Mentha piperita L. essential oil inactivates spoilage yeasts in fruit juices through the perturbation of different physiological functions in yeast cells.

This study evaluated the efficacy of the essential oil from Mentha piperita L. (MPEO) to inactivate cells of the potentially spoilage yeasts Candida albicans, Candida tropicalis, Pichia anomala and Saccharomyces cerevisiae in cashew, guava, mango and pineapple juices during 72 h of refrigerated storage. Damage in different physiological functions caused by MPEO in S. cerevisiae in cashew and guava juices were investigated using flow cytometry (FC). The effects of the incorporation of an effective anti-yeast MPEO dose on sensory characteristics of juices were also evaluated. MPEO displayed mini…

Phosphatidylserine is a component of the gradient-tracking machine in mating yeast.

Chemotropism, the ability to orient growth toward external chemical cues, is a fundamental process in diverse eukaryotic systems. During mating, budding yeast cells detect pheromone gradients from potential partners, locating them by assembling a gradient-tracking machine (GTM) at the plasma membrane that redistributes upgradient prior to polarized growth. Although membrane lipids are known to influence pheromone signaling and morphogenesis, their roles in pre-morphogenic gradient tracking have remained unclear. Here, we show that phosphatidylserine (PS), phosphatidylinositol-4,5-bisphosphate,

Enhancing myrcene biosynthesis in yeast through nuclear compartmentalization.

Myrcene is a high-value monoterpene extensively applied in the fragrance, flavor, and agricultural industries, yet its efficient microbial production remains challenging due to pathway competition and limited metabolic flux. Compartmentalization offers a unique strategy to spatially organize heterologous metabolic pathways in Saccharomyces cerevisiae, enabling improved pathway efficiency through physical separation from competing cytosolic metabolism. In this study, we engineered the S. cerevisiae nucleus as a synthetic metabolic compartment for myrcene biosynthesis. Screening of myrcene synth

Yeast biomimetic iron‑titanium oxides for electrochemical biosensing.

A yeast cell template technology is described to biomimetically synthesize FeTi oxides. The resulting samples, yeast Fe2O3 and yeast Fe2O3/TiO2, are then employed as active materials for the electrochemical detection of cancer-associated miRNAs. For the perfect matched cancer miR-141, electrochemical analysis shows a main oxidation peak at about +0.4 V, along with a reduction peak at about +0.30 V. For the detection of cancer miR-29c, the introduction of the Ti element is found to stabilize the structure of the resulting materials. These results suggest a potential of the biomimetic FeTi oxide

Yeast Membrane Hydration is Maintained Under Ethanol Exposure.

Yeasts are able to tolerate different environmental conditions, including stress situations. Given their broad applications in the food industry, their ability to adapt to stressful conditions is an active area of research. Lipid composition of the yeast membrane is affected by environmental stress, and thus, the regulation of the membrane biophysical properties under such conditions may be a key point for yeast adaptation. Although Saccharomyces cerevisiae is highly tolerant to ethanol, its growth is inhibited when this alcohol accumulates in the medium. Therefore, we studied the effect of et…

Advancing yeast metabolism for a sustainable single carbon bioeconomy.

Single carbon (C1) molecules are considered as valuable substrates for biotechnology, as they serve as intermediates of carbon dioxide recycling, and enable bio-based production of a plethora of substances of our daily use without relying on agricultural plant production. Yeasts are valuable chassis organisms for biotech production, and they are able to use C1 substrates either natively or as synthetic engineered strains. This minireview highlights native yeast pathways for methanol and formate assimilation, their engineering, and the realization of heterologous C1 pathways including CO2, in d…

Fanconi syndrome induced by red yeast rice supplement.

Fanconi syndrome is a disorder of the proximal renal tubules characterized by metabolic acidosis, renal glycosuria, and aminoaciduria. Recently, Fanconi syndrome caused by red yeast rice supplements has been reported. We encountered a 60-year-old woman with acute kidney injury and Fanconi syndrome caused by a red yeast rice supplement. The patient discontinued the supplements, leading to an improvement in her serum creatinine and urinary protein levels. Early detection and discontinuation of the suspected drugs are important for the treatment of drug-induced Fanconi syndrome.

Evidence for a hydrogen sulfide-sensing E3 ligase in yeast.

In yeast, control of sulfur amino acid metabolism relies upon Met4, a transcription factor that activates the expression of a network of enzymes responsible for the biosynthesis of cysteine and methionine. In times of sulfur abundance, the activity of Met4 is repressed via ubiquitination by the SCFMet30 E3 ubiquitin ligase, but the mechanism by which the F-box protein Met30 senses sulfur status to tune its E3 ligase activity remains unresolved. Herein, we show that Met30 responds to flux through the trans-sulfuration pathway to regulate the MET gene transcriptional program. In particular, Met3

Ergosterol promotes aggregation of natamycin in the yeast plasma membrane.

Polyene macrolides are antifungal substances, which interact with cells in a sterol-dependent manner. While being widely used, their mode of action is poorly understood. Here, we employ ultraviolet-sensitive (UV) microscopy to show that the antifungal polyene natamycin binds to the yeast plasma membrane (PM) and causes permeation of propidium iodide into cells. Right before membrane permeability became compromised, we observed clustering of natamycin in the PM that was independent of PM protein domains. Aggregation of natamycin was paralleled by cell deformation and membrane blebbing as reveal

Iron Deficiency and Recovery in Yeast: A Quantitative Proteomics Approach.

Iron is an essential element for life, as it is critical for oxygen transport, cellular respiration, DNA synthesis, and metabolism. Disruptions in iron metabolism have been associated with several complex diseases like diabetes, cancer, infection susceptibility, neurodegeneration, and others; however, the molecular mechanisms linking iron metabolism with these diseases are not fully understood. A commonly used model to study iron deficiency (ID) is yeast, Saccharomyces cerevisiae. Here, we used quantitative (phospho)proteomics to explore the early (4 and 6 h) and late (12 h) response to ID. We

The interplay between gut bacteria and the yeast Candida albicans.

The fungus Candida albicans is a ubiquitous member of the human gut microbiota. Hundreds or thousands of bacterial taxa reside together with this fungus in the intestine, creating a milieu with myriad opportunities for inter-kingdom interactions. Indeed, recent studies examining the broader composition - that is, monitoring not only bacteria but also the often neglected fungal component - of the gut microbiota hint that there are significant interdependencies between fungi and bacteria. Gut bacteria closely associate with C. albicans cells in the colon, break down and feed on complex sugars de

Impact of alanyl-tRNA synthetase editing deficiency in yeast.

Aminoacyl-tRNA synthetases (aaRSs) are essential enzymes that provide the ribosome with aminoacyl-tRNA substrates for protein synthesis. Mutations in aaRSs lead to various neurological disorders in humans. Many aaRSs utilize editing to prevent error propagation during translation. Editing defects in alanyl-tRNA synthetase (AlaRS) cause neurodegeneration and cardioproteinopathy in mice and are associated with microcephaly in human patients. The cellular impact of AlaRS editing deficiency in eukaryotes remains unclear. Here we use yeast as a model organism to systematically investigate the physi

[Red yeast rice, monacolin K, and pleiotropic effects.].

The extracts of red yeast rice represent a nutraceutical with proven cholesterol lowering effect. Its efficacy is proportional to the concentration on monacolin K in the extract that could reach the amount of 10 mg per daily dose. The daily assumption of monacolin K could then reduce LDL-cholesterol plasma levels by 15-25% in 6-8 weeks. The LDL-cholesterol reduction is associated with a proportional reduction in total cholesterolemia, non-HDL cholesterolemia, plasma apolipoprotein B, high-sensitivity C-reactive protein, and matrix metalloproteinases 2 and 9. Then, the red yeast rice lipid-lowe

Red Yeast Rice Preparations: Are They Suitable Substitutions for Statins?

Red yeast rice, a commercially available food supplement known to reduce serum cholesterol, has been repeatedly advocated as alternative therapy for hypercholesterolemic patients that refuse statins, cannot tolerate statin therapy's side effects, or request a "naturopathic" medicine. Red yeast rice contains a fungus (Monascus purpureus), which was utilized in the original production of lovastatin (MEVACOR, Merck & Co, Whitehouse Station, NJ), the first marketed pharmaceutical statin, and is chemically identical to such product. Their identical properties account for the similarity in therapeut

A genetic method to enhance the accumulation of S-adenosylmethionine in yeast.

S-Adenosylmethionine (SAM) is a key component of sulphur amino acid metabolism in living organisms and is synthesised from methionine and adenosine triphosphate by methionine adenosyltransferase. This molecule serves as the main biological methyl donor due to its active methylthio ether group. Notably, SAM has shown beneficial effects in clinical trials for the treatment of alcoholic liver disease, depression and joint pain. Due to the high potential value of SAM, current research efforts are attempting to develop a more rapid, cost-effective and higher yielding SAM production method than the
