Species Identification of the Major Japanese Encephalitis Vectors within the Culex vishnui Subgroup (Diptera: Culicidae) in Thailand Using Geometric Morphometrics and DNA Barcoding.

Japanese encephalitis (JE) is a viral infection of the brain caused by the Japanese encephalitis virus, which spreads globally, particularly in 24 countries of Southeast Asia and the Western Pacific region. In Thailand, the primary vectors of JE are Cx. pseudovishnui, Cx. tritaeniorhynchus, and Cx. vishnui of the Cx. vishnui subgroup. The morphologies of three mosquito species are extremely similar, making identification challenging. Thus, geometric morphometrics (GM) and DNA barcoding were applied for species identification. The results of cross-validation reclassification revealed that the G

Subconjunctival human dirofilariasis by Dirofilaria repens in the Mediterranean Basin.

PURPOSE: Human dirofilariasis cases have increased in Europe for the last few decades. We document an autochthonous case of dirofilariasis infection in the subconjunctival space of a 60-year-old Spanish man. OBSERVATIONS: The patient presented with pain in his right eye, which was diagnosed as epi-scleritis. In spite of the treatment administered, no improvement was reported. An external exam showed conjunctival congestion on the temporal part of the right eye bulbar conjunctiva. The anterior chamber showed no flare or cells in both eyes and dilated fundus exam was normal. Nevertheless, a lon

Synthesis of new series of quinoline derivatives with insecticidal effects on larval vectors of malaria and dengue diseases.

Mosquito borne diseases are on the rise because of their fast spread worldwide and the lack of effective treatments. Here we are focusing on the development of a novel anti-malarial and virucidal agent with biocidal effects also on its vectors. We have synthesized a new quinoline (4,7-dichloroquinoline) derivative which showed significant larvicidal and pupicidal properties against a malarial and a dengue vector and a lethal toxicity ranging from 4.408 µM/mL (first instar larvae) to 7.958 µM/mL (pupal populations) for Anopheles stephensi and 5.016 µM/mL (larva 1) to 10.669 µM/mL (pupae) for Ae

A retrospective study of environmental predictors of dengue in Delhi from 2015 to 2018 using the generalized linear model.

Dengue fever is a mosquito-borne infection with a rising trend, expected to increase further with the rise in global temperature. The study aimed to use the environmental and dengue data 2015-2018 to examine the seasonal variation and establish a probabilistic model of environmental predictors of dengue using the generalized linear model (GLM). In Delhi, dengue cases started emerging in the monsoon season, peaked in the post-monsoon, and thereafter, declined in early winter. The annual trend of dengue cases declined, but the seasonal pattern remained alike (2015-18). The Spearman correlation c

The Burden of Bancroftian Filariasis in Nigeria: A Review.

BACKGROUND: Lymphatic filariasis (LF) is a neglected tropical disease (NTD) vectored by mosquito; and people in rural areas are mostly at risk of infection. Pooling prevalence data across the six geo-political zones of Nigeria is expected to provide a clearer insight into the burden of the disease as this information could guide towards planning eradication programmes. METHODS: Search for pertinent literature was done on Google Scholar, African Journal Online (AJOL) and PubMed databases using relevant keywords. Studies on the prevalence of LF due to Wuchereria bancrofti in Nigeria were select

Transmission dynamics of dengue and chikungunya in a changing climate: do we understand the eco-evolutionary response?

INTRODUCTION: We are witnessing an alarming increase in the burden and range of mosquito-borne arboviral diseases. The transmission dynamics of arboviral diseases is highly sensitive to climate and weather and is further affected by non-climatic factors such as human mobility, urbanization, and disease control. As evidence also suggests, climate-driven changes in species interactions may trigger evolutionary responses in both vectors and pathogens with important consequences for disease transmission patterns. AREAS COVERED: Focusing on dengue and chikungunya, we review the current knowledge a

Host Associations Of Biting Midges (Diptera: Ceratopogonidae: Culicoides) Near Sentinel Chicken Surveillance Locations In Florida, USA.

Quantifying host use is important for understanding transmission of vector-borne pathogens. Despite the importance of biting midges (Diptera: Ceratopogonidae) in pathogen transmission, the vector-host relationships of most Culicoides species are poorly documented, even in locations where active arbovirus surveillance is conducted. Polymerase chain reaction-based blood-meal analysis was performed on 663 blood-engorged Culicoides females collected by 7 Florida mosquito control districts at 24 sentinel chicken arbovirus surveillance sites in 2017. A total of 638 blood meals were successfully anal…

[Association between malaria and low birth weight in Yaounde, Cameroon].

INTRODUCTION: Low birth weight (LBW) is an important predictor of newborn survival and development. Given its pathophysiology, malaria is presumed to be one of the risk factors for low birth weight. This study aims to determine the association between malaria in pregnant women (PW) and LBW (weight < 2500 g). METHODS: We conducted a case-control analytical study based on the administration of a questionnaire and an observation chart. We calculated the crude odds ratio (OR) and the adjusted odds ratio to determine this association. Logistic regression was applied to recognize the variables whic

Climate change may enable Aedes aegypti infestation in major European cities by 2100.

BACKGROUND: Climate change allows Aedes aegypti to infest new areas. Consequently, it enables the arboviruses the mosquito transmits -- e.g., dengue, chikungunya, Zika and yellow fever - to emerge in previously uninfected areas. An example is the Portuguese island of Madeira during 2012-13. OBJECTIVE: We aim to understand how climate change will affect the future spread of this potent vector, as an aid in assessing the risk of disease outbreaks and effectively allocating resources for vector control. METHODS: We used an empirically-informed, process-based mathematical model to study the feas

Discovery of an Aedes (Stegomyia) albopictus population and first records of Aedes (Stegomyia) aegypti in Canada.

A population of Aedes (Stegomyia) albopictus (Skuse) (Diptera: Culicidae), a vector of chikungunya, dengue, yellow fever, and Zika and West Nile viruses, has been detected in Windsor, Ontario, Canada from 2016 onwards. Here, we describe its seasonal distribution, as well as the various aquatic habitats from which this species was collected and its larval co-habitation. We collected immatures from tires, treeholes, extruded polystyrene foam containers, discarded plastic cups, old recycling bins and oviposition traps. Aedes albopictus larvae were collected with Aedes japonicus (Theobald), Anophe

Deciphering the dark proteome of Chikungunya virus.

Chikungunya virus (CHIKV) is a mosquito-borne alphavirus. The outbreak of CHIKV infection has been seen in many tropical and subtropical regions of the biosphere. Current reports evidenced that after outbreaks in 2005-06, the fitness of this virus propagating in Aedes albopictus enhanced due to the epistatic mutational changes in its envelope protein. In our study, we evaluated the prevalence of intrinsically disordered proteins (IDPs) and IDP regions (IDPRs) in CHIKV proteome. IDPs/IDPRs are known as members of a 'Dark Proteome' that defined as a set of polypeptide segments or whole protein w

Dengue outbreaks and the geographic distribution of dengue vectors in Taiwan: A 20-year epidemiological analysis.

Dengue fever is an important mosquito-borne viral infectious disease that mostly occurs in tropical and subtropical areas of the world. According to epidemiological data from the Center for Disease Control of Taiwan, more than 98.62% of outbreaks of indigenous total dengue cases were reported in the southern part of Taiwan. Southern Taiwan is an aggregate area encompassing Tainan, Kaohsiung, and Pingtung, all of which are located below the Tropic of Cancer (23º35'N). With a few exceptions, dengue outbreaks mainly occur in southern Taiwan which is highly associated or overlaps with the prevalen

What Does the Future Hold for Yellow Fever Virus? (I).

The recent resurgence of yellow fever virus (YFV) activity in the tropical regions of Africa and South America has sparked renewed interest in this infamous arboviral disease. Yellow fever virus had been a human plague for centuries prior to the identification of its urban transmission vector, the Aedes (Stegomyia) aegypti (Linnaeus) mosquito species, and the development of an efficient live-attenuated vaccine, the YF-17D strain. The combination of vector-control measures and vaccination campaigns drastically reduced YFV incidence in humans on many occasions, but the virus never ceased to circ

Small Molecules and Antibodies for Zika Therapy.

Zika virus (ZIKV) infection during pregnancy can cause devastating congenital abnormities or fetal demise. Zika virus infection could also cause Guillain-Barré syndrome in adults. Mosquito control, vaccine, and therapeutics are 3 potential, effective means to prevent ZIKV infection. Here we review the current status of ZIKV drug discovery. Both small molecule inhibitors and therapeutic antibodies have been identified, some of which have shown promising efficacy in mouse models. Most inhibitors were identified through screening US Food and Drug Administration-approved drugs and clinical trial c…

Effective population sizes of a major vector of human diseases, Aedes aegypti.

The effective population size (Ne ) is a fundamental parameter in population genetics that determines the relative strength of selection and random genetic drift, the effect of migration, levels of inbreeding, and linkage disequilibrium. In many cases where it has been estimated in animals, Ne is on the order of 10%-20% of the census size. In this study, we use 12 microsatellite markers and 14,888 single nucleotide polymorphisms (SNPs) to empirically estimate Ne in Aedes aegypti, the major vector of yellow fever, dengue, chikungunya, and Zika viruses. We used the method of temporal sampling to

Prospects and challenges of CRISPR/Cas genome editing for the study and control of neglected vector-borne nematode diseases.

Neglected tropical diseases caused by parasitic nematodes inflict an immense health and socioeconomic burden throughout much of the developing world. Current estimates indicate that more than two billion people are infected with nematodes, resulting in the loss of 14 million disability-adjusted life years per annum. Although these parasites cause significant mortality, they primarily cause chronic morbidity through a wide range of severe clinical ailments. Treatment options for nematode infections are restricted to a small number of anthelmintic drugs, and the rapid expansion of anthelmintic m

Antibody Profiling in Naïve and Semi-immune Individuals Experimentally Challenged with Plasmodium vivax Sporozoites.

BACKGROUND: Acquisition of malaria immunity in low transmission areas usually occurs after relatively few exposures to the parasite. A recent Plasmodium vivax experimental challenge trial in malaria naïve and semi-immune volunteers from Colombia showed that all naïve individuals developed malaria symptoms, whereas semi-immune subjects were asymptomatic or displayed attenuated symptoms. Sera from these individuals were analyzed by protein microarray to identify antibodies associated with clinical protection. METHODOLOGY/PRINCIPAL FINDINGS: Serum samples from naïve (n = 7) and semi-immune (n =

Regulators of endothelial integrity as severity predictors in dengue.

INTRODUCTION: Dengue is currently among the mosquito-borne diseases of greatest global impact. The clinical course of the disease can be unpredictable, so proper handling in its early stages is critical to ensure optimal outcomes. OBJECTIVE: To evaluate serum regulators of endothelial integrity (VEGF, sICAM-1, sEndoglina, Ang-1, and Ang-2) as predictive markers of dengue severity. MATERIALS AND METHODS: We conducted a case-control study nested in an appropriate cohort. Endothelial regulator levels were first measured by ELISA, after which analysis was performed using logistic regression of c

Chimeric Epitope Vaccine from Multistage Antigens for Lymphatic Filariasis.

Lymphatic filariasis, a mosquito-borne parasitic disease, affects more than 120 million people worldwide. Vaccination for filariasis by targeting different stages of the parasite will be a boon to the existing MDA efforts of WHO which required repeated administration of the drug to reduce the infection level and sustained transmission. Onset of a filaria-specific immune response achieved through antigen vaccines can act synergistically with these drugs to enhance the parasite killing. Multi-epitope vaccine approach has been proved to be successful against several parasitic diseases as it overc
