Morphological and molecular identification of Dirofilaria immitis from Jackal (Canis aureus) in North Khorasan, northeast Iran.

BACKGROUND & OBJECTIVES: The heartworm Dirofilaria immitis is an important mosquito-borne zoonotic nematode of domestic and wild mammals throughout the world, causing cardiopulmonary dirofilariasis. This parasite has been reported from carnivores in some provinces of Iran. However, in the present study, the occurrence of this filarial nematode is reported for the first time in wild canids of the North Khorasan Province, located in northeast Iran, based on morphological and molecular characteristics. METHODS: The carcasses of 45 golden jackals (Canis aureus), 16 foxes (Vulpes vulpes), 15 dogs

A Trypsin Inhibitor from Clitoria fairchildiana Cotyledons is Active Against Digestive Enzymes of Aedes aegypti Larvae.

Aedes aegypti, the principal mosquito vector of yellow fever, dengue fever and chikungunya fever virus-transmitted diseases, is an insect closely associated with humans and their housing habitats. As there is no commercially available vaccine, prevention is the most suggested form of avoiding disease spreading and a number of studies are being developed in order to give support to vector control operations. The present study reports on the identification of a trypsin inhibitor isolated from cotyledons of the Clitoria fairchildiana amazonic tree seeds, which was able to reduce by 87.93 % the ac

Nonstructural protein 2 (nsP2) of Chikungunya virus (CHIKV) enhances protective immunity mediated by a CHIKV envelope protein expressing DNA Vaccine.

Chikungunya virus (CHIKV) is an important emerging mosquito-borne alphavirus, indigenous to tropical Africa and Asia. It can cause epidemic fever and acute illness characterized by fever and arthralgias. The epidemic cycle of this infection is similar to dengue and urban yellow fever viral infections. The generation of an efficient vaccine against CHIKV is necessary to prevent and/or control the disease manifestations of the infection. In this report, we studied immune response against a CHIKV-envelope DNA vaccine (pEnv) and the role of the CHIKV nonstructural gene 2 (nsP2) as an adjuvant for

The NS3 protease and helicase domains of Japanese encephalitis virus trigger cell death via caspase‑dependent and ‑independent pathways.

Japanese encephalitis virus (JEV), a mosquito‑borne flavivirus, causes acute encephalitis and nervous damage. Previous studies have demonstrated that JEV induces apoptosis in infected cells. However, to date the mechanisms of JEV‑induced apoptosis are unclear. In order to identify the viral proteins associated with JEV‑induced apoptosis, pEGFP‑non‑structural protein 3 (NS3) 1‑619 (expressing the JEV NS3 intact protein, including the protease and helicase domains), pEGFP‑NS3 1‑180 (expressing the protease domain) and pEGFP‑NS3 163‑619 (expressing the helicase domain) were transfected into targe

Insect repellents and associated personal protection for a reduction in human disease.

Personal protection measures against biting arthropods include topical insect repellents, area repellents, insecticide-treated bednets and treated clothing. The literature on the effectiveness of personal protection products against arthropods is mainly limited to studies of prevention of bites, rather than prevention of disease. Tungiasis was successfully controlled by application of topical repellents and scrub typhus was reduced through the use of treated clothing. Successful reduction of leishmaniasis was achieved through the use of topical repellents, treated bednets and treated clothing

A detrimental role for invariant natural killer T cells in the pathogenesis of experimental dengue virus infection.

Dengue virus (DENV), a member of the mosquito-borne flaviviruses, is a serious public health problem in many tropical countries. We assessed the in vivo physiologic contribution of invariant natural killer T (iNKT) cells, a population of nonconventional lipid-reactive αβ T lymphocytes, to the host response during experimental DENV infection. We used a mouse-adapted DENV serotype 2 strain that causes a disease that resembles severe dengue in humans. On DENV challenge, splenic and hepatic iNKT cells became activated insofar as CD69 and Fas ligand up-regulation and interferon-γ production. C57BL/

Air conditioning and television as protective factors in arboviral encephalitis risk.

In California, the advent of television and air conditioned housing has coincided with a general decrease in mosquito-borne viral encephalitis cases in humans in the past 25 years. During this same period, levels of vector populations, virus activity in vectors, birds, and horses has been high at times. Air conditioning and television encourage persons to remain indoors during summer evenings, the primary time when infected Culex tarsalis transmit western equine encephalomyelitis and St. Louis encephalitis viruses. The attack rates for these 2 diseases in 33 counties in the central valley of C

Caregivers' perception of risk for malaria, helminth infection and malaria-helminth co-infection among children living in urban and rural settings of Senegal: A qualitative study.

The parasites causing malaria, soil-transmitted helminthiasis and schistosomiasis frequently co-exist in children living in low-and middle-income countries, where existing vertical control programmes for the control of these diseases are not operating at optimal levels. This gap necessitates the development and implementation of strategic interventions to achieve effective control and eventual elimination of these co-infections. Central to the successful implementation of any intervention is its acceptance and uptake by caregivers whose perception about the risk for malaria-helminth co-infecti

Overlaying human and mosquito behavioral data to estimate residual exposure to host-seeking mosquitoes and the protection of bednets in a malaria elimination setting where indoor residual spraying and nets were deployed together.

Characterizing persistent malaria transmission that occurs after the combined deployment of indoor residual spraying (IRS) and long-lasting insecticidal nets (LLINs) is critical to guide malaria control and elimination efforts. This requires a detailed understanding of both human and vector behaviors at the same temporal and spatial scale. Cross-sectional human behavior evaluations and mosquito collections were performed in parallel in Magude district, Mozambique. Net use and the exact time when participant moved into each of five environments (outdoor, indoor before bed, indoor in bed, indoor

The impact of temperature on the bionomics of Aedes (Stegomyia) aegypti, with special reference to the cool geographic range margins.

The mosquito Aedes (Stegomyia) aegypti (L.), which occurs widely in the subtropics and tropics, is the primary urban vector of dengue and yellow fever viruses, and an important vector of chikungunya virus. There is substantial interest in how climate change may impact the bionomics and pathogen transmission potential of this mosquito. This Forum article focuses specifically on the effects of temperature on the bionomics of Ae. aegypti, with special emphasis on the cool geographic range margins where future rising temperatures could facilitate population growth. Key aims are to: 1) broadly defi

From gibson assembly to cell-free expression: A guide to recombinant plasmid and toscana virus N protein production.

Toscana virus (TOSV) is a mosquito-borne phlebovirus endemic to the Mediterranean region and an important cause of arboviral infections in humans, particularly during the summer months. Although TOSV infections are often asymptomatic or mild, the virus can cause neuroinvasive disease, including meningitis and encephalitis, highlighting the need for rapid and reliable tools for its detection and characterization. N protein was identified as a promising diagnostic target for TOSV due to its accumulation in infected cells and its role as primary immune target. The aim of this study was to establi

Bacterial Extracellular Vesicles (BEVs) Derived from Chryseobacterium Inhibit Dengue Virus Infection by Disrupting Its Structural Integrity.

Dengue virus (DENV) infection poses a significant global health threat, and current prevention and treatment strategies are limited by challenges of lacking effective mosquito control measures and antibody-dependent enhancement. This study reports that bacterial extracellular vesicles (BEVs) secreted by a soil bacterium Chryseobacterium aquifrigidense M24 exhibit potent anti-DENV activity by triggering the structural disintegration of DENV particles prior to cellular entry in a dose-dependent manner. Mechanistic investigations revealed that BEVs interact with the viral envelope, inducing prema…

Antiphasic Arctangent-Arccotangent (AAA) probes: A kinetically orthogonal design for single-tube, real-time, and low-crosstalk multiplex LAMP.

The rapid and discriminative detection of multiple pathogens is crucial for effective disease control, yet remains a significant challenge for point-of-care testing (POCT). Herein, we report a novel probe design strategy-Antiphasic Arctangent-Arccotangent (AAA) probes-for kinetically orthogonal, low-crosstalk multiplex detection in a single closed tube via loop-mediated isothermal amplification (LAMP). The AAA probe system comprises two kinetically complementary probes: the Arctangent Probe (ATP), a stem-loop probe that yields a rising fluorescence signal, and the Arccotangent Probe (ACP), a g

Decoding and engineering temperature-sensitive lethality in Ceratitis capitata for pest control.

The Sterile Insect Technique (SIT) is a species-specific and environmentally friendly method for effectively controlling pest insect populations based on releasing reared, sterile insects into infested areas. Sex sorting in rearing facilities, enabling male-only releases, is necessary to ensure SIT programs are efficient, cost-effective and, in case of mosquito control, also safe. This can be greatly facilitated by genetic sexing strains (GSS), exhibiting sex-specific phenotypic markers. However, the development of GSS remains challenging. The construction of a temperature-sensitive lethal (ts…

The hygienic insecticide dimefluthrin induced neurodevelopmental deficits and behavioral disorders in zebrafish (Danio rerio) larvae.

Hygienic insecticides are applied directly to the living environment and are closely related to human life. Dimefluthrin (DIM) is one of the most widely used hygienic insecticides globally. However, with increasing mosquito resistance, both the concentration and duration of DIM usage have risen, prompting public concerns regarding its neurotoxic risks, especially for immunocompromised children. Therefore, this study evaluated DIM's neurotoxic risk from multiple perspectives using zebrafish larvae as a model, which was valuable for the future risk control of sanitary insecticides. The findings
