Helicobacter pylori requires genetic agility to infect new hosts and establish long-term colonization of changing gastric environments. In this study, we analyzed H. pylori genetic adaptation in the Mongolian gerbil model. This model is of particular interest because H. pylori-infected gerbils develop a high level of gastric inflammation and often develop gastric adenocarcinoma or gastric ulceration. We analyzed the whole genome sequences of H. pylori strains cultured from experimentally infected gerbils, in comparison to the genome sequence of the input strain. The mean annualized single nucl

The flaviviruses comprise a number of arthropod-transmitted human disease agents that cause significant and increasing health threats in major parts of the world. The development of new vaccines is of vital importance, but the stringent need for safety, efficacy and cost-effectiveness together with the problems associated with the specific immune pathogenesis of some flavivirus infections impose significant challenges to innovative vaccine research. Using tick-borne encephalitis virus (TBEV) as a model, the viral capsid protein gene was recently identified as a novel target for generating flav

In this study, we examined the neuroanatomy of dyslexic (14 males, four females) and control (19 males, 13 females) children in grades 4-6 from a family genetics study. The dyslexics had specific deficits in word reading relative to the population mean and verbal IQ, but did not have primary language or motor deficits. Measurements of the posterior temporal lobe, inferior frontal gyrus, cerebellum and whole brain were collected from MRI scans. The dyslexics exhibited significantly smaller right anterior lobes of the cerebellum, pars triangularis bilaterally, and brain volume. Measures of the r

Mendelian forms of hypertension have delivered a treasure trove of novel genes. To date, the molecular mechanisms of five such syndromes have been largely clarified, including glucocorticoid-remediable aldosteronism, Liddle's syndrome, apparent mineralocorticoid excess, an activating mutation of the mineralocorticoid receptor, and pseudohypoaldosteronism type 2. Each of these conditions features salt sensitivity with increased sodium and volume reabsorption by the kidney and low plasma renin activity. None of the gene loci for these syndromes has been convincingly linked to hypertension in the

Primary open-angle glaucoma (POAG) is a complex disorder characterized by a progressive and treatable degeneration of the optic nerve. TIGR/myocilin (MYOC) gene mutations are found in approximately 4% of all POAG patients. Populations with frequent founder effects, such as the French-Canadians, offer unique advantages to implement genetic testing for the disorder. To assess molecular diagnosis for POAG in this population, we determined the prevalence of TIGR/MYOC mutations in 384 unrelated glaucoma patients, 38 ocular hypertensive subjects and 18 affected families (180 patients). We further an

Leucocyte adhesion deficiency (LAD) is an autosomal-recessive genetic disease that is characterized clinically by severe bacterial infections and caused by mutations in the CD18 gene that codes for the beta2 integrin subunit. A patient with a severe LAD phenotype was studied and the molecular basis of the disease was identified as a single homozygous defect in a Herpes virus saimiri (HVS)-transformed T-cell line. The defect identified involves a deletion of 171 bp in the cDNA that encodes part of the proteic extracellular domain. This genetic abnormality was further studied at the genomic DNA

Sacral agenesis is a heterogeneous group of congenital anomalies in which most cases are sporadic but rare familial forms also occur. Although one gene has been mapped to chromosome 7q36 in families with hemisacrum, associated with anorectal atresia and presacral mass, it is clear that the genetic aetiology of these disorders is complex and other genes remain to be discovered. Some years ago, the idea of T (Brachyury) as a candidate gene for sacral agenesis was raised, because tail abnormalities associated with T and the t complex, on mouse chromosome 17, resemble spinal defects seen in man. T

Abnormal beta-hexosaminidase beta chain cDNA clones were isolated from a library constructed from cultured fibroblasts of a patient with a juvenile form of Sandhoff disease (genetic beta-hexosaminidase A and B deficiency). Sequence analysis of a cDNA clone isolated from these fibroblasts contained an extra 24-base segment between exons 12 and 13. This segment was identified as the 3' terminus of intron 12. The remainder of the coding sequence was completely normal. The same 24-base insertion was found in four additional clones by sequencing. Restriction mapping analysis of seven other clones w

Phytoremediation of cadmium (Cd)-contaminated agricultural soils requires suitable germplasm and an improved understanding of the genetic factors underlying Cd accumulation. Here, we evaluated a genetically diverse panel of 165 Brassica juncea accessions and identified BJ20 as a high-Cd-accumulating genotype. Across two years, two sites, and two developmental stages, BJ20 consistently accumulated more Cd than the low-accumulating genotype BJ25. Mean shoot Cd accumulation reached 111.21 μg plant⁻¹ in BJ20, compared with 55.07 μg plant⁻¹ in BJ25. However, BJ20 had a lower root-to-shoot transloca

BACKGROUND: The monocarboxylate transporter 1 (MCT1) is a key mediator of lactate transport, and genetic polymorphisms may influence perioperative lactate dynamics. We investigated the association between the MCT1 rs1049434 polymorphism and postoperative lactate levels and outcomes following major abdominal surgery.
METHODS: This prospective observational study included patients undergoing major abdominal surgery. Postoperative lactate trajectories were analysed using linear mixed-effects models with a random intercept for patients and fixed effects for time, genotype, and their interaction.

Mastophorus muris is a prevalent nematode parasite of rodents that can cause pathogenic manifestations or mortality. As the only 1 species in the genus Mastophorus and the sole member of the family Spirocercidae, its biology, molecular identification, and population genetic structure remain poorly studied. In this study, the complete mitochondrial genome of M. muris was fully assembled and annotated through high-fidelity next-generation sequencing for the first time, to resolve its molecular architecture with nucleotide-level precision. The 13,668 bp mitogenome encodes 36 genes, including 12 p

Insertion of fluorescent reporter genes into viral genomes is a powerful tool for monitoring infection. In coronaviruses, this is commonly achieved by replacing accessory ORFs, thereby deleting endogenous gene functions. An alternative strategy is to manipulate viral RNA synthesis by inserting copies of the viral transcription regulatory sequence (TRS), which drives the transcription of viral subgenomic RNAs. However, coronavirus transcription is tightly regulated, and these modifications frequently disrupt native subgenomic RNA synthesis and attenuate viral growth. Here, we describe a reporte

X-linked Duchenne muscular dystrophy (DMD), caused by mutations in a gene coding several dystrophin isoforms from independent promoters, is associated with a range of brain-related comorbidities. Intellectual disability is variably expressed depending on the position of the genetic variants, intelligence quotients being negatively correlated with the number of deficient brain dystrophins. However, the genetic basis of other phenotypes underlying cognitive deficits and neuropsychiatric disorders, such as executive dysfunctions and attention deficit/hyperactivity disorder (ADHD), is still unclea

INTRODUCTION: Genetic disorders are major causes of morbidity and mortality in neonatal intensive care units (NICUs), yet traditional tests like karyotyping and chromosomal microarray (CMA) often fail to provide a diagnosis, resulting in a "diagnostic odyssey". Trio whole-genome sequencing (WGS) has emerged as a comprehensive tool capable of detecting a wide range of variants beyond protein-coding regions, potentially offering higher diagnostic yields and rapid, actionable results for critical care management.
METHODS: This prospective study evaluated 16 neonates with multiple congenital anom

Alzheimer's disease (AD) is a complex neurodegenerative disorder influenced by various factors, including genetic and exposure-related. Certain combinations of these factors may promote AD more substantially than others. APOE4 is the strongest genetic risk factor for AD. Traffic-related air pollution (TRAP) and infections are important exposure-related AD risk factors. Here we investigated how the interplay between a history of infections and chronically high exposure to TRAP (highTRAP) impacts the subsequent risk of AD and other dementias (AD+) in carriers and non-carriers of APOE4 in UK Biob

Background/Objectives: Atrial fibrillation (AF), the most prevalent form of clinical cardiac arrhythmia globally, is associated with markedly increased morbidity, mortality, and socio-economic expenditure. Accumulating strong evidence highlights genetic abnormalities underpinning its etiopathogenesis. A recent investigation has demonstrated that mutations in the TBX20 gene, which codes for a T-box transcription factor essential for proper cardiovascular development and structural remodeling, contribute to familial AF. Nevertheless, the mutational prevalence and spectrum of this gene in patient

BACKGROUND/OBJECTIVES: Opioid use disorder (OUD) is caused by a complex interplay between genetic and non-genetic factors. DNA methylation is an epigenetic mechanism that modulates gene expression. Data on DNA methylation and opioid addiction and treatment are limited. This association study was designed to assess the difference in genome-wide methylation patterns between individuals with OUD in methadone maintenance treatment (MMT) (n = 114) and those with OUD who achieved long-term abstinence (>10 years) without mu opioid receptor agonist treatment (n = 136).
METHODS: Differential DNA methy

Thrombotic thrombocytopenic purpura (TTP) is a thrombotic microangiopathy caused by severe deficiency of the metalloprotease ADAMTS13. TTP has 2 subtypes: immune (iTTP) and congenital (cTTP). Autoimmune mechanisms underlie iTTP; however, the genetic factors influencing disease risk, relapse risk, and response to treatment are incompletely understood. In cTTP, although variants are described throughout the ADAMTS13 gene, the effect of many of these variants is uncertain. We conducted a comprehensive literature review of both iTTP and cTTP, incorporating findings from genome-wide association stu
