Outcome-Based Decision-Making Algorithm for Treating Patients with Primary Aldosteronism.

BACKGROUND: Optimal management of primary aldosteronism (PA) is crucial due to the increased risk of cardiovascular and cerebrovascular diseases. Adrenal venous sampling (AVS) is the gold standard method for determining subtype but is technically challenging and invasive. Some PA patients do not benefit clinically from surgery. We sought to develop an algorithm to improve decision- making before engaging in AVS and surgery in clinical practice. METHODS: We conducted the ongoing Korean Primary Aldosteronism Study at two tertiary centers. Study A involved PA patients with successful catheteriza

AIBx, Artificial Intelligence Model to Risk Stratify Thyroid Nodules.

Background: Current classification systems for thyroid nodules are very subjective. Artificial intelligence (AI) algorithms have been used to decrease subjectivity in medical image interpretation. One out of 2 women over the age of 50 years may have a thyroid nodule and at present the only way to exclude malignancy is through invasive procedures for those that are suspicious on ultrasonography. Hence, there exists a need for noninvasive objective classification of thyroid nodules. Some cancers have benign appearance on ultrasonogram. Hence, we decided to create an image similarity algorithm ra

Machine learning based screening of biomarkers associated with cell death and immunosuppression of multiple life stages sepsis populations.

Sepsis is a condition resulting from the uncontrolled immune response to infection, leading to widespread inflammatory damage and potentially fatal organ dysfunction. Currently, there is a lack of specific prevention and treatment strategies for sepsis across different age groups. Programmed Cell Death (PCD) can regulate the enrichment of effector immune cells or regulatory immune cells, providing a new perspective for immunotherapy. Within the framework of computational biology and machine learning strategies, and against the backdrop of global multicenter sepsis cohort data, this study aims

A single-cell and machine learning framework identifies CAFs-associated signatures linking stromal heterogeneity to immune regulation in pancreatic cancer.

BACKGROUND: Cancer-associated fibroblasts (CAFs) are key components of the tumor microenvironment (TME) in pancreatic ductal adenocarcinoma (PDAC), contributing to tumor progression, metabolic reprogramming, and immune suppression. However, the functional heterogeneity of CAFs and their prognostic and immunological significance remain incompletely understood. This study aimed to characterize CAFs heterogeneity in PDAC and develop a robust CAFs-associated signature (CAFAS) for prognostic prediction and immune stratification. METHODS: Single-cell RNA sequencing (scRNA-seq) datasets from PDAC we
