Quantitative structure-binding relationships (QSBR) and artificial neural networks: improved predictions in drug:cyclodextrin inclusion complexes.

The application of the principal neural network architecture, namely the multilayer perceptron (MLP), have been developed for obtaining sufficient quantitative structure-binding relationships (QSBR) with high accuracy. To this end a dataset of 17 barbiturates as guests complexing to alpha- and beta-cyclodextrins was examined and the results compared to that of Lopata et al (J. Pharm. Sci., 74, (1995)) who studied the same problem using multiple regression analysis. A series of new and improved algorithms other than the "old fashion" and problematic steepest descent were examined for training t

Neurochemically informed network control theory reveals energetic dysregulation of altered brain state dynamics in obsessive-compulsive disorder.

AIM: Previous studies assessed dynamic functional connectivity in obsessive-compulsive disorder (OCD) by the sliding-window approach, limiting sensitivity to rapid neural fluctuations. Moreover, the mechanisms underlying dynamic transitions between brain states also remain unknown. Therefore, the study aimed to explore the dynamic neural mechanisms of OCD by characterizing state dynamic patterns and the underlying energy basis. METHODS: The study recruited 198 OCD patients and 109 healthy controls, characterizing altered state dynamic patterns and underlying control energy in OCD by integrati

Emotion Detection from EEG Signals Using Machine Deep Learning Models.

Detecting emotions is a growing field aiming to comprehend and interpret human emotions from various data sources, including text, voice, and physiological signals. Electroencephalogram (EEG) is a unique and promising approach among these sources. EEG is a non-invasive monitoring technique that records the brain's electrical activity through electrodes placed on the scalp's surface. It is used in clinical and research contexts to explore how the human brain responds to emotions and cognitive stimuli. Recently, its use has gained interest in real-time emotion detection, offering a direct approa

Toward personalized circuit-based closed-loop brain-interventions in psychiatry: using symptom provocation to extract EEG-markers of brain circuit activity.

Symptom provocation is a well-established component of psychiatric research and therapy. It is hypothesized that specific activation of those brain circuits involved in the symptomatic expression of a brain pathology makes the relevant neural substrate accessible as a target for therapeutic interventions. For example, in the treatment of obsessive-compulsive disorder (OCD), symptom provocation is an important part of psychotherapy and is also performed prior to therapeutic brain stimulation with transcranial magnetic stimulation (TMS). Here, we discuss the potential of symptom provocation to i

Quantifying the calcification of abdominal aorta and major side branches with deep learning.

AIM: To explore the possibility of a neural network-based method for quantifying calcifications of the abdominal aorta and its branches. MATERIALS AND METHODS: In total, 58 computed tomography (CT) angiography volumes were selected from a dataset of 609 to represent different stages of sclerosis. The ground truth segmentations of the abdominal aorta, coeliac trunk, superior mesenteric artery, renal arteries, common iliac arteries, and their calcifications were delineated manually. Two V-Net ensemble models were trained, one for segmenting arteries of interest and another for calcifications. T

Intrinsic neural timescales mediate the cognitive bias of self - temporal integration as key mechanism.

Our perceptions and decisions are not always objectively correct as they are featured by a bias related to our self. What are the behavioral, neural, and computational mechanisms of such cognitive bias? Addressing this yet unresolved question, we here investigate whether the cognitive bias is related to temporal integration and segregation as mediated by the brain's Intrinsic neural timescales (INT). Using Signal Detection Theory (SDT), we operationalize the cognitive bias by the Criterion C as distinguished from the sensitivity index d'. This was probed in a self-task based on morphed self- a

On construction of transfer learning for facial symmetry assessment before and after orthognathic surgery.

Orthognathic surgery (OGS) is frequently used to correct facial deformities associated with skeletal malocclusion and facial asymmetry. An accurate evaluation of facial symmetry is a critical for precise surgical planning and the execution of OGS. However, no facial symmetry scoring standard is available. Typically, orthodontists or physicians simply judge facial symmetry. Therefore, maintaining accuracy is difficult. We propose a convolutional neural network with a transfer learning approach for facial symmetry assessment based on 3-dimensional (3D) features to assist physicians in enhancing

Thalamic Reticular Dysfunction as a Circuit Endophenotype in Neurodevelopmental Disorders.

Diagnoses of behavioral disorders such as autism spectrum disorder and schizophrenia are based on symptomatic descriptions that have been difficult to connect to mechanism. Although psychiatric genetics provide insight into the genetic underpinning of such disorders, with a majority of cases explained by polygenic factors, it remains difficult to design rational treatments. In this review, we highlight the value of understanding neural circuit function both as an intermediate level of explanatory description that links gene to behavior and as a pathway for developing rational diagnostics and t

Distinct EEG functional network alterations in Parkinson's disease and multiple system atrophy.

Parkinson's disease (PD) and multiple system atrophy (MSA) are alpha-synuclein-related neurodegenerative disorders with overlapping motor and non-motor symptoms but distinct pathology, clinical management, and prognosis. Early differential diagnosis, particularly between PD and the parkinsonian subtype of MSA (MSA-P), remains a major clinical challenge. Yet, how their distinct underlying neurodegeneration differentially disrupts large-scale neural coordination remains poorly understood, limiting the development of objective neuroimaging biomarkers. Here, we aimed to characterize EEG cortical n

Individuality in Visual Representation of Pulse: A Comprehensive Analysis.

Individuality, a foundational concept of traditional medicine, has been increasingly challenged by the standardization of modern healthcare. The emergence of wearable technology offers a scalable pathway toward precision health. Here, we explored the comprehensive visual representation of wearable photoplethysmography signals from a single device using the phase-amplitude coupling method. We demonstrated that the coupling diagrams contain visually distinguishable regions of physiological frequency components, including those consistent with reference breathing rates (R-squared value of 0.86 fo

Temporal prediction captures retinal spiking responses across animal species.

The retina's role in visual processing has been viewed as two extremes: an efficient compressor of incoming visual stimuli, akin to a camera, or a predictor of future stimuli. Addressing this dichotomy, we developed a spiking neural network model of the retina trained on natural movies under metabolic-like constraints to either encode the present or to predict future scenes. When optimized for efficient temporal prediction ~100 ms into the future, the model not only captures retina-like receptive fields and their mosaic-like organizations, but also exhibits complex retinal processes such as la

Explainable/interpretable artificial intelligence and knowledge graph neural network for synthetic lethality prediction: overview, challenges, and future directions.

Synthetic lethality (SL) provides a powerful foundation for targeted oncology by identifying gene pairs whose simultaneous disruption is fatal to cancer cells while sparing healthy tissue. However, mapping these interactions experimentally across the human genome is restricted by massive combinatorial scale and high costs. To address this, computational approaches specifically graph neural networks (GNNs) have become the standard for modeling these complex biological relationships. Hence, by integrating heterogeneous knowledge graph (KG) and multi-omics data, GNNs can effectively capture the t

Intelligent machine learning solutions with Bayesian regularization backpropagation adaptive networks for differential systems of Maize streak virus diseases.

Maize streak virus, a cause of maize streak disease, poses a major threat to food security that significantly reduces yields of maize, a crucial food crop, thereby jeopardizing the volatile social and economic stability of subsistence growers across the planted field. This communication emphasizes the predictive forecasting of epidemic dynamics by including three control interventions: chemical control, quarantine, and preventive, together with the parameter estimations. The study integrates machine predictive neural networks with Bayesian regularization (NNBNRS) backpropagation for an efficie

Multimodal Autoencoder-Based Anomaly Detection Reveals Clinical-Radiologic Heterogeneity in Pulmonary Fibrosis.

Background: Pulmonary fibrosis (PF) and post-infectious fibrotic lung disease are characterized by marked heterogeneity in radiologic patterns, physiologic impairment, and clinical presentation. Conventional analytic approaches often fail to capture non-linear and multimodal relationships between structural imaging findings and functional limitation. Integrating imaging-derived representations with clinical and functional data using artificial intelligence (AI) may provide a more comprehensive characterization of disease heterogeneity. Objectives: The objective of this study was to develop and

High-resolution 3D flow reconstruction of cerebrospinal fluid microcirculation using physics-informed neural network: Conceptualization and application to a large animal model.

In this study, we present a novel Physics-Informed Neural Network (PINN) framework that reconstructs 3D flows using planar velocity projections from arbitrarily oriented planes. The method is designed for the reconstruction of low-Reynolds-number flows typical of cerebrospinal fluid (CSF) in the subarachnoid space (SAS). The method utilizes a projection loss function combined with gradient smoothing regularization during network training. We show that plane orientation with perturbations of 0.05 (relative to the main flow axis) or greater is sufficient axial data for accurate reconstruction. A
