Effect in rats of simultaneous prenatal exposure to ochratoxin A and aflatoxin B1. I. Maternal toxicity and fetal malformations.

Ochratoxin A (OA) and Aflatoxin B1 (AFB1), the food borne mycotoxins are produced by several fungal species of the genera Aspergillus and Penicillium. To determine the teratogenic effects, these mycotoxins were administered orally either individually or in combination to the pregnant Wistar rats on days 6-15 of gestation. OA and AFB1 were dissolved in corn oil and different doses of OA (0.125, 0.25, 0.50, and 0.75 mg/kg), AFB1 (0.125, 0.25, 0.50, and 1.00 mg/kg), and a combination of OA+AFB1 (0.125+0.125; 0.25+0.50; 0.50+0.25 mg/kg) were given by gastric intubation to rats. During dosing perio

Determinants of aflatoxin exposure in young children from Benin and Togo, West Africa: the critical role of weaning.

BACKGROUND: Dietary exposure to high levels of the fungal toxin, aflatoxin, occurs in West Africa, where long-term crop storage facilitates fungal growth. METHODS: We conducted a cross-sectional study in Benin and Togo to investigate aflatoxin exposure in children around the time of weaning and correlated these data with food consumption, socioeconomic status, agro-ecological zone of residence, and anthropometric measures. Blood samples from 479 children (age 9 months to 5 years) from 16 villages in four agro-ecological zones were assayed for aflatoxin-albumin adducts (AF-alb) as a measure of

Intranasal instillation of aflatoxin B(1) in rats: bioactivation in the nasal mucosa and neuronal transport to the olfactory bulb.

Aflatoxin B(1) (AFB(1)) may be present in moldy dust. Inhalation of contaminated dust particles may result in high local exposure of the nasal mucosa. The present study was designed to assess bioactivation and toxicity of AFB(1) in the nasal mucosa after intranasal administration of the mycotoxin in rats and also to examine if translocation of the mycotoxin occurs from the nasal mucosa to the brain along olfactory neurons. Female Sprague-Dawley rats were given (3)H-AFB(1) (0.2, 1 or 20 microg) intranasally and were sacrificed at various intervals (1 h to 20 d). Tissues were examined autoradiog

Effects of volatile aldehydes from Aspergillus-resistant varieties of corn on Aspergillus parasiticus growth and aflatoxin biosynthesis.

The fungi Aspergillus flavus and Aspergillus parasiticus produce a potent class of hepatocarcinogens known as aflatoxins. Corn-derived volatile compounds have been previously found to affect growth and aflatoxin production in A. flavus. In this study, the effects on A. parasiticus of three corn-derived volatile compounds, n-decyl aldehyde, hexanal and octanal, were measured. These three compounds were previously found to be variably expressed in five Aspergillus-resistant maize strains and three susceptible strains. In this study, A. parasiticus radial growth was restricted least by n-decyl al…

Experimental induction of liver fibrosis in young guinea pigs by combined application of copper sulphate and aflatoxin B1.

Aflatoxin B1 alone (0.05 mg resp. 0.037 mg/kg/d), copper alone (6.6 mg/kg/d or 200 mg/l drinking water) or a combination of both was administered orally for 6 months to young guinea pigs from the first/second day of life. In the copper group there were no pathomorphological changes. For the aflatoxin B1 group, liver damage was established. In the combined group, liver injury was more frequent and more severe compared to the aflatoxin B1 group and biliary copper excretion was diminished compared with the copper group. Histologically, only the livers of this group exhibited degeneration, atrophy

Protection against aflatoxin B1-induced hepatic toxicity as short-term screen of cancer chemopreventive dithiolethiones.

Dithiolethiones are an important class of cancer chemopreventive agents. More than 50 new dithiolethione analogs were synthesized for structure-activity studies. Using selected dithiolethiones, studies were designed to measure protection against the hepatotoxicity of aflatoxin B1 (AFB1) and relate it to the protection against carcinogenicity. Young male F344 rats were pretreated with 0.1 or 0.3 mmol dithiolethiones/kg body wt and challenged with toxic doses of AFB1 (50 micrograms/100 g rat/day) on 2 successive days. One day later, the protection from hepatotoxicity was assessed by measuring se

Overexpression of cytochrome P-450 isoforms involved in aflatoxin B1 bioactivation in human liver with cirrhosis and hepatitis.

Studies were carried out to test the hypothesis that inflammatory liver disease increases the expression of specific cytochrome P-450 isoenzymes involved in aflatoxin B1 (AFB) activation. The immunohistochemical expression and localization of various human cytochrome P-450 isoforms, including CYP2A6, CYP1A2, CYP3A4, and CYP2B1, were examined in normal human liver and liver with hepatitis and cirrhosis. The constitutive expression of CYP3A4 in normal liver showed a characteristic pattern of distribution in centrilobular hepatocytes, whereas CYP1A2, CYP2A6, and CYP2B1 were expressed uniformly th

Fatty and hemorrhagic liver and kidney syndrome in breeding hens caused by aflatoxin B1 and heat stress in the Sudan.

Fatty liver in animals is commonly associated with toxic or nutritional disease. In tropical Africa, fatty liver in poultry is of considerable economic importance. However, it's occurrence and association with renal damage have not yet been reported. We now report the effects of hemorrhagic and fatty hepatorenal syndrome in Hybro breeding hens naturally exposed to heat stress and fed low level of aflatoxin B1.

A dual-monomer magnetic imprinted sorbent for fast removal and trace-level monitoring of hazardous aflatoxin B1.

Aflatoxin B1 (AFB1) is a highly toxic contaminant commonly found in foods, and its trace detection faces severe interference from complex matrices. To achieve efficient enrichment and rapid detection of AFB1, this study designed and synthesized a magnetic dual-functional monomer molecularly imprinted polymer (MMIP) with magnetic nanoparticles as the core and dummy templated ECC as the imprinting molecule. Through surface imprinting technology combined with methacrylic acid and styrene as dual functional monomers, an imprinted layer with specific recognition sites was constructed, while systema

Linking Phytochemical Diversity to Aflatoxin Suppression: LC-MS/MS Metabolomics of Trichilia dregeana Bark Extracts.

Trichilia dregeana has a rich phytochemical diversity and biological activity; however, information on its metabolomic profile and antimycotoxigenic potential is limited. This study investigated different extracts of T. dregeana bark obtained with various solvents (water, ethanol, ethyl acetate, and methanol), assessing their chemical composition using LC-MS and their inhibitory activity against the aflatoxin produced by Aspergillus fungi. LC-MS analysis identified metabolites belonging to several secondary metabolite classes, including flavonoids, phenolic acids, lignan glycosides, cardiac gl

D-shaped optical fiber lossy mode resonance biosensor for label-free detection of aflatoxin B1.

Aflatoxin B1 (AFB1) is a highly toxic mycotoxin that poses a serious threat to food safety and human health. To achieve rapid, sensitive and specific detection of AFB1, a label-free D-shaped optical fiber sensor based on lossy mode resonance (LMR) was developed in this study. The sensor was fabricated by depositing a SnO2 thin film on the polished surface of a D-shaped optical fiber as the LMR sensitive layer and immobilizing anti-AFB1 antibodies on the SnO2 surface to construct a specific biorecognition interface. The specific binding between AFB1 in the sample and the immobilized anti-AFB1 a

Contact Occupancy and Per-Residue Energetic Contributions Distinguish Aflatoxin B1 Nanobodies with Different Binding Affinities.

Aflatoxin B1 (AFB1) is a potent foodborne mycotoxin for which nanobodies offer compact, engineerable detection reagents, yet the molecular basis of their affinity differences remains poorly defined. Here, closely related nanobodies G8, NB28, and NB26 were compared. Fluorescence correlation spectroscopy showed G8 had the lowest dissociation constant for AFB1-BSA, followed by NB28 and NB26. To resolve this affinity separation, we performed triplicate 3 μs molecular dynamics simulations for each complex. While all complexes remained stable, G8 maintained broader, persistent aromatic and hydrophob

Aflatoxin B1 Toxicity in Animal Models: Biomarker-Guided Mechanisms, Systemic Injury, and Precision Mitigation Strategies.

Aflatoxin B1 (AFB1) is a highly toxic mycotoxin which can be carried over into animal products and cause deterioration of livestock productivity when fed to livestock and wildlife. This review proposes a biomarker-guided framework for improving the early assessment of AFB1 exposure and toxicological responses in animal models. Oral exposure leads to the absorption of AFB1, which is bioactivated in the liver to the reactive AFB1-exo-8,9-epoxide that causes DNA and protein adduct formation, inflammation, mitochondrial apoptosis, and other effects. Cytochrome P450 activation and glutathione-depen

Flavonoids as multi-target protective agents against aflatoxin B1 toxicity: mechanistic convergence and translational challenges.

Aflatoxin B1 (AFB1) is the most widespread and toxic mycotoxin found in food and feed. It is both chemically and physically stable and shows resistance to degradation during processing. AFB1 can cause damage to multiple organs, including the liver, immune system, nervous system, kidneys, reproductive system, intestines, and genetic material. Therefore, the development of a safe and effective antidote is urgently needed. Flavonoids are a class of natural polyphenols widely found in fruits, vegetables, and herbs, which can mitigate AFB1-induced organ toxicity through various mechanisms. However,
