Inhibition of proliferation and differentiation of human B-lymphocytes by a biscoclaurine alkaloid.

A biscoclaurine alkaloid, cepharanthine, is known to be a potent inhibitor of snake venom-induced hemolysis by interaction with the lipid bilayer of the membrane. The drug also interferes with the ion channel intracellularly. In this study, we examined the effect of cepharanthine on human B-cell functions. Small dense B-cells from tonsil samples were isolated using a Percoll density gradient from non-rosetted cells and were used as the target cells. Cepharanthine inhibited the proliferation of the lymphocytes and antibody production of human B-lymphocytes. The inhibitory effect of cepharanthin

A comparison of the catecholamine releasing effect of the khat alkaloids (-)-cathinone and (+)-norpseudoephedrine.

Consumption of khat leaves produces symptoms that are similar to those caused by amphetamine. The experiments reported in the following describe the effect of the two khat alkaloids (-)-cathinone and (+)-norpseudoephedrine at the cellular level. Both were shown to induce release from physiological catecholamine storage sites. Peripheral noradrenaline sites (rabbit heart) were observed to be more sensitive than CNS dopamine sites (rat nucleus accumbens). In the latter organ, which is believed to be involved in amphetamine hypermotility, (-)-cathinone was considerably more potent than (+)-norpse…

The long persistence of pyrrolizidine alkaloid-derived pyrrole-protein adducts in vivo: Kinetic study following multiple exposures of a pyrrolizidine alkaloid containing extract of Gynura japonica.

Gynura japonica (also named Tusanqi in Chinese) is used as a folk herbal medicine for treating blood stasis or traumatic injury. However, hundreds of hepatic sinusoidal obstruction syndrome (HSOS) cases have been reported after consumption of preparations made from G. japonica because it contains large amounts of hepatotoxic pyrrolizidine alkaloids (PAs). To date, blood pyrrole-protein adducts (PPAs) are suggested as biomarkers for the diagnosis of PA-induced HSOS in clinics. However, the concentration of PPAs in the blood is greatly affected by several factors including the amount of PA expos…

Promising approaches for simultaneous enhancement of medicinally significant benzylisoquinoline alkaloids in opium poppy.

Benzylisoquinoline alkaloids (BIAs) produced in opium poppy have been evidenced to heal patients suffering from various diseases. They, therefore, hold an integral position in the herbal drug industry. Despite the adoption of several approaches for the large-scale production of BIAs, opium poppy remains the only platform in this purpose. The only disadvantage associated with producing BIAs in the plant is their small quantity. Thus, recruiting strategies that boost their levels is deemed necessary. All the methods which have been employed so far are just able to enhance a maximum of two BIAs. …
