The actin-binding protein synaptopodin (Synpo) regulates the cytoskeleton and organization of the endoplasmic reticulum, thereby amplifying intracellular Ca2+ signaling. Knockout (KO) mouse models have been used to study the role of Synpo in kidney and brain functions, where it supports stress fiber formation, as well as long-term potentiation (LTP) and learning, respectively. Here, we generated Synpo KO rats using CRISPR-Cas9, and show they are viable but have reduced body weight after postnatal days 35-45, along with shorter limb bone length. Their basal kidney function is normal into early

Synucleinopathies are a group of neurodegenerative disorders characterized pathologically by the presence of neuronal and/or oligodendrocyte inclusions composed of aggregated alpha-synuclein (αS). These disorders can be divided into two major sub-types: Lewy body disease and multiple system atrophy, which have distinct clinical and pathological profiles. As is seen with other neurodegenerative diseases, recent evidences suggest that the conformation and molecular organization of αS amyloid filaments deposited in brains might dictate the distinct pathological and clinical profiles in synucleino

Prostate cancer remains one of the most common malignant tumors among men worldwide, and treatment options are limited in the advanced stage. To address this challenge, we have developed a pH-responsive copper porphyrin metal-organic framework (CuTCPP-MOF), which integrates photodynamic therapy (PDT) and chemodynamic therapy (CDT). It enables efficient electron transfer and ROS generation. Under 488 nm laser irradiation, MOF acts as a self-sensitizing photosensitizer to generate single-state oxygen (1O2), which gradually releases Cu2+ in an acidic environment. Subsequently, it is reduced to Cu

Cytarabine (Ara-C) is a frontline chemotherapeutic agent for acute myeloid leukemia and related hematological malignancies; however, its narrow therapeutic window, rapid metabolic clearance, and pronounced interpatient variability necessitate the development of robust analytical tools for precise monitoring in both pharmaceutical formulations and biological fluids. To address this challenge, copper-doped nitrogen carbon dots (Cu-NCDs) were engineered for colorimetric and fluorometric Ara-C detection. The Cu-NCDs integrate peroxidase-mimetic activity with inherent fluorescence, enabling simulta

BACKGROUND: Successful recruitment of study participants is a challenging component of research, and recruitment barriers are amplified in safety-net hospital (SNH) settings. However, engaging historically underrepresented groups in research is critically important to improve health disparities and outcomes. We summarize challenges we encountered while recruiting patients with COVID-19 from the emergency department (ED), actions to improve inclusivity, and implementation hurdles in an SNH setting.
METHODS: We conducted an observational study at the largest safety-net hospital in New England,

Luminescent metal-organic frameworks (LMOFs) provide exceptional advantages over conventional materials in detecting harmful gases. However, challenges remain, such as limited stability, unclear fluorescence signal changes and insufficient mechanistic understanding. In this study, we developed two types of lanthanide metal-organic frameworks (Ln-MOFs) exhibiting rapid (<5 s) and reversible fluorescence "turn-on" responses to vapor exposure. Specifically, Ln-TCPE demonstrates a swift response to HCl vapor via a unique dual-channel mechanism: highly dipolar HCl molecules stabilize the conformati

Osteomyelitis caused by methicillin-resistant Staphylococcus aureus (MRSA) biofilms poses a major therapeutic challenge due to persistent infection and bone loss. Optimizing anti-infection and promoting bone repair are the main goals to improve the efficiency of osteomyelitis treatment. Herein, we present an ultrasound (US)-actived Cu-BTO@Gua composite piezoelectric sonosensitizer, created by conjugating guanidine (Gua) groups, a component that permeates the biofilm matrix, onto US-absorbing Cu-doped barium titanate (BTO). The guanidine groups demonstrate strong affinity for matrices abundant

Noninvasive phototherapy with functional preservation is considered to be a promising cancer therapeutic method. However, the clinical application of tumor phototherapy is severely restrained by the lack of appropriate multimodal phototherapy agents exhibiting an ideal tissue penetration depth to maximize the antitumor efficiency as well as to maintain important tissue functions. Herein, an innovative near-infrared ray (NIR)-triggered photodynamic-photocatalytic-photothermal therapy (PDT-PCT-PTT) agent based on an atomically dispersed cobalt single-atom enzyme (Co-SAE) anchored on hollow N-dop

Uric acid (UA), a crucial biomarker for chronic metabolic disorders such as gout and hyperuricemia, necessitates non-invasive monitoring for effective disease management. However, wearable sensor for real-time UA detection face significant challenges due to the low UA concentration (<100 μM) in human sweat and the difficulty of multi-functional integration. Here, we developed an integrated wearable photo-electrochemical (PEC) sensor for UA detection in sweat. The sensor comprises iontophoresis electrodes, micro-channel, PEC detection unit and signal system, successfully integrating the functio

Food freshness monitoring and volatile amine detection are key to food safety. In this study, we demonstrated the applicability of mixed-valence rhenium oxide quantum dots (MV-ReOQDs), synthesized via the hydrothermal reaction of α-cyclodextrin and rhenium ion precursors, in triethylamine (TEA) sensing. Spectroscopic correlation techniques showed that the developed MV-ReOQDs possessed mixed-valent rhenium, α-cyclodextrin as capped ligand, partially carbonized surface, and amorphous phase structure. The multiple oxidation states in the MV-ReOQDs facilitated electron precipitation, contributing

In this work there was investigated the synergistic effect of the nanomaterials-the Montmorillonite (MMT) and the vanadium pentoxide (V2O5) on the polyvinyl alcohol (PVA)/starch composite. The composite films were prepared by the solvent casting method. The characterization of the composites showed that the addition of the MMT and the V2O5 to PVA/starch composite decreased the water solubility and water absorption capacity of the film. Both of the reinforcement materials enriched values of thermal conductivity and thermal stability of the composite. The TG/DTA and universal testing machine (UT

The iron nanozyme-based colorimetric method, which is widely applied for biosubstrate detection in in vitro diagnosis (IVD), faces some limitations. The optimal catalytic conditions of iron nanozymes necessitate a strong acidic environment, high temperature, and other restrictive factors; additionally, the colorimetric results are highly influenced by optical interferences. To address these challenges, iron nanozymes doped with various transition elements were efficiently prepared in this study, and notably, the manganese-modified one displayed a high catalytic activity owing to its electron t

Fluorine NMR is a highly sensitive technique for delineating the conformational states of biomolecules and has shown great utility in drug screening and in understanding protein function. Current fluorinated protein tags leverage the intrinsic chemical shift sensitivity of the 19F nucleus to detect subtle changes in protein conformation and topology. This chemical shift sensitivity can be amplified by embedding the fluorine or trifluoromethyl reporter within a pyridone. Due to their polarizability and rapid tautomerization, pyridones exhibit a greater range of electron delocalization and corre

Maternal smoking, including both traditional cigarettes and electronic ones, is a significant modifiable risk factor associated with adverse perinatal outcomes, especially in twin pregnancies. This narrative review aims to explore the impact of maternal smoking on obstetric and neonatal outcomes in twin pregnancies, which inherently carry a higher risk of complications. A literature search was conducted using the PubMed and EMBASE databases, selecting studies published between January 1994 and October 2024. The findings demonstrate a clear association between smoking and increased risks of pre

Magnesium alloys present a compelling prospect for absorbable implant materials in orthopedic and trauma surgery. This study evaluates an ultra-high purity, lean magnesium-calcium alloy (X0), both with and without plasma electrolytic oxidation (PEO) surface modification, in comparison to a clinically utilized WE43 magnesium alloy. It is shown that the mechanical properties of X0 can be tuned to yield a high-strength material suitable for bone screws (with an ultimate tensile strength of 336 MPa) or a ductile material appropriate for intraoperatively deformable plates (with an elongation at fra

Four of seven Patagonian maras (Dolichotis patagonum) at a zoological institution developed acute neurologic signs that progressed to tetraparesis and death. All affected were young adult females (10 mon-5 yr old) that presented over 11 d. Clinical signs were rapidly progressive and unresponsive to supportive therapies. Two of the four individuals were found deceased 4 d after hospitalization. Two individuals were euthanized due to poor prognosis and decline after 6 and 8 d, respectively. Simultaneously, an additional mara developed mild and self-resolving clinical signs, including a kyphotic

Herein, we report a supersensitive and specific detection of Salmonella employing nanocatalysis of silver nanoparticle (AgNp). A nanobioprobe was developed employing specific antibody (Ab) that binds to a peptide present in transmembrane protein of Salmonella. We have studied 7 surface-exposed peptide hits from conserved virulence proteins (PagC, ST50, PagN, CdtB and FliC). These peptides were experimentally evaluated by BLI (Bio layer interferometry) for their reactivity towards antisera raised against an admix of major Salmonella serogroups. The most promising peptide was used to generate Ab

Sesamol (SM) is a potent natural antioxidant that can quench free radicals and modulate the cholinergic system in the brain, thereby ameliorating memory and cognitive impairment in Alzheimer's disease patients. Moreover, the total antioxidant capacity can be amplified by synergistic interactions between different antioxidants. Here, we constructed a ternary heterojunction graphitic carbon nitride/cupric sulfide/titanium dioxide (g-C3N4/CuS/TiO2) photoelectrochemical (PEC) sensor for the quantification of SM and its synergistic interactions with other antioxidants. Crucially, the Schottky barri

The elimination of formaldehyde at room temperature holds immense potential for various applications, and the incorporation of a catalyst rich in surface hydroxyl groups and oxygen significantly enhances its catalytic activity towards formaldehyde oxidation. By employing a coprecipitation method, we successfully achieved a palladium domain confined within the manganese carbonate lattice and doped with iron. This synergistic effect between highly dispersed palladium and iron greatly amplifies the concentration of surface hydroxyl groups and oxygen on the catalyst, thereby enabling complete oxid

Hyperconjugative aromaticity (HA) frequently appears in metalla-aromatics, but its effect on photophysical properties remains unexplored to date. Herein, we reveal two different HA scenarios in nearly isostructural triaurated indolium and benzofuranylium compounds. The biased HAs show a discernible effect on the spatial arrangement of metal atoms and thus tailor metal parentage in frontier orbitals and the HOMO-LUMO energy gap. Theoretical calculations and structural analyses demonstrate that HA not only influences the degree of electron delocalization over the trimetalated aromatic rings but
