Clinical and Histological Effects of Calcium Hydroxyapatite Filler in the Orofacial Region: A Study in Rats.

BACKGROUND: Calcium hydroxyapatite (CaHA) dermal filler has been increasingly used in facial aesthetic procedures. OBJECTIVE: To investigate clinical and histological changes associated with calcium hydroxyapatite (CaHA) dermal filler in the orofacial region. MATERIALS AND METHODS: Forty-eight female Wistar rats were divided into CaHA and control groups. The material was applied in the ventral tongue and the submandibular region; the animals were euthanized after 7, 30, and 90 days. RESULTS: After 7 days, yellowish nodules with a firm consistency were observed on the tongue. In 2 animals, t

Mutant α-synuclein and aging reduce neurogenesis in the acute 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine model of Parkinson's disease.

Neurogenesis, the production of new neurons from less differentiated precursor cells, normally occurs in adult brains in the subventricular zone (SVZ) of the lateral ventricles and the subgranular zone of the hippocampal dentate gyrus. Neurogenesis declines with aging. In previous studies, neurogenesis was stimulated by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine hydrochloride (MPTP) in young animals. In this study, we examined the effect of acute MPTP administration and mutant α-synuclein A53T on neurogenesis and migration of newborn neurons in the aged (23-month) vs. young (2-month) rodent

Targeting super-enhancer-driven SKIL transcription by CDK7 inhibitor THZ1 to suppress gastric cancer progression.

BACKGROUND: Gastric cancer (GC) is a lethal malignancy characterized by high incidence, mortality, and limited treatment options. Transcriptional addiction is a key cancer hallmark that drives tumor pathogenesis, making its inhibition a promising therapeutic strategy for GC. The study aims to investigate the roles and mechanisms of super-enhancer (SE)-driven oncogenic transcriptional addiction in GC progression and to identify novel targetable vulnerabilities. METHODS: We utilized cellular and animal models to assess the effects of THZ1 treatment and CDK7 knockdown on GC progression. RNA seque

Amniotic Membrane (AM) Covered Airway Stent Mitigates Airway Injury and Complications: A Comparative In Vivo Study in a Porcine Model.

BACKGROUND: Airway stenting is a critical intervention to restore airway patency, yet complications such as stent migration, mucostasis, airway injury, and granulation tissue formation remain major concerns. Although various covering materials have been explored, the effectiveness of amniotic membrane (AM) as a stent cover has not been evaluated. This study is the first to assess airway-related complications of a novel AM-covered self-expandable metallic stent (SEMS) compared with a commercially available synthetic polymer-covered SEMS. To compare the incidence of stent-related complications b

OSBPL3 promotes colorectal cancer progression through modulation of TGF-β signaling and immune cell infiltration.

BACKGROUND: Colorectal cancer (CRC) is the third most common cancer worldwide, with prognosis often limited by disease progression. The role of oxysterol-binding protein-like 3 (OSBPL3) in various cancers remains unclear. This study aimed to investigate the expression, biological function, and relationship of OSBPL3 with the tumor microenvironment (TME) in CRC, and to assess its potential as a therapeutic target. METHODS: We employed bioinformatic analysis, immunohistochemistry (IHC), Cell Counting Kit-8 (CCK-8) assay, Western blot (WB), ethynyl deoxyuridine (EdU) incorporation assay, wound h

Characterizing trajectories of innate immune cells in larval zebrafish.

It is well established from in vitro studies of immune cells that stimulation by a wide range of potential signals leads to motility and morphology changes. How these physical behaviors manifest inside a living animal remains unclear due to limitations of conventional imaging and analysis approaches. Here, we establish a quantitative framework for imaging and tracking neutrophil and macrophage dynamics in larval zebrafish, spanning a large fraction of the animal for multi-hour timescales with few-minute temporal resolution. We focus especially on the gut, examining innate immune responses to d

Determinants of skin sensitisation potential.

Skin sensitisation is an important toxicological endpoint. The possibility that chemicals used in the workplace and in consumer products might cause skin sensitisation is a major concern for individuals, for employers and for marketing. In European REACH (Registration, Evaluation, and Authorisation of Chemicals) legislation, the sensitising potential should therefore be assessed for chemicals below the 10 ton threshold. Development of methods for prediction of skin sensitisation potential without animal testing has been an active research area for some time, but has received further impetus wi

Detection of cochlear dysfunction by the measurement of transiently evoked otoacoustic emissions in guinea pigs with autoimmune-induced labyrinthitis.

For the evaluation of functional inner ear lesions caused by an autoimmune-induced labyrinthitis, transiently evoked otoacoustic emissions (TEOAEs) were detected in guinea pigs before and after transfer of sensitized lymphocytes from animals suffering from a labyrinthitis induced by the foreign protein keyhole limpet hemocyanin (KLH). Initially TEOAEs were detectable from 47 of 62 ears (76%) in healthy guinea pigs. These animals then were used as recipients for sensitized lymphocytes from donors exclusively. Three months after cell transfer the incidence of TEOAEs was reduced to 48% in animals

Reproductive and morphological traits distinguish early- and late-maturing Pacific lamprey (Entosphenus tridentatus).

Pacific lamprey, an anadromous fish native to the North Pacific Ocean, are imperiled in parts of their range. Many aspects of their biology are not fully understood, including the physiology of their spawning migration. Pacific lamprey exhibit two divergent reproductive life histories: early-maturing and late-maturing. Interior Columbia River early-maturing lamprey complete the final stages of gonadal maturation ~ 10 months after entering freshwater. From the same migrating cohort, the late-maturing lamprey wait ~ 22 months to complete maturation. This study sought to characterize the reproduc

Bull trout (Salvelinus confluentus) exhibit binge-feeding and digestive flexibility during seasonal resource pulses associated with salmon migrations.

Resource pulses are infrequent, ephemeral events of resource hyperabundance that can represent important feeding opportunities for consumers. To capitalize on pulsed resources, consumers can exhibit behavioural and physiological traits including binge-feeding and phenotypic plasticity of digestive physiology, although expression of these traits has not been observed simultaneously. Further, past studies of binge-feeding have largely focused on times and locations where resources were highly concentrated, ignoring potential temporal and spatial variation in consumer responses. We investigated t

Cryptochrome 4b protein is probably irrelevant for radical pair-based magnetoreception in the European robin.

Avian cryptochrome 4 (Cry4) protein is a putative magnetosensitive molecule facilitating precise long-distance navigation in migratory birds. Two splice variants of Cry4 have been reported in the European robin (Erithacus rubecula), namely ErCry4a and ErCry4b. ErCry4a protein is known to generate magnetically sensitive radical pairs for magnetoreception through electron transfer between the flavin adenine dinucleotide (FAD) cofactor and tryptophan residues within the protein. However, little is known about the ErCry4b isoform. We therefore characterize the properties of ErCry4b to determine wh

Reduction of coastal lighting decreases seabird strandings.

Artificial light at night (ALAN) is negatively impacting numerous species of nocturnally active birds. Nocturnal positive phototaxis, the movement towards ALAN, is exhibited by many marine birds and can result in stranding on land. Seabird species facing major population declines may be most at risk. Leach's Storm-Petrels (Hydrobates leucorhous) are small, threatened seabirds with an extensive breeding range in the North Atlantic and North Pacific Oceans. The Atlantic population, which represents approximately 40-48% of the global population, is declining sharply. Nocturnal positive phototaxis

Sod farms drive habitat selection of a migratory grassland shorebird during a critical stopover period.

Migratory shorebirds are one of the fastest declining groups of North American avifauna. Yet, relatively little is known about how these species select habitat during migration. We explored the habitat selection of Buff-breasted Sandpipers (Calidris subruficollis) during spring and fall migration through the Texas Coastal Plain, a major stopover region for this species. Using tracking data from 118 birds compiled over 4 years, we found Buff-breasted Sandpipers selected intensively managed crops such as sod and short-stature crop fields, but generally avoided rangeland and areas near trees and

Migrating Porcupine Quills: Sudden Death of a Yearling American Black Bear (Ursus americanus) at a Wildlife Rehabilitation Center.

A yearling, intact, apparently healthy male American black bear (Ursus americanus) died peracutely at a rehabilitation facility in Ontario, Canada while overwintering, after recovery from porcupine quilling a few months previously. The postmortem examination findings support that porcupine quill migration should be a differential for cause of sudden death in wildlife.

Incorporating multidimensional behavior into a risk management tool for a critically endangered and migratory species.

Conservation of migratory species exhibiting wide-ranging and multidimensional behaviors is challenged by management efforts that only utilize horizontal movements or produce static spatial-temporal products. For the deep-diving, critically endangered eastern Pacific leatherback turtle, tools that predict where turtles have high risks of fisheries interactions are urgently needed to prevent further population decline. We incorporated horizontal-vertical movement model results with spatial-temporal kernel density estimates and threat data (gear-specific fishing) to develop monthly maps of spati

Application of exponential random graph models to determine nomadic herders' movements in Senegal.

Understanding human and animal mobility patterns is a key to predict local and global disease spread. We analysed the nomad herds' movement network in a pilot area of northern Senegal and used exponential random graph models (ERGM) to investigate the reasons behind these movements. We interviewed 132 nomadic herders to collect information about nomad herd structures, movements, and reasons for taking specific routes or gathering in certain areas. We constructed a spatially explicit network with villages as the nodes and nomad herds' movements as the connecting edges. The final ERGM showed that

New observations - with older ones reviewed - on mass migrations in millipedes based on a recent outbreak on Hachijojima (Izu Islands) of the polydesmid diplopod (Chamberlinius hualienensis, Wang 1956): Nothing appears to make much sense.

Mass aggregations and migrations of millipedes despite numerous attempts to find causes for their occurrences are still an enigma. They have been reported from both southern and northern hemisphere countries, from highlands and lowlands of both tropical and temperate regions and they can involve species belonging to the orders Julida and Spirobolida, Polydesmida and Glomerida. According to the main suggestions put forward in the past, mass occurrences in Diplopoda occur: (1) because of a lack of food and a population increase beyond sustainable levels; (2) for the purpose of reproduction and i

How far north are migrant birds transporting the tick Ixodes scapularis in Canada? Insights from stable hydrogen isotope analyses of feathers.

Lyme disease is emerging in Canada because of northward range expansion of the tick vector Ixodes scapularis. It is hypothesised that I. scapularis feeding on passerine birds migrating north in spring are important in founding new I. scapularis populations leading to northward range expansion. However, there are no studies on how far north I. scapularis may be carried, only inferences from passive tick surveillance. We used stable hydrogen isotope (δ(2)H) analysis of rectrices collected from northward migrating, I. scapularis-carrying, passerine birds captured in Canada to estimate how far nor

Resolving the roles of immunity, pathogenesis, and immigration for rabies persistence in vampire bats.

Bats are important reservoirs for emerging infectious diseases, yet the mechanisms that allow highly virulent pathogens to persist within bat populations remain obscure. In Latin America, vampire-bat-transmitted rabies virus represents a key example of how such uncertainty can impede efforts to prevent cross-species transmission. Despite decades of agricultural and human health losses, control efforts have had limited success. To establish persistence mechanisms of vampire-bat-transmitted rabies virus in Latin America, we use data from a spatially replicated, longitudinal field study of vampir
