Electroretinogram response changes in a guinea pig myopia model treated with different atropine concentrations.

PURPOSE: We used electroretinography (ERG) to assess whether different concentrations of atropine alter the electrical activity of retinal cells during inhibition of myopia progression. MATERIAL AND METHODS: Myopia was established in 14-day-old guinea pigs using -10.00 diopter spectacle lenses in the right eye. Topical atropine eye drops (0.01% and 1%) and sterile water (vehicle) were subsequently administered to the right eyes starting at 2 weeks postnatally. All animals in each group underwent refraction and ERG measurements at weeks 2, 4, 6, and 8. RESULTS: The degree of myopia decreased …

Evaluating the effect of 0.125% atropine on foveal microvasculature using optical coherence tomography angiography.

This study investigated alterations in the foveal microvasculature following the administration of 0.125% atropine in myopic children. In this prospective study, 63 eyes from 36 individuals aged 5-18 years with myopia were administered 0.125% atropine for myopia control. After administration, foveal microvascular parameters such as the area and perimeter of the foveal avascular zone (FAZ), the acircularity index (AI) of FAZ, and foveal vessel density were evaluated longitudinally. The effect of atropine on foveal microvasculature was analyzed using a linear mixed model. One month after atropin…

An Ocular Exposure Prediction for Topical Atropine in Human Using Physiologically Based Pharmacokinetic Modeling.

Developing a mathematical model to predict the distribution and bioavailability of atropine in human eyes is an insight approach for clinical practice. This study aims to develop a human ocular physiologically based pharmacokinetic (PBPK) modeling for the ophthalmic drug atropine and explore possible mechanisms by which atropine reduces myopia progression in children. The Ocular Compartment Absorption and Transit (OCAT™) model was employed to describe the ocular distribution of atropine following administration at different dosages in both rabbits and humans. The PBPK model enables the extrapo…

Analysis of treatment response about low-dose (0.01%) atropine eye-drops in myopic children.

INTRODUCTION: Myopia usually commences during primary school and progresses until the mean age of 16 years. Topical low-dose (0.01%) atropine eye-drop appears to be safe and efficacious for myopia control in children. However, in some cases, a higher concentration of atropine is required in some cases because low-dose atropine treatment is not effective. METHODS: This is a retrospective study among young myopic children between 5 and 15 years with myopia progression > 0.50 D/year. We selected patients treated with low-dose atropine (0.01%) eye-drops for 12 months and conducted a comparative a…

Comparison of microvolt T-wave alternans measurements using atrial pacing compared to atropine administration.

BACKGROUND: Microvolt T-wave alternans (MTWA) has been associated with malignant ventricular arrhythmias in patients (pts) with structural heart disease. MTWA has been shown to be a strong heart rate-dependent arrhythmia marker. However, in clinical practice some pts in which MTWA should be assessed are unable to perform physical exercise to increase heart rate due to various reasons. METHODS: In this study, we investigated the feasibility of noninvasive MTWA measurement by using intravenous atropine to increase heart rate and compared the results to MTWA measurement by right atrial (RA) paci…

Oesophago-gastric junction opening function: assessment using ultrasound imaging and the effects of atropine.

Relaxation and opening of the oesophago-gastric junction (OGJ) is crucial for oesophageal transport. A novel ultrasound technique was used to determine OGJ opening, before and after atropine, in 12 normals. An ultrasound probe, a solid-state pressure transducer and an infusion tube were placed inside a 20-mm diameter bag, which was placed across the OGJ. At various bag pressures ultrasound transducer was pulled across the bag. Acquired B-mode ultrasound images were converted into M-mode image to display the oesophagus, OGJ and stomach. At low bag-pressure (< 20 mmHg), the OGJ but not oesophagu…

[Holter monitoring, exercise test and atropine test in isolated congenital atrioventricular block in children].

Twenty-four patients with isolated congenital heart block were investigated by 24-hour Holter monitoring at an average age of 9.3 +/- 5.5 years. Six patients were symptomatic and 18 were asymptomatic. Eight asymptomatic patients underwent exercise stress tests and an atropine test was performed in 10 asymptomatic patients to evaluate the capacity to accelerate the heart rate. The symptomatic patients were older than the asymptomatic patients. None of the parameters which analyse ventricular rate were significantly different in the two groups of patients. Significant ventricular arrhythmias (Lo…

[Physiopathologic mechanism of ventricular fibrillation induced by atropine. Report of two cases (author's transl)].

The authors report on two cases of ventricular fibrillation induced by bolus of atropine injected for diagnostic purpose. The electrophysiologic effect of adrenergic stimulation following the parasympathetic blockade, is probably the responsible mechanism rather than myocardial ischemia in the first patient, affected by complete a-v block with narrow QRS. The second patient, affected by coronary heart disease and suspected sick sinus syndrome, had more probably a ventricular fibrillation because of a worsening of the underlying myocardial ischemia due to the increase of the cardiac rate. the r…
