Preclinical Testing of 3D Printed, Cell Loaded Hydrogel Based Corneal Substitutes on Rabbit Model.

Many people lose their vision due to corneal stroma injuries of the eye and the golden solution is transplantation of allografts from donors. Unfortunately, the limited availability of donor tissue, risk of disease transmission, and immune rejection are serious handicaps. However, implants made of biomaterials can be used as substitutes. In this study, cell-loaded and cell-free, methacrylated gelatin (GelMA) implants are 3D printed and tested under in vitro conditions. The samples are physically characterized for their printability, equilibrium water content, compressive mechanical strength, a

Advances in the application of smart materials in the treatment of ophthalmic diseases.

Smart materials dynamically sense and respond to physiological signals like reactive oxygen species (ROS), pH, and light, surpassing traditional materials such as poly(lactic-co-glycolic acid), which have high drug loss rates and limited spatiotemporal control. These innovative materials offer new strategies for ophthalmic treatments, with core advantages including targeted delivery via ROS-sensitive nanocarriers, precise regulation through microvalves, and multifunctional integration, such as glucose-responsive contact lenses that create a "sensing-treatment" loop. However, challenges remain,

Optimization of fused filament fabrication process parameters to improve the compressive properties of PEEK and PEKK biomaterials.

Fused filament fabrication (FFF) is increasingly being adopted to create polymeric orthopedic devices. FFF process parameters determine the mechanical performance of final printed parts; hence, optimizing for appropriate levels of strength is critical for load-bearing applications. We utilized a Taguchi L-9 orthogonal array to investigate the impact of nozzle temperature (TN), chamber temperature (TCh), layer height (LH), and print speed (PS) on the compressive properties of cylindrical polyether-ether-ketone (PEEK) and polyether-ketone-ketone (PEKK). The printed specimens were examined using

Biological responses to biomaterials: a review.

Biomaterials stimulate diverse biological responses, including inflammation, wound healing, foreign body reactions, and fibrous encapsulation, all critical for evaluating biocompatibility and effectiveness. These responses are influenced by the material's physicochemical and biological properties, such as composition, texture, and surface characteristics. Adverse reactions, such as severe inflammation or fibrous encapsulation, can hinder tissue integration, jeopardizing patient health and increasing healthcare costs. This review aimed to summarize the current scientific evidence on biological

Hydrogel Foams Containing Superabsorbent Particles for Wound Care Applications.

Herein, we report an innovative approach to design hydrogel foams by embedding superabsorbent polymer (SAP) particles with the aim of improving wound dressing applications. A cross-linking process was used to prepare hydrogel foams by just adding 3 components: an NCO-terminated three-arm prepolymer, SAP particles, and water. Variations in the chemical compositions allow creation of hydrogel foams with different material properties that can be applied to wounds with different characteristics. Hydrogel foams that are stable to electron beam sterilization are mechanically stable and have adjustab

Bioresorbable High-Strength HA/PLLA Composites for Internal Fracture Fixation.

In modern surgery, the internal fixation plates fabricated from hydroxyapatite/poly(L-lactide) (HA/PLLA) composites encounter clinical limitations in fracture treatment due to their inadequate mechanical properties. In this work, pressure-induced flow (PIF) technique is employed to address this limitation. Under optimal processing conditions (140 °C and 250 MPa), the HA/PLLA composites exhibit an impressive flexural strength of 199.2 MPa, which is comparable to that of human cortical bone, the strongest bone tissue in the body. The tensile strength and the notched Izod impact strength are clos

Effect of 20-μm-Thin Ceramic Coatings of Hydroxyapatite, Bioglass, GB14 and β-TCP With Copper on the Ingrowth Behavior of Femoral Implants.

Aseptic loosening and infection are still the main reasons for revision surgery. The contact of the implant is made through the surface, which is coated with calcium phosphate ceramics for better integration into the bone. The aim of the present work is to optimize these coatings by making them thinner (20 μm) and doping them with copper to add antimicrobial functionality. Four different coating materials were used: hydroxyapatite, Bioglass, GB14, and beta tricalcium phosphate. The coatings were applied by high velocity suspension flame spraying. The titanium rods with the coatings were then i

Feasibility of intratracheal tracheostomy sealing: anatomical adaptation and biocompatibility of a second-generation prototype in cadaveric and porcine models.

After decannulation, air leak through the tracheostoma can impair cough and voice. We evaluated a second-generation temporary intratracheal sealing disc that seals the stoma from within. Two preclinical sub-studies were performed: Sub-study 1 cadaveric deployment to assess anatomical fit, seal integrity, and removability; and Sub-study 2, a 7-day porcine implantation with anatomical compatibility and blinded histology. In cadavers, the sealing disc maintained airway patency and sealed the stoma, withstanding peak intratracheal pressures up to 180 cmH2O; removal through a near-healed opening re

A high frequency ultrasound-based platform to non-destructively quantify geometric, acoustic and mechanical properties of thin, engineered soft connective tissues in vitro.

Assessment of temporal changes in the functional properties of in vitro tissue engineered (TE) constructs is typically done by destructive endpoint analysis, which increases experimental resources, analysis time, and financial burden. To overcome the limitations of existing practices, a novel Modular High Frequency Ultrasound Bulge Testing System (mHFUS-BTS) was designed to perform repeated, non-destructive and non-invasive characterization of cell-seeded biomaterial sheets throughout culture. Quantitative HFUS was validated to accurately measure the acoustic properties (i.e., speed of sound,

Effect of non-contact induction heating on HA coatings and bone cement, an ex vivo study.

BACKGROUND: Prosthetic joint infection is a serious complication that can arise after total joint replacement surgery. When bacteria colonise an orthopaedic implant, they form biofilms that protect them from their environment, making them difficult to remove. Treatment is further complicated by a global rise of antimicrobial resistance. These protective mechanisms make treatment of prosthetic joint infection increasingly complex. Non-contact induction heating is an upcoming technology that uses heat to eradicate bacteria that are present on the surface of metallic implants. This study aims to

Self-Propelled Tubular Micromotors Powered by Hydrogen Bubbles under Mild Conditions: A Major Step toward Biological Applications with Live Cells.

Polymer-based tubular micromotors, featuring an inner layer of Pt nanoparticles (PtNPs), exhibit vigorous propulsion by emitting H2 bubbles in an aqueous ammonia borane (NH3BH3) solution. The hydrolysis of NH3BH3 on the PtNPs facilitates the continuous release of H2 gas from the open-end terminus, driving its forward movement. Unlike conventional O2 bubbles' systems that rely on hydrogen peroxide (H2O2) as fuel, these micromotors can operate in the presence of live cells within the NH3BH3 medium. Consequently, micromotors functionalized with the lectin concanavalin A demonstrate the capability

Endothelium-Mimicking Materials: A "Rising Star" for Antithrombosis.

The advancement of antithrombotic materials has significantly mitigated the thrombosis issue in clinical applications involving various medical implants. Extensive research has been dedicated over the past few decades to developing blood-contacting materials with complete resistance to thrombosis. However, despite these advancements, the risk of thrombosis and other complications persists when these materials are implanted in the human body. Consequently, the modification and enhancement of antithrombotic materials remain pivotal in 21st-century hemocompatibility studies. Previous research ind

Electrochemically Grown Hole-Rich NiO(OH) Thin Films toward Hole-Mediated Very Fast and Selective Enzyme-Free Electrochemical Sensing of Dopamine under Simulated Environment.

This work aimed to develop an enzyme-free semiconductor-assisted electrochemical technique for the selective detection of the neurotransmitter dopamine. In this case, electrochemically grown nickel oxyhydroxide [NiO(OH)] thin films were chosen to fabricate the sensing platform, i.e., the electrodes. Chronoamperometry was used to deposit the films on indium tin oxide (ITO) coated glass substrates. The films were thoroughly characterized to establish their structure, composition, phase purity, and electrochemical attributes. Electrochemical sensing characteristics were investigated by means of c

Structure, Properties and Degradation of Self-Assembled Fibrinogen Nanofiber Scaffolds.

Self-assembled fibrinogen nanofibers are promising candidates for skin tissue engineering due to their biocompatibility and ability to mimic the native blood clot architecture. Here, we studied the structure-property relationship and degradation of rehydrated fibrinogen nanofibers prepared by salt-induced self-assembly, focusing on the effect of scaffold layering, cross-linking time and freeze-drying. Optimal fiber stability was achieved with cross-linking by formaldehyde (FA) vapor, while treatment with liquid aldehydes, genipin, EDC, and transglutaminase failed to preserve the nanofibrous ar

Effect of Thermomechanical Processing Induced Retained Strain on In Vitro, In Vivo Biodegradation Response and Cytocompatibility of Mg Alloy.

The implications of retained strain on in vitro and in vivo degradation behavior as well as cytocompatibility of the Mg-4Zn-0.5Ca-0.8Mn alloy is comprehensively studied. The retained strain is induced in homogenized specimens by hard-plate hot forging (HPHF) at a temperature (523 K) lower than the recrystallization temperature of the alloy. The retained strain generated during deformation process deteriorated the corrosion response of the deformed alloy as compared to its homogenized counterpart. The strained area of deformed specimen with high dislocation density promoted defect generation (o

Wearable Pressure Sensors Based on MXene/Tissue Papers for Wireless Human Health Monitoring.

Though the widely available, low-cost, and disposable papers have been explored in flexible paper-based pressure sensors, it is still difficult for them to simultaneously achieve ultrahigh sensitivity, low limit and broad range of detection, and high-pressure resolution. Herein, we demonstrate a novel flexible paper-based pressure sensing platform that features the MXene-coated tissue paper (MTP) sandwiched between a polyimide encapsulation layer and a printing paper with interdigital electrodes. After replacing the polyimide with weighing paper in the MTP pressure sensor, the silver interdigi

Corrosion resistance of a Nitinol ocular microstent: Implications on biocompatibility.

Nitinol is commonly used in medical implants due to its unique thermomechanical properties of shape memory and superelasticity. Free nickel has the potential to induce biological responses that may be a concern for permanent implants manufactured from nickel-containing alloys. Although there are extensive reports on the effects of surface treatments on corrosion behavior in cardiovascular Nitinol implants, there is a lack of data on corrosion resistance and impact on biocompatibility for ocular implants. Therefore, the objective of this study was to determine localized corrosion and nickel elu
