Kinetics of DNA Repair in Vicia faba Meristem Regeneration Following Replication Stress.

The astonishing survival abilities of Vicia faba, one the earliest domesticated plants, are associated, among other things, to the highly effective replication stress response system which ensures smooth cell division and proper preservation of genomic information. The most crucial pathway here seems to be the ataxia telangiectasia-mutated kinase (ATM)/ataxia telangiectasia and Rad3-related kinase (ATR)-dependent replication stress response mechanism, also present in humans. In this article, we attempted to take an in-depth look at the dynamics of regeneration from the effects of replication i

Morphologic features of binucleated lymphocytes to assess the diagnosis of persistent B-cell polyclonal lymphocytosis or other mature B-cell neoplasms.

The observation of binucleated lymphocytes (BNLs) on a peripheral blood smear is essential to the diagnosis of persistent polyclonal B-cell lymphocytosis (PPBL). Only a few case reports have mentioned their presence in other contexts, mainly mature B-cell neoplasms such as chronic lymphocytic leukemia or reactive circumstances such as multiple sclerosis undergoing natalizumab treatment. We sought to investigate whether any particular morphologic features of BNL were more specific to PPBL than other diseases. We reviewed peripheral blood smears of a series of patients affected by PPBL or harbor

[Electron microscopy observation on pre- and post-treatment skin of dorsum pedis of chyluric patient treated with heat-clearing and hemostatic drugs--the lymphangial wall damage being the pathogenesis of chyluria].

Heat-clearing and hemostatic drugs were effective in treating chyluria. In order to explore the curative mechanism, a group of 10 in-patients with chyluria were chosen and treated with heat-clearing and hemostatic drugs for 45 days. Some pre- and post-treatment skin from dorsum pedis of the patients were taken off and observed under electron microscope. It showed that the walls of lymphatic capillaries were seriously damaged before treatment and rehabilitated wall after treatment. The pinocytotic vesicles were packed tightly in the endothelial cells of capillaries before treatment and their nu

Characteristics of myocytes isolated from hearts of renovascular hypertensive guinea pigs.

A model of renovascular hypertension has been developed in the guinea pig using the Goldblatt (2-kidney, 1-clip) operation. Systolic and diastolic blood pressures were significantly increased 3 and 7 wk after the operation, but levels fell to control values at 11 wk. The two-dimensional areas of myocytes isolated from the hearts of Goldblatt-operated (GB) animals were larger than those in control cells at 3 wk (cf. 3,397 +/- 87 and 2,208 +/- 125 microns 2, P < 0.01), and the difference was maintained at 7 and 11 wk. No change in cell contraction or relaxation characteristics were seen at eithe

A morphological study of glial cells in the hypoglossal nucleus of the cat during nerve regeneration.

The cat hypoglossal nerve and nucleus have been used as a model for the study of the occurrence and time course of modifications in the size and composition of the perineuronal glial cell population as they relate to cytological changes in the nerve cell body and the initiation and progress of axon regeneration. Animals were killed at 2, 5, 10, 20, 35, 65, and 115 days after crush injury to the hypoglossal nerve. At 5 days after surgery, growth cones and regenerating unmyelinated axons were present at the lesion site, but no conspicuous changes were apparent in the nerve cell bodies. At 10 day

Neuronal adjustments in developing nuclear centers of the chick embryo following transplantation of an additional optic primordium.

Following transplantation of an additional optic primordium into the orbital mesenchyme of chick embryos of approximately 2 days of incubation age, the changes in cell number in the ciliary ganglion, accessory oculomotor and trochlear nuclei were studied at various stages of development. Cell counts were made at 1-day intervals from days 9 through 15 for ciliary ganglion, and from days 13 through 15 for the accessory oculomotor and trochlear nuclei. Cell counts for the ciliary ganglion on days 9 and 11 were similar on the operated and control sides which suggests that grafting of an additional

CEBPD aggravates apoptosis and oxidative stress of neuron after ischemic stroke by Nrf2/HO-1 pathway.

CCAAT enhancer binding protein delta (CEBPD) is a transcription factor and plays an important role in apoptosis and oxidative stress, which are the main pathogenesis of ischemic stroke. However, whether CEBPD regulates ischemic stroke through targeting apoptosis and oxidative stress is unclear. Therefore, to answer this question, rat middle cerebral artery occlusion (MCAO) reperfusion model and oxygen-glucose deprivation/reoxygenation (OGD/R) primary cortical neuron were established to mimic ischemic reperfusion injury. We found that CEBPD was upregulated and accompanied with increased neurolo

Cell type-specific expression of FoxP2 in the ferret and mouse retina.

Although the anatomical and physiological properties of subtypes of retinal ganglion cells (RGCs) have been extensively investigated, their molecular properties are still unclear. Here, we examined the expression patterns of FoxP2 in the retina of ferrets and mice. We found that FoxP2 was expressed in small subsets of neurons in the adult ferret retina. FoxP2-positive neurons in the ganglion cell layer were divided into two groups. Large FoxP2-positive neurons expressed Brn3a and were retrogradely labeled with cholera toxin subunit B injected into the optic nerve, indicating that they are RGCs

NOS expression in nigral cells after excitotoxic and non-excitotoxic lesion of the pedunculopontine tegmental nucleus.

The substantia nigra (SN) receives afferents from cholinergic neurons of the pedunculopontine tegmental nucleus (PPTg), a neuronal population that shows high levels of nitric oxide synthase (NOS), the enzyme responsible for the synthesis of nitric oxide. We have investigated the effects of the injection in PPTg of two neurotoxins, kainic acid (an excitotoxic neurotoxin), and ethylcholine mustard azirinium ion (AF64A, a non-excitotoxic neurotoxin), upon the SN cells of the rat, by using choline acetyltransferase (ChAT) immunohistochemistry as a marker of cholinergic neurons, and nicotinamide ad

Yields and chondrogenic potential of primary synovial mesenchymal stem cells are comparable between rheumatoid arthritis and osteoarthritis patients.

BACKGROUND: Mesenchymal stem cells derived from the synovial membrane (synovial MSCs) are a candidate cell source for regenerative medicine of cartilage and menisci due to their high chondrogenic ability. Regenerative medicine can be expected for RA patients with the inflammation well-controlled as well as OA patients and transplantation of synovial MSCs would also be a possible therapeutic treatment. Some properties of synovial MSCs vary dependent on the diseases patients have, and whether or not the pathological condition of RA affects the chondrogenesis of synovial MSCs remains controversia

Atlas of Plasmodium falciparum intraerythrocytic development using expansion microscopy.

Apicomplexan parasites exhibit tremendous diversity in much of their fundamental cell biology, but study of these organisms using light microscopy is often hindered by their small size. Ultrastructural expansion microscopy (U-ExM) is a microscopy preparation method that physically expands the sample ~4.5x. Here, we apply U-ExM to the human malaria parasite Plasmodium falciparum during the asexual blood stage of its lifecycle to understand how this parasite is organized in three-dimensions. Tropisetron inhibits high glucose-induced calcineurin/NFAT hypertrophic pathway in H9c2 myocardial cells.

OBJECTIVES: Cardiomyocyte hypertrophy is an important structural feature of diabetic cardiomyopathy. Calcineurin/nuclear factor of activated T-cell (NFAT) pathway plays a central role in the pathogenesis of cardiac hypertrophy. The purpose of this study was to investigate the effects of tropisetron, a novel calcineurin inhibitor, on high glucose (HG)-induced cardiomyocyte hypertrophy and its underlying mechanism. METHODS: H9c2 myocardial cells were treated with tropisetron or cyclosporine A 1 h before exposure to HG for 48 h. KEY FINDINGS: Exposure to HG resulted in enhanced cell size, prote

Channelopathies (Olmsted Syndrome)-causing point mutations of TRPV3 disrupt cellular thermal homeostasis in human keratinocytes.

TRPV3 is a thermosensitive and non-selective cation channel activated at physiological temperatures. It is abundantly expressed in skin keratinocytes, and point mutations in TRPV3 cause a rare genetic disorder, a channelopathy known as Olmsted Syndrome (OS). TRPV3-mutations that cause Olmsted Syndrome show major defects in lysosomal functions and many other cellular abnormalities, such as defects in cell adhesion, cell size and mitochondrial metabolism. In this work, we used a library of thermosensitive organelle probes to explore the sub-cellular thermal spectrum of the OS-causing TRPV3 mutan

TRPV4 Orchestrates Distinct Calcium and Cell Growth Responses in Non-Tumorigenic and Clear Cell Renal Carcinoma-Derived Cells.

Clear cell renal cell carcinoma (ccRCC) is the most common subtype of kidney cancer and often exhibits resistance to conventional therapies. Calcium signaling plays a critical role in cancer progression, and the non-selective cation channel TRPV4 (Transient Receptor Potential Vanilloid 4) has been implicated in tumorigenesis across multiple cancer types. However, its specific role in ccRCC remains poorly understood. In this study, we investigated TRPV4 subcellular localization and functional activity in ccRCC-derived cell lines (786-O and Caki-1) compared to non-tumorigenic renal epithelial ce

Blastoid variant of mantle cell lymphoma in palatine tonsil.

Blastoid variant of mantle cell lymphoma (MCL) is an aggressive and extremely rare malignancy. MCL may be diagnosed in lymph nodes and/or extranodal sites exhibiting a poor prognosis. MCL with primary presentation in palatine tonsils has been rarely reported. Herein, we report the case of a 73-year-old man with a painless nodular mass on the right palatine tonsil. A biopsy was performed, and microscopic analysis revealed a neoplasm composed of small to medium sized lymphocytes with finely dispersed chromatin, roundish nucleus and many mitoses. The tumor cells were positive for CD20 (L26), CD5

Downregulated parafibromin expression is a promising marker for pathogenesis, invasion, metastasis and prognosis of gastric carcinomas.

Parafibromin is a protein encoded by the hyperparathyroidism 2 oncosuppressor gene and its downregulated expression is involved in pathogenesis of parathyroid carcinomas. To clarify the roles of parafibromin expression in tumourigenesis and progression of gastric carcinomas, it was examined by immunohistochemistry (IHC) on tissue microarray containing gastric carcinomas (n = 508), adenomas (n = 45) and gastritis (n = 49) with a comparison of its expression with clinicopathological parametres of carcinomas. Gastric carcinoma cell lines (MKN28, AGS, MKN45, KATO-III and HGC-27) were studied for p

DREADDs-Gi mediated inhibition of myeloid cells leads to anti-inflammatory signalling, shrinkage of microglia and acute heart failure in Cx3cr1Cre mice.

Little is known about the functional consequences of real-time changes to innate immune-cell states. While chemogenetic tools have become increasingly used for probing immune-brain interactions, the consequences of manipulating myeloid cell activity states remain poorly understood. In this study, we found that acute activation of Gi-coupled DREADDs (hM4Di) in Cx3cr1⁺ myeloid cells severely suppresses cardiac function and induces a predominantly anti-inflammatory signalling state in mice. This surprising finding prompted further investigation. Using high-resolution ultrasound and electrocardiog
