The artemisinin-induced dormant stages of Plasmodium falciparum exhibit hallmarks of cellular quiescence/senescence and drug resilience.

Recrudescent infections with the human malaria parasite, Plasmodium falciparum, presented traditionally the major setback of artemisinin-based monotherapies. Although the introduction of artemisinin combination therapies (ACT) largely solved the problem, the ability of artemisinin to induce dormant parasites still poses an obstacle for current as well as future malaria chemotherapeutics. Here, we use a laboratory model for induction of dormant P. falciparum parasites and characterize their transcriptome, drug sensitivity profile, and cellular ultrastructure. We show that P. falciparum dormancy

Identification of stable housekeeping genes in mouse liver for studying carbon tetrachloride-induced injury and cellular senescence.

Acute liver injury (ALI) presents a challenging problem worldwide, prompting extensive research efforts. Cellular senescence has been found to be induced following ALI, and targeting cellular senescence has shown therapeutic potential. Therefore, understanding the expression of senescence-related genes in ALI can help to explore pathogenesis and treatment of this common disease. Carbon tetrachloride (CCl4) is commonly used to study ALI. Although polymerase chain reaction (PCR) is a convenient and economical molecular biology technique widely used in basic medicine, research on selecting suitab…

RAS-activated PI3K/AKT signaling sustains cellular senescence via P53/P21 axis in experimental models of psoriasis.

BACKGROUND: Psoriasis is a chronic immune-mediated skin disease in which upper epidermal keratinocytes exhibit a senescent-like phenotype. In psoriatic skin, a variety of inflammatory cytokines can activate intracellular pathways including phosphatidylinositol 3-kinase (PI3K)/AKT signaling and RAS effectors. AKT and RAS participate to cellular senescence, but currently their role in senescence responses occurring in psoriasis have not yet been investigated. OBJECTIVE: The role of AKT molecular axis and RAS activation was evaluated in the context of cellular senescence in psoriasis disease. M…

Characterization of cellular senescence in radiation ulcers and therapeutic effects of mesenchymal stem cell-derived conditioned medium.

BACKGROUND: Radiation ulcers are a common and severe injury after uncontrolled exposure to ionizing radiation. The most important feature of radiation ulcers is progressive ulceration, which results in the expansion of radiation injury to the nonirradiated area and refractory wounds. Current theories cannot explain the progression of radiation ulcers. Cellular senescence refers to as irreversible growth arrest that occurs after exposure to stress, which contributes to tissue dysfunction by inducing paracrine senescence, stem cell dysfunction and chronic inflammation. However, it is not yet cle…

Ferroptosis resistance cooperates with cellular senescence in the overt stage of nonalcoholic fatty liver disease/nonalcoholic steatohepatitis.

Cellular senescence and ferroptosis are the two main, fine-tuned processes in tissue damage restraint; however, they can be overactivated in pathologies such as nonalcoholic fatty liver disease/nonalcoholic steatohepatitis (NAFLD/NASH), becoming dangerous stimuli. Senescence is characterized by a decline in cell division and an abnormal release of reactive oxygen species (ROS), and ferroptosis is represented by iron deposition associated with an excessive accumulation of ROS. ROS and cellular stress pathways are also drivers of NAFLD/NASH development. The etiology of NAFLD/NASH lies in poor di…

Activation of DNA Damage Response and Cellular Senescence in Cardiac Fibroblasts Limit Cardiac Fibrosis After Myocardial Infarction.

Cardiac fibrosis plays an important role in cardiac remodeling after myocardial infarction (MI). The molecular mechanisms that promote cardiac fibrosis after MI are well studied; however, the mechanisms by which the progression of cardiac fibrosis becomes attenuated after MI remain poorly understood. Recent reports show the role of cellular senescence in limiting tissue fibrosis. In the present study, we tested whether cellular senescence of cardiac fibroblasts (CFs) plays a role in attenuating the progression of cardiac fibrosis after MI. We found that the number of γH2AX-positive CFs increas…

Downregulation of peroxiredoxin-3 by hydrophobic bile acid induces mitochondrial dysfunction and cellular senescence in human trophoblasts.

Intrahepatic cholestasis of pregnancy (ICP) is a pregnancy-specific disorder characterised by raised bile acids in foetal-maternal circulati … t cells. Moreover, silencing of PRDX3 in trophoblast cell line HTR8/SVneo induced growth arrest and cellular senescence via activation of p38-mitogen-activated protein kinase (MAPK) and induction of p21WAF1/CIP and p16INK4A. Additionally, enhanced cellular senescence, determined by senescence-associated beta-galactosidase staining, was obviously attenuated by p38-MAP …

Intermittent Supplementation With Fisetin Improves Physical Function and Decreases Cellular Senescence in Skeletal Muscle With Aging: A Comparison to Genetic Clearance of Senescent Cells and Synthetic Senolytic Approaches.

Excess cellular senescence contributes to age-related increases in frailty and reductions in skeletal muscle strength. In the present study, we determined the efficacy of oral intermittent treatment (1 week on-2 weeks off-1 week on) with the natural flavonoid senolytic fisetin to improve frailty and grip strength in old mice. Further, the effects of fisetin on physical function were evaluated in young mice. We performed bulk RNA sequencing of quadricep skeletal muscle to determine the cell senescence-related signaling pathways modulated by fisetin. We also assessed the relative effects of fise…

Organoids for modeling cellular senescence and age-related pathology: From experimental reconstruction to mechanistic and translational applications.

Cellular senescence, characterized by irreversible cell cycle arrest and the acquisition of a senescence-associated secretory phenotype (SASP), is a fundamental hallmark of aging and a key driver of numerous age-related diseases. However, conventional two-dimensional cell cultures and animal models fail to fully replicate the complex cellular architecture, multicellular interactions, and tissue-specific microenvironments in which senescence develops in vivo. Organoids, three-dimensional tissue models derived from pluripotent or adult stem cells, have emerged as physiologically relevant platfor…

Targeting DYRK1A with harmine abrogates the FOXO1-NF-κB axis to alleviate cellular senescence and renal fibrosis.

Proximal renal tubule epithelial cell (TEC) senescence and its associated senescence-associated secretory phenotype (SASP) drive renal fibrosis, yet the underlying molecular mechanism remains elusive. Dual-specificity tyrosine phosphorylation-regulated kinase 1 A (DYRK1A) accelerates cellular senescence, and its specific inhibitor Harmine is hypothesized to mitigate renal interstitial fibrosis (RIF) by targeting DYRK1A to alleviate TEC senescence. To verify this, we established a hypoxic HK-2 (human proximal TEC line) senescence model and a mouse unilateral ureteral obstruction (UUO) model, wi…

Crosstalk between exosomes and cellular senescence: A core vicious loop promoting α-synuclein pathology in Parkinson's disease.

Parkinson's disease is characterized by α-synuclein aggregation, dopaminergic neuron loss, and neuroinflammation. Emerging evidence reveals complex crosstalk between exosomes and cellular senescence in Parkinson's disease pathogenesis. Exosomes mediate α-synuclein transmission and neuroinflammation, while delivering protective factors such as miRNAs that inhibit cellular senescence. Senescent cells secrete pro-inflammatory factors via mitochondrial dysfunction and the senescence-associated secretory phenotype, linking to Parkinson's disease mitophagy defects. This forms a vicious "senescence-p…

Dapagliflozin attenuates cellular senescence and mitochondrial dysfunction in diabetic kidney disease: potential involvement of AMPK/Sirt3 signaling.

Cellular senescence contributes to the pathology of diabetic kidney disease (DKD). Besides, mitochondrial dysfunction and cellular senescence are closely associated pathological processes that may influence one another during kidney injury progression. Dapagliflozin has demonstrated its renal protective benefits independent of the glucose-lowering effects, yet whether it acts via modulating cellular senescence is poorly understood. The design of our study is to explore the potential renal protective mechanism of dapagliflozin, focusing on cellular senescence and mitochondrial function. The str…

Dehydrodiisoeugenol ameliorates DSS-induced ulcerative colitis by suppression of cellular senescence through inhibiting AKT and activating GSK3B.

Ulcerative colitis (UC), a common inflammatory bowel disease (IBD), is characterized by chronic mucosal inflammation. Our previous work established a link between cellular senescence and UC, suggesting that ameliorating cell senescence could be a therapeutic strategy for UC. In this study, we identified dehydrodiisoeugenol (DDIE) as an anti-senescence drug through screening in a DSS-induced colonic epithelial cell senescence model. In a DSS-induced acute mouse model of UC, DDIE effectively suppressed colonic senescence and significantly alleviated UC. Network pharmacology, colonic tissue trans…

Melatonin Alleviates Diabetic Cardiomyopathy by Inhibiting Cellular Senescence via Activation of the MT2/SIRT1/Nrf2 Signaling Pathway.

Diabetic cardiomyopathy (DCM) is one of the most severe cardiovascular complications of diabetes mellitus (DM), yet effective treatment strategies remain lacking. Cellular senescence is a principal risk factor for multiple cardiovascular diseases, contributing significantly to cardiac dysfunction and disease progression. Melatonin, an endogenous hormone primarily secreted by the pineal gland, exerts cardioprotective effects against DCM. However, the underlying molecular mechanism remains incompletely understood, particularly whether melatonin ameliorates DCM by suppressing cardiomyocyte senesc…

GATA4-Driven Transcription of HtrA1 Promotes Cellular Senescence in Ménière's Disease and Age-Related Audio-Vestibular Dysfunction.

Ménière's disease (MD), a chronic inflammatory disorder with age-related increased incidence, exhibits poorly understood pathogenesis and limited therapeutic options. Here, we demonstrate that cellular senescence, marked by mitochondrial damage, reactive oxygen species accumulation, and senescence-associated secretory phenotype (SASP), is prevalent in the vestibular tissue of MD patients and an endolymphatic hydrops mouse model. The transcription factor GATA4 is upregulated in MD and mice, and its genetic deletion in hair cells alleviates LPS-induced audio-vestibular dysfunction and cellular s…

27-hydroxycholesterol impairs placental development via p53/p21/Cdk6 pathway: Implications for nutrient transport and cellular senescence.

Aberrant placental development and function contribute to various pregnancy complications. 27-hydroxycholesterol (27-OHC), a recognized mediator linking hypercholesterolemia and metabolic diseases, has an undefined role in placental development. This study investigates the impact of 27-OHC on placental development and its underlying mechanisms, particularly in relation to cellular senescence. Pregnant mice were subcutaneously administered either 27-OHC (27-OHC group) or normal saline (control group) during gestation. Subsequently, placentas underwent spatial transcriptome (ST) sequencing. The …

Elucidating Cellular Senescence-related Genes in Benign Prostatic Hyperplasia Through Mendelian Randomization and Single-cell RNA Sequencing.

BACKGROUND: Benign prostatic hyperplasia (BPH) is a widely observed disorder in older men, with substantial evidence indicating that cellular senescence serves a pivotal function in its progression. This investigation seeks to pinpoint cellular senescence-related genes causally connected with BPH and to examine their expression and regulatory networks across distinct prostate cells. METHODS: Using exposure data from the eQTLGen database and outcome data from both FinnGen Consortium and UKB database, Mendelian randomization was utilized to determine cell senescence genes that are causally link…

Identification of cellular senescence-associated hub genes and molecular targeting mechanisms of thalidomide in lupus nephritis (LN).

Systemic lupus erythematosus (SLE), a chronic autoimmune disease, progresses to lupus nephritis (LN) in 50-60% of patients, driving end-stage renal disease (ESRD). Identifying LN-associated cellular senescence hub genes and drug targets is critical for elucidating pathogenesis and advancing targeted therapies. Integrated transcriptomic data from LN patients (GSE61635, GSE121239; n = 441) were analyzed to identify differentially expressed genes (DEGs) using the limma package (|log2FC| > 0.5 and FDR < 0.05). Cellular senescence-associated differentially expressed genes (CS-DEGs) were further fil…

ROS-degradable hydrogel delivering LOXL2-LNPs rescues disc degeneration by synchronously suppressing cellular senescence and oxidative damage.

BACKGROUND: Intervertebral disc degeneration (IDD) is a chronic degenerative disorder marked by nucleus pulposus cells (NPCs) senescence and … complements the intracellular anti-aging effect of LOXL2, blocking the cascade of "oxidative stress-cellular senescence-ECM degradation". In the rat IDD model, LNP-LOXL2@Gel not only reduced nucleus pulposus senescence, but also inhibited ECM catabolism by decreasing the expression of ROS in the microenvironment, thus partially restoring the physiological function of …
