The enantiomeric separation of triadimenol and triadimefon on a Chiralpak AD column using supercritical fluid chromatography, was studied in this work. The effect of different modifiers (methanol, ethanol and 2-propanol) was tested, with methanol and ethanol providing the best results for the enantiomeric separation of the two compounds. A high-pressure liquid chromatography method has been developed for the analysis in urinary calculi of six purines: uric acid, 2, 8-dihydroxyadenine, xanthine, hypoxanthine, allopurinol, and oxypurinol. A mixture of seven penicillins was separated on a C-8 column and detected using pulsed amperometric detection (PAD). Due to the polarity range of the penicillin mixture, a gradient program was necessary to produce a reasonable separation, causing some baseline shifting. Application of a flow program, where the solvent flow rate is varied within the separation, was also examined, and found to cause only a small shift in baseline response for PAD. Further examination of a variety of different flow programs was undertaken to characterize the baseline and analyte response under these conditions. N

A rapid screening method for detecting low levels of tetrachloroethylene (perchloroethylene, PCE) in olive oils has been developed using headspace capillary gas chromatography. Modification of this method allows quantitative results to be obtained down to 0.001 mg kg-1. Results obtained show that olive oil samples received in this laboratory over a two year period have not contained PCE residues in excess of the European Commission guidelines.

Hypoxanthine-guanine phosphoribosyltransferase (HPRT) catalyzes the conversion of hypoxanthine and guanine to IMP and GMP, respectively, in the presence of 5-phosphoribosyl-1-pyrophosphate. This study describes a method for the quantitative determination of residual tetrachloroethylene or perchloroethylene (PCE) in olive oil by headspace-gas chromatography. The analysis of the chlorinated product is performed by electron-capture gas chromatography, using a fused silica capillary column, CP Sil 8 CB. The absolute detection limit for PCE was 1 pg. Forty samples of olive oil and 5 other commodities have been analysed. Only one sample exceeded the proposed maximum level of 1 mg/kg.

Buffered solid dosage forms containing aspirin, magnesium hydroxide, and aluminum hydroxide are blended with acidic ethanol to extract the aspirin and salicylic acid rapidly. The resulting preparation is then immediately injected onto a 4.6 mm x 3 cm 5 micron reverse-phase column. Aspirin and free salicylic acid are determined simultaneously. The run time is less than 2 min. The total time from the initiation of sample extraction to completion of the separation is less than 5 min.
