Using confirmatory factor analysis to identify symptom clusters in individuals with acquired disabilities.

PURPOSE: Acquired disabilities, such as spinal cord injury, traumatic brain injury, limb loss (or other major limb injury/illness), or stroke, are typically accompanied by an array of physical and emotional symptoms and social challenges that interfere with recovery and affect quality of life (QOL). Despite advances in personalized medicine, interrelationships between symptoms remain unclear, and clinicians often treat individual symptoms in isolation. The objective of this study was to identify clusters of symptoms that occur across sudden-onset disability populations, with a long-term goal o

Phenotypic Clustering of Addictions Reveals Impulsivity Links to Internalizing Disorders: A Network Analysis.

BACKGROUND: Substance-related and behavioral addictions frequently co-occur and are increasingly conceptualized as part of an interconnected psychopathological framework. Addictive behaviors are linked to internalizing symptoms, including depression, anxiety, and stress, through shared vulnerability. Although impulsivity, anhedonia, and stress-related processes have been proposed as transdiagnostic factors, their relative roles as central or bridging mechanisms remain unclear. This study aimed to clarify the network structure of multiple addictions by jointly modeling substance-related and beh

A Dyadic Analysis of Posttraumatic Stress Clusters and Sexual Satisfaction with U.S. Veteran Couples.

Posttraumatic stress symptoms (PTSS) are associated with adverse relationship outcomes, including sexual satisfaction. This is particularly true for veteran samples; yet, these associations have rarely been studied in dyadic contexts. This study aimed to examine the individual and dyadic associations among DSM-5 PTSS clusters and sexual satisfaction in trauma-exposed, veteran couples. The study included 125 treatment-seeking veterans (i.e., men) and their civilian partners (i.e., women), where at least one partner reported a lifetime history of trauma exposure. An actor-partner interdependence

Spatial clusters, temporal behavior, and risk factors analysis of rabies in livestock in Ecuador.

Rabies, a globally distributed and highly lethal zoonotic neglected tropical disease, has a significant impact in South America. In Ecuador, animal rabies cases are primarily linked to livestock, and hematophagous bats play a crucial role in disease transmission. This study aims to identify temporal trends, spatial patterns, and risk factors for animal rabies in Ecuador between 2014 and 2019. Epidemiological survey reports from the official Animal Rabies Surveillance Program of the Phyto and Zoosanitary Regulation and Control Agency of Ecuador (AGROCALIDAD) were used. The Animal Rabies Surveil

Analysis of symptom clusters amongst adults with anorexia nervosa: Key severity indicators.

This study used cluster analysis to explore clinically relevant subgroups of adult patients with anorexia nervosa (AN). Patients were clustered based on their body mass index (BMI), eating disorder symptomatology, anxiety and depression symptoms and autistic characteristics. The difference between clusters in work and social functioning, duration of illness, bingeing and purging behaviour, previous hospitalisations and number of comorbidities was also investigated. Two meaningful clusters emerged: a higher symptoms cluster with more severe eating pathology, anxiety, depression, and more autist…

Time-varying proportional odds model for mega-analysis of clustered event times.

Mega-analysis, or the meta-analysis of individual data, enables pooling and comparing multiple studies to enhance estimation and power. A challenge in mega-analysis is estimating the distribution for clustered, potentially censored event times where the dependency structure can introduce bias if ignored. We propose a new proportional odds model with unknown, time-varying coefficients, and random effects. The model directly captures event dependencies, handles censoring using pseudo-values, and permits a simple estimation by transforming the model into an easily estimable additive logistic mixe

Metabolic Clustering Analysis as a Strategy for Compound Selection in the Drug Discovery Pipeline for Leishmaniasis.

A lack of viable hits, increasing resistance, and limited knowledge on mode of action is hindering drug discovery for many diseases. To optimize prioritization and accelerate the discovery process, a strategy to cluster compounds based on more than chemical structure is required. We show the power of metabolomics in comparing effects on metabolism of 28 different candidate treatments for Leishmaniasis (25 from the GSK Leishmania box, two analogues of Leishmania box series, and amphotericin B as a gold standard treatment), tested in the axenic amastigote form of Leishmania donovani. Genetic and biochemical analysis of the antibiotic biosynthetic gene clusters on the Streptomyces linear plasmid.

We extensively analyzed the giant linear plasmid pSLA2-L in Streptomyces rochei 7434AN4, a producer of two structurally unrelated polyketide antibiotics, lankacidin and lankamycin. It was found that amine oxidase LkcE oxidizes an acyclic amine to an imine, which is in turn converted to the 17-membered carbocyclic lankacidin. Heterologous expression and translational fusion experiments indicated the modular-iterative mixed polyketide biosynthesis of lankacidin. Concerning to lankamycin biosynthesis, starter unit biosynthesis and the post-PKS modification pathway were elucidated by feeding and g

Molecular analysis of Coxiella burnetii in Germany reveals evolution of unique clonal clusters.

The causative agent of Q fever, Coxiella burnetii, is a query agent occurring naturally all over the world. We studied 104 German Coxiella burnetii strains/DNA samples obtained between 1969 and 2011 using a 14 microsatellite marker Multiple-locus variable-number of tandem repeat (VNTR) analysis (MLVA) technique. We were able to divide our collection into 32 different genotypes clustered into four major groups (A-D). Two of these (A and C) formed predominant clonal complexes that covered 97% of all studied samples. Group C consisted exclusively of cattle-associated isolates/DNA specimens, while
