Collagens of normal and cirrhotic human liver.

The collagens of normal and end-stage alcoholic cirrhotic human liver were investigated. Mild conditions of pepsin digestion were employed to preserved the more pepsin sensitive type IV collagen molecules while increasingly more harsh conditions were used which solubilized the type I and type III hepatic collagens. The total hepatic collagen content was elevated from 1.8-2.1 in normal liver to 7.3-8.2 percent in the end-stage alcoholic liver. The graded pepsin digestion resulted in solubilization of 76-90 percent of the total hepatic collagen. The genetically distinct types of collagen were fr…

Isolation and characterization of collagen in Peyronie's disease.

Peyronie's disease is characterized histologically by excessive collagen deposition in the lesion. We examined the collagen types in Peyronie's disease plaque tissues compared to unaffected tissues from the same patient, other control tissues, and Dupuytren's contracture. Gel electrophoresis of pepsin-solubilized collagen demonstrated the presence of type I collagen and an increased content of type III collagen in plaque tissue. Increased type III collagen was detected in apparently normal tissue adjacent to the plaque and in Dupuytren's lesion, confirming previous findings. Although the cause…

Identification of collagen fibrils in scleroderma skin.

Skin from early and late stages of scleroderma has been shown to contain large amounts of thin (30-40 nm diameter) collagen fibrils that may be present in bundles or intermingled with large diameter fibrils (90-120 nm). The nature of these fibrils is unknown. Skin biopsies were obtained from involved areas of nine patients with progressive systemic sclerosis (PSS), one case of generalized morphea, one case of morphea, and six normal controls. Intact skin was analyzed by immunoelectron microscopy (IEM), while extracts were subjected to sodium dodecyl polyacrylamide gel electrophoresis (SDS-PAGE…

The collagen eye patch: a preliminary report.

This study is a preliminary report on the use of freeze-dried-collagen eye patches in the treatment of a selected group of inflammatory eye conditions. The early responses to treatment have been encouraging in cases of allergic conjunctivitis, blepharitis, and pterygium. In addition, the collagen patches have exhibited a significant beneficial effect in the resolution of ectropion.

UCLA conference. Biochemistry of collagen in diseases.

Collagen, the major extracellular matrix component, comprises a family of closely related, yet genetically distinct proteins that provide physiologic tensile properties to the tissues. The recent progress made in understanding the normal biochemistry and biology of collagens has permitted us to identify different levels at which errors might be introduced into the structure and metabolism of this protein. Several acquired and heritable diseases are now known to involve distinct abnormalities in collagen. This conference reviews some of the latest discoveries in the biochemistry of collagen and…

[How to explore the microangiopathy of collagen diseases].

Numerous methods for the exploration of the microangiopathy of collagen disease exist, but their perspective differs as a function of their use to assist early diagnosis, to establish a prognosis, to provide follow-up data or to attempt better understanding of the mechanism of these affections. Progressive systemic sclerosis is a very suitable collagen disease with which to illustrate these methods, since in this affection the microangiopathy is frequent, diffuse, of early onset and also formally implicated in the pathogenic rank. Biopsy specimens provide details of morphologic anomalies of th…

Collagen genes and proteins in osteogenesis imperfecta.

Type I collagen is a heteropolymer of alpha 1(I) and alpha 2(I) chains, each of which is a separate product of genes localised to chromosomes 17 and 7 respectively. Molecular defects of type I collagen produce a group of inherited disorders of connective tissue primarily affecting bones, which are easily broken and collagen depleted (osteogenesis imperfecta). Sillence classifies these diseases into four groups, two of which are autosomal dominant and relatively mild, the others being either genetic lethals or responsible for very severe progressive disease. Here we describe two specific molecu…

Collagenous colitis: the clinical and morphological features.

A woman aged 23 years is described who had watery diarrhoea and colicky abdominal pain with normal results on sigmoidoscopy and barium enema. Rectal biopsy showed a thick layer of collagen in the lamina propria below the free epithelial surface. This lesion extended throughout the length of the descending colon. The only change revealed by electron microscopy was an increase in the thickness of the sub-epithelial collagen layer which is normally present in the colon. The maturation of the fibroblasts was normal. The lesion in this patient may be considered as a small change from the normal sit…

[Pulmonary collagens and their pathology (author's transl)].

Collagen is a macro-molecule of polypeptide chains arranged in a triple helix. The biosynthesis and degradation of collagen require complex enzymes systems; considerable progress has been made in their understanding in recent years. Collagen is the most important connective tissue of the body and five types are present in the lower respiratory tract. Genetic disorders of collagen (e.g. the Ehlers-Danlos or Marfan's syndrome) may affect the lung. Furthermore certain pulmonary diseases such as fibrosis are associated with changes in the ratio of the different types of collagen. Collagen can no l…

Brain pathology in the collagen vascular diseases.

Neuropathologic examination of an autopsy series of 54 patients of various types of CVD revealed a very high frequency of pathologic changes both in brain parenchyma (in 81%) and vessels (in 78%). A broad but continuous spectrum of primary vascular alterations was observed, ranging from fibrinoid deposits in intact or necrotizing vessel walls to fibrohyalinosis and endothelial proliferations. In acute SLE showing LE cells within brain tissues, immune complex deposits were observed for the first time in brain vessels, in addition to similar deposits in the plexus chorioideus and in hematoxylin

Primary biliary cirrhosis as a collagen disease.

Primary biliary cirrhosis is a disease of the small bile ducts with altered immunologic responsiveness often associated with various collagen diseases. All appear to be mediated by immune complex deposition. They probably are all different clinical manifestations of the same disease with different expressions determined by genetic and acquired factors. Therapy should be directed to the most immediate life-threatening problem.

[Unusual forms of collagen fibrils in dermatoses].

Electron microscopic examinations of the skin from foci of lesions in scleroderma, psoriasis, parapsoriasis, and lupus erythematosus revealed two types of unusual collagen structures consisting of loosely packed fibrils 5--8 nm in diameter with a cross-striation period of 150--170 and 80--90 nm. The former were localized in the epi- or perineurium of pulpless nervous fibers of the derma, the latter among collagen fibers. These structures appear to occur rather widely in collagen metabolism disorders (tumours of neurogenic origin, some skin tumours, collagen diseases).

Cellular sensitivity to collagen in rheumatoid arthritis.

Using an assay to measure antigen-induction of a lymphocyte mediator, LIF, we detected cellular sensitivities to native human types II and III collagens in three-quarters of a group of 50 patients with rheumatoid arthritis. There was no cellular response to type I collagen. Cellular reactivities to collagen were absent in a group of 41 patients who had other kinds of arthritis, such as osteoarthritis, crystalline-induced synovitis, or arthropathies associated with a high prevalence of the HLA-B27 antigen. Lymphocytes, responding to an unknown persistent antigenic stimulus, are thought to play …

Chemistry of the crosslinking of collagen during tanning.

The materials commonly used for crosslinking collagen as part of the process of converting animal hides into leather fall into three main groups: mineral tannages, aldehyde tannages, and "vegetable" tannages. The most important mineral crosslinking agents are hydrated basic chromium III sulfate complexes. These compounds form extended polynuclear coordination complexes containing hydroxol, oxo, and sulfato bridges into which ionized carboxyl groups on collagen enter readily as coordinating ligands accomplishing crosslinking. On pH adjustment and partial drying, highly stable complexes are form…

Collagen biosynthesis in liver disease of the alcoholic.

Percutaneous liver biopsies obtained from patients with a history of chronic alcoholism and normal liver, fatty liver, alcoholic hepatitis, or active cirrhosis were incubated with tritiated proline to determine the pattern of collagen biosynthesis in these conditions. Incorporation of labeled proline and hydroxyproline into salt-soluble and insoluble fractions of collagen was evaluated by radiochemical analysis and tissue localization documented by autoradiography. Biopsy specimens of alcoholic hepatitis and cirrhosis exhibit a significant increase in the amount of radioactive proline and hydr…
