Gamma-glutamyltransferase and immunoglobulin G in intestinal maturation: Pilot study in preterm and term infants and the influence of breast milk.

Gamma-glutamyltransferase (γGT), a key enzyme in glutathione metabolism, is abundant in colostrum and may indicate neonatal gastrointestinal maturation, although normative serum values and associations with breast milk intake remain unclear, particularly in preterm infants. In this prospective study, serum γGT was measured in 84 neonates, including 36 born before 32 weeks' gestation and 48 born at or after 32 weeks, across the first 4 postnatal weeks. Breast milk γGT was analyzed in samples from 107 mothers, and correlations with colostrum intake were assessed in 66 neonates. Serum γGT remaine…

Triacylglycerols containing branched palmitic acid ester of hydroxystearic acid (PAHSA) are present in the breast milk and hydrolyzed by carboxyl ester lipase.

Breast milk is a complex mixture containing underexplored bioactive lipids. We performed an observational case-control study to compare the … eferred substrates among tested triacylglycerol estolide isomers. This explained exceptionally high colostrum levels of 5-PAHSA in the VB group. CS and preterm birth negatively affected colostrum lipidome, including 5-PAHSA levels, but the lipidomic profiles normalized in mature milk. Mothers delivering term babies vaginally produce colostrum rich in 5-PAHS …

Dynamics and drivers of the human breast milk microbiome across lactation stages.

INTRODUCTION: Human breast milk (HBM) not only provides all the nutrients and microbiota for the initial development of infants but is also … plex ecosystem, and we found the network central nodes that change during the transition from human colostrum to human mature milk and various stage specific microbial markers. In addition, we also found that the mode of delivery and intrapartum antibiotic prophylaxis during delivery affected the structure and diversity of the microbial community of colostr …

Production of Recombinant S1 Protein of Porcine Epidemic Diarrhea Virus in Recombinant CHO Cells for Application in Indirect ELISA.

The N-terminal S1 subunit of the spike protein (S1 protein) of porcine epidemic diarrhea virus (PEDV) is recognized as a potential diagnostic antigen for detecting anti-PEDV immunoglobulin A (IgA) levels in colostrum, a key indicator for assessing passive immunity against PEDV infection. Given the advantages of producing Fc-fusion proteins in mammalian cells, it is important to investigate how the addition of the Fc region affects the diagnostic performance of the PEDV-S1 protein. In this study, we successfully produced full-length PEDV-S1 protein fused with an Fc region (PEDV-S1-Fc protein) v…

Exploratory cohort study to determine if dry cow vaccination with a Salmonella Newport bacterin can protect dairy calves against oral Salmonella challenge.

BACKGROUND: Salmonellosis is a major cause of morbidity and mortality in neonatal calves, often occurring before preventative vaccines can be administered. HYPOTHESIS/OBJECTIVE: To evaluate the protective effect on calves of colostrum from cows vaccinated with a commercially available Salmonella Newport bacterin against a Salmonella Typhimurium challenge. ANIMALS: Twenty Holstein bull calves from a university dairy farm. METHODS: Nonrandomized placebo-controlled trial in which colostrum was harvested from 30 cows that received 2 doses of either Salmonella bacterin or saline before calving. …

Hyper-Immune Bovine Milk as an Immunological and Nutritional Supplement for COVID-19.

UNLABELLED: Many different strategies have been used to fight against the Coronavirus disease (COVID-19) pandemic as a therapeutics or prophylaxis approaches. However, not enough attention has been paid to general and specific immune factors and nutritional components found in hyper-immunized dairy products. Hyper-immune bovine colostrum (HBC) has been used against many different respiratory and gastrointestinal tracts infections during past decades. An isolated dairy farm was established, and nine mixed Holstein X Simmental dairy cattle in their 6-7 months of gestation period were chosen for …

The influence of colostral leukocytes on the course of an experimental Escherichia coli infection and serum antibodies in neonatal calves.

Three hours post natum, 20 calves were orally infected with 10(9) colony forming units of an enteropathogenic E. coli. Immediately after infection and on 2 days subsequent, ten of the calves (COL-) received cell-deprived pooled colostrum and the other ten (COL+) pooled colostrum supplemented with colostral cells. The COL+ calves excreted significantly less bacteria of the infectious strain with their faeces in the first week after infection and reached the lower limit of detectability earlier than the COL- calves. Colostral leukocytes obtained from cows which developed clinical mastitis (n = 2…

Newborn rats in the murine typhus enzootic infection cycle: studies on transplacental infection and passively acquired maternal antirickettsial antibodies.

This study focused attention on the newborn rat as a possible significant participant in the highly successful enzootic cycle of murine typhus. We examined the influence of maternal Rickettsia typhi (R. mooseri) infection in rats on the offspring with respect to the possible vertical transmission of R. typhi and the passive transfer of maternal antirickettsial antibodies. Transmission of R. typhi by rickettsemic pregnant rats did not occur either transplacentally during gestation to their fetuses or postnatally through colostrum and milk to their newborn. The rickettsial burden of the placenta…

Joint-ill and other bacterial infections on thoroughbred studs.

From a perinatal survey carried out on Thoroughbred studs, information was obtained on the incidence, mortality and long-term effects of joint-ill and other bacterial infections. Many of these infections occurred in foals with a history of other perinatal abnormalities, and in foals in which receipt of colostrum was delayed. There was no direct relationship between the incidence of infection and maternal age, but prenatal lactation and loss of colostrum were more frequent in older mares. There was a significant reduction in the incidence of infections in foals given neomycin and framomycin dur…

Prions in milk from ewes incubating natural scrapie.

Since prion infectivity had never been reported in milk, dairy products originating from transmissible spongiform encephalopathy (TSE)-affected ruminant flocks currently enter unrestricted into the animal and human food chain. However, a recently published study brought the first evidence of the presence of prions in mammary secretions from scrapie-affected ewes. Here we report the detection of consistent levels of infectivity in colostrum and milk from sheep incubating natural scrapie, several months prior to clinical onset. Additionally, abnormal PrP was detected, by immunohistochemistry and…

Delayed-type hypersensitivity responses induced by bovine colostral components.

Transfer factor-type substances obtained from leukocytic cells and fluid portions of bovine colostrum caused effective passive transfer of delayed-type hypersensitivity responses across species barriers. Passive transfer of Brucella abortus sensitivity was obtained with equal regularity when using components derived from peripheral blood and colostrum of dams sensitized at 3 and 9 months of age. Colostral feedings to calves caused the passive transfer of delayed-type hypersensitivity as early as 2 days after parturition. The findings indicated that colostral components were important in the pr…

Effects of age, nursing, and oral IGF1 supplementation on neonatal porcine cervical development.

Nursing supports neonatal porcine uterine and testicular development, however, lactocrine effects on cervical development are undefined. Studies were conducted to determine the effects of i) age and the imposition of the lactocrine-null state from birth (postnatal day 0 (PND0)) by milk replacer feeding on cervical histology; ii) imposition of the lactocrine-null state for 2 days from birth on cervical cell proliferation, as reflected by proliferating cell nuclear antigen immunostaining; and iii) a single feeding of colostrum or milk replacer, administered at birth, with or without oral IGF1, o…

Characterization of potential sources of secretory immunoglobulin A reaching the rumen and its effects on microbial growth and fermentation.

Secretory immunoglobulin A (SIgA) is a major mucosal antibody known to modulate intestinal microbial ecology in nonruminant species; however, its role in the rumen remains largely unexplored. The objectives of this study were to identify potential sources of SIgA reaching the rumen and to evaluate whether colostrum-derived SIgA can influence rumen bacterial growth and fermentation. In study 1, SIgA concentrations were quantified in colostrum and milk from 15 multiparous Holstein cows during the first 42 d of lactation and in saliva from their respective calves during the first 42 d of life. Mi…

Effect of dry period immunization of Salmonella Dublin latent carriers with a commercial live culture vaccine on intrauterine transmission based on the presence of precolostral antibodies in offspring.

Salmonella Dublin latent carrier cows represent a high risk for infection of newborn calves via intrauterine transmission and shedding of bacteria in feces and colostrum at calving. Vaccination of these latent carrier dams during late gestation boosts immunity against Salmonella Dublin. This could reduce the activation of the dormant bacteria during the periparturient immune dysfunction period, thereby reducing the risk of early-life infection in the offspring. Thus, the objective of this study was to evaluate the extent to which vaccinating Salmonella Dublin latent carrier cows at dry-off wit…

Developments of reproductive management and biotechnology in the pig.

This review aims to describe changes in production environment, management tools and technology to alleviate problems seen with the present hyperprolific sow model. Successful parturition in the pig includes the possibility to express adequate maternal behaviour, rapid expulsion of piglets, complete expulsion of placenta, elimination of uterine contamination and debris, neonatal activity and colostrum intake. We focus on management of large litters, including maternal behaviour, ease of parturition, colostrum production, piglet quality parameters and intermittent suckling. There are also some …

The kinetics of maternal and self-developed Streptococcus suis-specific antibodies.

BACKGROUND: Streptococcus suis (S. suis) infections are responsible for a large disease burden in piglets during the nursery phase, compromising animal welfare and increasing antibiotic use. The immune gap caused by decreased maternal-derived antibodies (MDA) and insufficient levels or functionality of acquired antibodies in weaned pigs could play a role in the increased susceptibility to S. suis infections. To better understand this, two studies were performed. Study I evaluated the associations between sow antibodies in colostrum and serum, birth parameters (e.g., birth weight, colostrum int…

Exploring pathogen-specific somatic cell patterns in dairy ewes during early lactation.

The postpartum period is a critical period for mastitis susceptibility in dairy ewes, yet the species-specific effects of mastitis pathogens on milk SCC and differential inflammatory cell counts (DICC) remain largely unknown. Therefore, this study investigates pathogen-specific impacts on microscopic SCC and DICC in milk and colostrum from Lacaune dairy ewes during early lactation. A total of 586 colostrum and milk samples were collected from 50 ewes (100 udder halves) at 6 time points: on the day of parturition and at 1, 3, 7, 15, and 30 d postpartum. Milk and colostrum samples were examined …
