Reaction of human hemoglobin HbA0 and two cross-linked derivatives with hydrogen peroxide: differential behavior of the ferryl intermediate.

Functional concerns regarding hemoglobin-based red cell substitutes have generally centered on two parameters: (a) oxygen binding and delivery properties and (b) stabilization of the hemoglobin tetramer to prevent dimerization. Strategic chemical cross-linking and site-directed mutagenesis have produced proteins that have both physiological oxygen binding characteristics and a markedly prolonged retention time in the circulation. The presence of a large amount of redoxactive iron outside the red blood cell, however, raises some concerns about the potential for toxic side effects, many involvin

A sensitive radioimmunoassay for fludrocortisone in human plasma.

Fludrocortisone has been a mainstay of therapy for orthostatic hypotension for many years. Clinical experience suggests that there exists a substantial interindividual variation in responsiveness to the drug. To assess this, we have developed an assay that permits measurement of the low concentrations of fludrocortisone found in human plasma. Fludrocortisone was detected by radioimmunoassay. A polyclonal rabbit antibody, raised against dexamethasone which cross-reacts strongly with fludrocortisone, was reacted with either standard or unknown samples in the presence of [125I]fludrocortisone-3-T

Cutaneous vasculitis in the newborn of a mother with cutaneous polyarteritis nodosa.

Vasculitis in an infant of a woman who had a long history of cutaneous polyarteritis nodosa is reported. During the neonatal period the child developed cutaneous vasculitis manifested by livedo reticularis, cutaneous nodules, and acral necrosis. The infant's vasculitis remitted by age 7 months. This is the third such report and strongly suggests the presence of a circulating factor that is capable of crossing the placenta and inducing cutaneous polyarteritis nodosa.

Chronic presence of blood circulating anti-NMDAR1 autoantibodies impairs cognitive function in mice.

High titers of anti-NMDAR1 autoantibodies in brain cause anti-NMDAR1 encephalitis that displays psychiatric symptoms of schizophrenia and/or other psychiatric disorders in addition to neurological symptoms. Low titers of anti-NMDAR1 autoantibodies are reported in the blood of a subset of the general human population and psychiatric patients. Since ~0.1-0.2% of blood circulating antibodies cross the blood-brain barriers and antibodies can persist for months and years in human blood, it is important to investigate whether chronic presence of these blood circulating anti-NMDAR1 autoantibodies may

Absence of Metformin in Fetal Circulation Following Maternal Administration in Late Gestation Pregnant Sheep.

In human pregnancy, metformin administered to the mother crosses the placenta resulting in metformin exposure to the fetus. However, the effects of metformin exposure on the fetus are poorly understood and difficult to study in humans. Pregnant sheep are a powerful large animal model for studying fetal physiology. The objective of this study was to determine if maternally administered metformin at human dose-equivalent concentrations crosses the ovine placenta and equilibrates in the fetal circulation. To test this, metformin was administered to the pregnant ewe via continuous intravenous infu

Real-time imaging of trapping and urease-dependent transmigration of Cryptococcus neoformans in mouse brain.

Infectious meningitis and encephalitis is caused by invasion of circulating pathogens into the brain. It is unknown how the circulating pathogens dynamically interact with brain endothelium under shear stress, leading to invasion into the brain. Here, using intravital microscopy, we have shown that Cryptococcus neoformans, a yeast pathogen that causes meningoencephalitis, stops suddenly in mouse brain capillaries of a similar or smaller diameter than the organism, in the same manner and with the same kinetics as polystyrene microspheres, without rolling and tethering to the endothelial surface

The Interaction Between Mental Distress and Opioid Maintenance Treatment Impacts Levels of Circulating Cytokines.

Opioid use modulates parts of the immune system, including cytokines, but with disparate results. Furthermore, several studies have demonstrated a positive correlation between circulating proinflammatory cytokines and mental distress of various kinds. The aim of this study was to investigate the relationship between self-reported mental health symptoms and peripheral circulating cytokines in opioid maintenance treatment patients to see whether the previous disparate results could at least in part be explained by an interaction with mental distress. In a cross-sectional study, we investigated l

Neuropilin-1 Associated Molecules in the Blood Distinguish Poor Prognosis Breast Cancer: A Cross-Sectional Study.

Circulating plasma and peripheral blood mononuclear (PBMCs) cells provide an informative snapshot of the systemic physiological state. Moreover, they provide a non-invasively accessible compartment to identify biomarkers for personalized medicine in advanced breast cancer. The role of Neuropilin-1 (NRP-1) and its interacting molecules in breast tumor tissue was correlated with cancer progression; however, the clinical impact of their systemic levels was not extensively evaluated. In this cross-sectional study, we found that circulating and tumor tissue expression of NRP-1 and circulating place

Circulating fibrocytes as biomarker of prognosis in Hermansky-Pudlak syndrome.

RATIONALE: The rate of progression of most interstitial lung diseases (ILD) is unpredictable. Fibrocytes are circulating bone marrow-derived cells that have been implicated in the pathogenesis of lung fibrosis. Hermansky-Pudlak syndrome (HPS), a genetic cause of ILD in early adulthood, allows for study of biomarkers of ILD in a homogeneous population at near-certain risk of developing fibrotic lung disease. OBJECTIVES: To test the hypothesis that, in subjects with HPS, the number or phenotype of circulating fibrocytes predicts progression and outcome of ILD. METHODS: We measured circulating

Childhood bullous pemphigoid: report of a case with characterization of the targeted antigens.

The clinical and immunopathologic features of children with acquired subepidermal blistering disorders show considerable overlap, and their classification frequently requires characterization of the targeted antigens. A 8-month-old boy developed a generalized subepidermal blistering disorder with striking palmoplantar involvement. The patient's serum contained antibodies reacting against the epidermal side of 1 M sodium chloride separated normal human skin. Immunoblotting analysis demonstrated circulating IgG autoantibodies that reacted against a eukaryotic recombinant form of human bullous pe

Localized adaptive evolution in VP1 drives antigenic divergence of feline calicivirus despite high sequence conservation.

Feline calicivirus (FCV) evolves rapidly under immune pressure, yet the genetic basis of antigenic escape remains poorly understood, particularly when sequence divergence is limited. We isolated 13 FCV strains from cats in Jilin Province, China, between 2018 and 2024, and characterized their genetic and antigenic properties. Phylogenetic analysis of the major capsid protein VP1 revealed the co-circulation of multiple lineages, including isolates clustering within clades associated with virulent systemic disease (VSD) and grouping with strains previously identified in dogs. Cross-neutralization

Molecular Prevalence and Genetic Characterization of Bovine Tick-Borne Protozoa in Thai and Imported Beef Cattle in Thai-Myanmar Border and Infesting Ticks from Kanchanaburi Province, Thailand.

Tick-borne protozoa (TBP), including Babesia spp. and Theileria spp., cause substantial health and productivity losses in cattle. In Thailand, most epidemiological studies have focused on dairy herds, while beef cattle remain underinvestigated. This study assessed TBP infections in beef cattle and their ticks at the Thai-Myanmar border. Blood samples were collected from 158 beef cattle, including local animals from Thong Pha Phum and Sangkhlaburi districts and cross-border cattle imported from Myanmar. Engorged ticks, predominantly Rhipicephalus microplus, were removed and identified morpholog

Adenovirus-mediated gene therapy for mucopolysaccharidosis VII: involvement of cross-correction in wide-spread distribution of the gene products and long-term effects of CTLA-4Ig coexpression.

Recombinant adenoviruses expressing human beta-glucuronidase (AxCAhGUS) and CTLA-4Ig (AxCACTLA-4Ig) were generated and therapeutic efficacy was investigated using a murine model of mucopolysaccharidosis type VII (MPSVII). Seven days after the intravenous administration of AxCAhGUS, high levels of beta-glucuronidase (GUSB) activity were observed in the liver, spleen, heart, lung, kidney, and serum, while viral DNA was predominantly detected in the liver. To investigate the contribution of in vivo cross-correction of GUSB between the liver and other organs, we injected the serum obtained from th

[Value and limits of serological methods: electrosyneresis and E.L.I.S.A. for the study of the prevalence of histoplasmosis in Mali].

The authors have looked for circulating antibodies on 168 subjects for the study of the prevalence of histoplasmosis in Mali. Two serologic systems were employed: with counter-immunoelectrophoresis precipitating bands were detected in 42% of sera, while 57% were positive with Enzyme-linked immunosorbent assay. E.L.I.S.A. appear less specific because many cross-reactions were observed with other diseases.

[Neonatal Fc receptor : role and implications in maternal fetal medicine].

During pregnancy, the mother's IgG immunoglobulins cross the -placenta via the neonatal Fc receptor (FcRn), enabling the fetus to acquire passive immunity. In the presence of maternal allo- or auto-antibodies, placental transfer of these pathogenic antibodies mediated by FcRn can cause diseases in the fetus and/or the newborn. FcRn blockade therefore appears to be a therapeutic strategy in these high-risk pregnancies, firstly by reducing IgG recycling, -thereby reducing its concentration in the maternal circulation, and secondly by blocking placental transfer. The promising results of a recent

[Acute brain stem syndromes].

The brain stem consists of the mesencephalon, pons, and medulla oblongata and contains the nuclei of the cranial nerves III to XII, their complex connectivity, the ascending and descending fibre tracts, and the cerebellar connections. The corticobulbar and corticospinal tracts and the medial lemniscus cross its midline. Vital functions such as wakefulness, respiration, swallowing, and circulation are controlled there. Here we discuss acute brain stem syndromes of vascular, inflammatory, and toxic-metabolic origin. They present frequently with alternating symptoms, i.e. ipsilateral cranial nerv

[Conventional surgery continues to be the best option for surgical treatment of aortic valve disease. Arguments in favor].

It is premature to try to compare the results of aortic valve surgery obtained by the so-called minimally-invasive techniques, with those obtained with the use of the conventional median sternotomy approach. The experience accumulated with this type of limited access surgery is small and remains anecdotal. We reviewed one hundred consecutive aortic valve patients operated at the Mayo Clinic. There was one hospital death. There were no instances of post-operative bleeding or mediastinitis. Average duration of extracorporeal circulation was 72 minutes; average duration of aortic cross-clamp was
