Mechanism of action of the platelet function inhibitor from Vipera russelli siamensis snake venom.

Human platelet aggregation induced by ADP, adrenaline, collagen or thrombin was inhibited by the venom inhibitor. Heating reduced both its phospholipase A2 enzymatic and anti-aggregatory activities, although not in parallel. The inhibitor caused significant dose-related inhibitory effects on the clot retraction of rabbit platelet-rich plasma caused by thrombin, while platelet malondialdehyde formation stimulated by thrombin was not affected. Furthermore, the venom inhibitor increased basal cyclic AMP levels in platelets, while cyclic GMP content was slightly lowered, but not in a dose-dependen…

Changes in cGMP levels on meiosis reinitiation of starfish oocytes.

An intracellular level of cGMP (but not cAMP) transiently decreased the reinitiation of oocyte maturation in the starfish, Asterias amurensis. Exogenously applied cGMP inhibited hormone-induced maturation. Methylxanthines inhibited oocyte maturation by suppressing the decrease in cGMP levels. These results suggest that a decrease in cGMP levels is a prerequisite for meiosis reinitiation of starfish oocytes.

Synergistic Effects of Pyrrosia lingua Caffeoylquinic Acid Compounds with Levofloxacin Against Uropathogenic Escherichia coli: Insights from Molecular Dynamics Simulations, Antibiofilm, and Antimicrobial Assessments.

Urinary tract infections (UTIs), primarily caused by uropathogenic Escherichia coli (UPEC), have high morbidity and recurrence rates. Resistance to levofloxacin hydrochloride (LEV), a commonly used treatment for UTIs, is increasingly problematic, exacerbated by biofilm formation mediated by interactions between cyclic di-GMP (c-di-GMP or CDG) and YcgR. In this study, we identified three caffeoylquinic acid compounds from Pyrrosia lingua-chlorogenic acid (CGA), sibiricose A5 (Si-A5), and 3-O-caffeoylquinic acid methyl ester (CAM)-that target YcgR through molecular docking. Biological assays rev

Direct evaluation of Pseudomonas aeruginosa biofilm mediators in a chronic infection model.

Biofilms contribute to Pseudomonas aeruginosa persistence in a variety of diseases, including cystic fibrosis, burn wounds, and chronic suppurative otitis media. However, few studies have directly addressed P. aeruginosa biofilms in vivo. We used a chinchilla model of otitis media, which has previously been used to study persistent Streptococcus pneumoniae and Haemophilus influenzae infections, to show that structures formed in vivo are biofilms of bacterial and host origin within a matrix that includes Psl, a P. aeruginosa biofilm polysaccharide. We evaluated three biofilm and/or virulence me

Evidence for amylase release by cGMP via cAMP-dependent protein kinase in rat parotid acinar cells.

Amylase release from the rat parotid gland is primarily mediated by a cAMP-dependent protein kinase (PKA). We previously reported that cGMP/cGMP-dependent protein kinase (PKG) signaling evokes amylase release. In the present study, we investigated whether cGMP-mediated amylase release might be due to cGMP/PKA signaling, as well as cGMP/PKG pathway. Activation of PKA by cGMP was required 100-1000-fold greater concentration than activation by cAMP in a parotid cytosol fraction. Synergistic activation of PKA by the combination of physiological cAMP and low concentration of cGMP was observed. Amyl

TDP-43 Puts the STING in ALS.

In a recent study, Yu et al. demonstrated that TAR DNA-binding protein of 43 kDa (TDP-43) causes inflammation in amyotrophic lateral sclerosis (ALS) by triggering mitochondrial (mt)DNA release into the cytoplasm, which subsequently activates the cytoplasmic DNA-sensing cyclic GMP-AMP synthase (cGAS)/stimulator of interferon genes (STING) pathway. These results suggest that inhibition of cGAS/STING could help mitigate inflammation-related neuropathology in ALS.

VpsR Directly Activates Transcription of Multiple Biofilm Genes in Vibrio cholerae.

Vibrio cholerae biofilm biogenesis, which is important for survival, dissemination, and persistence, requires multiple genes in the Vibrio polysaccharides (vps) operons I and II as well as the cluster of ribomatrix (rbm) genes. Transcriptional control of these genes is a complex process that requires several activators/repressors and the ubiquitous signaling molecule, cyclic di-GMP (c-di-GMP). Previously, we demonstrated that VpsR directly activates RNA polymerase containing σ70 (σ70-RNAP) at the vpsL promoter (P vpsL ), which precedes the vps-II operon, in a c-di-GMP-dependent manner by stimu

Immunohistochemical description of cyclic nucleotide phosphodiesterase (PDE) isoenzymes in the human labia minora.

INTRODUCTION: Up until now, only minimal research has been carried out on those female genital organs known to contribute to the normal cycle of sexual arousal and orgasm. Some findings indicated that there might be a significance of cyclic nucleotide-mediated pathways in the control of the normal function of female genital tissues. AIM: To elucidate, by means of immunohistochemistry, the distribution of the phosphodiesterase (PDE) isoenzymes 1, 3, 4, 5, 10, and 11 in the human labia minora. MAIN OUTCOME MEASURES: The amount of immunohistochemical staining specific for cyclic adenosine monop

Talazoparib engages innate immune activation via PARP trapping-dependent cGAS/STING activation in Ewing Sarcoma.

Ewing sarcoma (EwS) shows a limited clinical response to poly (ADP-ribose) polymerase (PARP) inhibitors (PARPi), despite promising preclinical data. In this study, we compared five PARPi with different PARP-trapping capacities in PDX-derived cell lines and mouse models. Talazoparib, the strongest PARP-trapping agent, showed markedly greater efficacy than olaparib or veliparib. It triggered extensive DNA damage, micronuclei formation, and activation of the cyclic GMP-AMP synthase (cGAS)/stimulator of interferon genes (STING) pathway, leading to robust type I interferon and pro-inflammatory cyto…

[Research progress on the correlation and treatment of the cGAS-STING pathway in systemic lupus erythematosus].

Systemic lupus erythematosus (SLE) is a chronic autoimmune disease, characterized by inflammation and immune-mediated damage to multiple organ systems. Emerging evidence demonstrates that the cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) signaling pathway plays a pivotal role in the pathogenesis of SLE. The cGAS-STING pathway is a pattern recognition pathway of the innate immune system. After cGAS senses double-stranded DNA or endogenous DNA in the cytoplasm, it generates 2'3'-cyclic GMP-AMP to mediate intracellular signal transduction. This process activates STING, whi…

The cGAS-STING pathway in pulmonary infectious and sterile inflammation: differences, connections, and therapeutic implications.

The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) signaling pathway plays a pivotal role in mediating both infectious and sterile pulmonary inflammation, exhibiting distinct dynamic characteristics under different contexts. The activation kinetics of the cGAS-STING pathway are highly dependent on disease context. In many acute infections, the cGAS-STING pathway is often activated rapidly and induces a strong type I interferon (IFN-I) response, which helps control pathogens. In contrast, chronic sterile injury is usually associated with endogenous damage-associated molecul…

Pathological cardiac hypertrophy is a key risk factor for heart failure. It is associated with increased interstitial fibrosis, cell death a … or the regression. The action of VEGF is mediated by VEGF receptor-1, whose activation is linked to cyclic GMP-dependent protein kinase-1 (PKG-1) signaling pathways, and inhibition of cyclic GMP degradation leads to regression of pathological cardiac hypertrophy. Most of these pathways are regulated by hypoxia-inducible factor. Potential therapeutic targets …

Insulin-stimulated NAD(P)H oxidase activity increases migration of cultured vascular smooth muscle cells.

BACKGROUND: We reported that insulin stimulates NAD(P)H oxidase activity but not migration of cultured rat vascular smooth muscle cells (VSMCs). Because angiotensin II (Ang II) increases NAD(P)H oxidase activity in these cells, we wished to determine whether insulin stimulates migration of Ang II-treated VSMCs by synergistically stimulating enzyme activity. METHODS: Cultured rat VSMC superoxide anion (O2-) production, cyclic GMP production, and migration were measured by lucigenin luminescence, immunoassay, and wound closure rate, respectively. Nitric oxide (NO) scavenging was measured by inh…

Activation of the cystic fibrosis transmembrane conductance regulator by cGMP in the human colonic cancer cell line, Caco-2.

Intestinal chloride (Cl-) secretion can be induced by the heat-stable enterotoxin (STa) from Escherichia coli via generation of cGMP. We investigated the regulatory pathway responsible for cGMP-mediated Cl- secretion in the human colonic carcinoma cell line Caco-2 using whole-cell voltage clamp techniques. Cyclic GMP or cAMP induced a 5-fold increase in Cl- conductance (gCl) in the presence of intracellular ATP and 3-isobutyl-1-methylxanthine. Current activation by cGMP persisted in the presence of the type I cGMP-dependent protein kinase (PKG) inhibitor, KT5823, but was inhibited by the speci…

N-terminal truncation of VC0395_0300 protein from Vibrio cholerae does not lead to loss of diguanylate cyclase activity.

The bacterial secondary messenger bis-(3',5')-cyclic-dimeric-guanosine monophosphate (c-di-GMP) has been implicated in the pathogenesis of Vibrio cholerae, due to its significant role in regulating the virulence, biofilm formation and motility of the host organism. The VC0395_0300 protein from V. cholerae, possessing a GGEEF sequence has been established as a diguanylate cyclase (DGC) capable of catalyzing the conversion of two GTP molecules to form cyclic-di-GMP. This in turn, plays a crucial role in allowing the organism to adopt a dual lifestyle, thriving both in human and aquatic systems.
