Sustained Release of Carvacrol Aldehyde from Hydroxyapatite for Osteosarcoma Inhibition.

The clinical management of bone cancer and infections remains a critical challenge due to limitations in localized drug delivery and rising antibiotic resistance. In our previous studies, we demonstrated carvacrol's (CA) osteogenic and antibacterial potential when loaded onto 3D-printed TCP scaffolds. However, the rapid release and limited retention of CA restricted long-term efficacy. Herein, we develop a hydroxyapatite (HA) substrate to locally deliver CA and its aldehyde derivative (carvacrol aldehyde) (CA-CHO) to enhance its biological properties. CA-CHO is synthesized via SnCl2-mediated o

Herbal terpenoids activate autophagy and mitophagy through modulation of bioenergetics and protect from metabolic stress, sarcopenia and epigenetic aging.

Small molecular food components contribute to the health benefits of diets rich in fruits, vegetables, herbs and spices. The cellular mechanisms by which noncaloric bioactives promote healthspan are not well understood, limiting their use in disease prevention. Here, we deploy a whole-organism, high-content screen in zebrafish to profile food-derived compounds for activation of autophagy, a cellular quality control mechanism that promotes healthy aging. We identify thymol and carvacrol as activators of autophagy and mitophagy through a transient dampening of the mitochondrial membrane potentia

Carvacrol modulates antioxidant enzymes, DNA integrity, and apoptotic markers in zearalenone-exposed fetal rat liver.

Maternal exposure to zearalenone (ZEA), a mycotoxin, can impact fetal liver development. This study investigated the protective effects of carvacrol (CRV) against ZEA-induced fetal liver damage. Thirty-two pregnant rats were allocated to four groups (eight rats/group); control, CRV (75 mg/kg), ZEA (5 mg/kg), and co-treated group (ZEA + CRV). The animals were given their doses during the gestation period. Maternal exposure to ZEA revealed a significant increase in the malondialdehyde (MDA) level in the fetal liver. In contrast, glutathione S-transferase (GST), superoxide dismutase (SOD), and ca

Carvacrol Derivatives as Antifungal Agents: Synthesis, Antimicrobial Activity and in Silico Studies on Carvacryl Esters.

Chemical modifications of natural monoterpenoids to various derivatives have been reported to result in enhancement of biological activities when compared to parent compounds. In this context a well-known biocide and food additive, carvacrol, served as a basic scaffold onto which a phenolic functionality transformation by introducing acyl groups was performed. By using this simple methodology, we obtained a small series of 25 esters. For each of the obtained compounds we have performed structural characterization, in vitro antimicrobial testing and in silico calculation of physico-chemical, ph

Bio-inspired multifunctional smooth lubricant-infused surface for residue-free, antimicrobial, and real-time freshness-monitoring food packaging.

Residual contamination, microbial growth, and the inability to monitor freshness in real-time are critical challenges in food packaging that affect safety and increase waste. Herein, we propose a novel multifunctional smooth lubricant-infused surface (SLIS) designed to address these issues. The SLIS was fabricated by infusing a porous carnauba wax coating with a functional water-in-oil (W/O) emulsion containing carvacrol (dissolved in oleic acid) and anthocyanin (dissolved in water). This SLIS exhibited excellent residue-free properties against common food liquids, and the addition of carvacro

Terpenes and terpenoids modulating TRPV channels in diabetes: role in fibrosis and inflammatory signaling.

Transient receptor potential vanilloid (TRPV) channels are non-selective cation channels that mediate thermosensation, nociception, osmotic balance, and metabolic regulation. In diabetes, altered TRPV channel expression and decreased calcium influx key pathways modulates, include NF-κB, AMPK, and TGF-β, promoting extracellular matrix accumulation and fibrotic remodelling. The limited availability of pathway specific interventions highlights TRPV channels as potential therapeutic target in diabetes. Recently, natural phytoconstituents have shown to exhibit TRPV channel modulating properties. St
