Bio-based nanophotothermal systems as smart drug delivery platforms for multimodal synergistic cancer therapy-a review.

Conventional protocols for cancer treatment are usually accompanied by serious limitations, which can be addressed using innovative, minimally invasive methods, such as photothermal therapy (PTT). But, using this method as a standalone therapy is usually accompanied by uneven heat distribution within tumor tissues, and the risk of cancer recurrence. Integrating PTT with complementary therapeutic modalities can help prevent these issues. The development of nanophotothermal systems (NPTSs) as multipurpose, multimodal platforms for smart drug delivery has greatly broadened the therapeutic landsca

Multiplex Detection and Quantification of miRNAs in Drug Delivery Systems Using a Signal-Off Electrochemical Platform.

Lipid nanoparticles (LNPs) are widely used for the delivery of therapeutic microRNAs (miRNAs), especially in coformulated systems where multiple targets are combined to enhance therapeutic efficacy. However, the simultaneous quantification of miRNAs within LNP formulations remains a major analytical challenge for pharmaceutical quality control. This work presents a dual-working electrochemical biosensor for multiplexed, selective, and amplification-free quantification of two miRNAs (miR-4676 and miR-6503) encapsulated in LNPs. The biosensor was fabricated on a dual-working screen-printed graph

Engineering responsive polymeric drug delivery systems via bioorthogonal and click chemistry: From molecular design to function.

Stimuli-responsive polymer systems have been rapidly gaining attention for the targeted release of therapeutics. Such systems often require the construction of complex, multi-functional materials. Advancements in click and bioorthogonal reactions have therefore been increasingly used to prepare these polymeric materials, both through their synthesis in the lab as well as in their assembly in vivo at the target site. Furthermore, such reactions can even be used to trigger therapeutic release. This review will cover the main bioorthogonal and click reactions used for polymeric delivery systems i

Rheology-guided experimental design of dynamically crosslinked keratin/ι-carrageenan hydrogels as potential drug delivery systems.

This study establishes a deterministic framework for designing 3D-printable, dynamically crosslinked hydrogel inks for controlled drug delivery via Direct Ink Writing (DIW). By blending cysteine-S-sulfonated keratin with ι-carrageenan through Ca2+ ionotropic gelation, shear-responsive networks were created. A Design of Experiments (DoE) approach generated a predictive rheology-to-printability map correlating composition with storage modulus (G'), flow stress (τf), and printability index (Pr). Advanced Large Amplitude Oscillatory Shear (LAOS) profiling revealed a transition from ductile, entang

Self-Nanoemulsifying Drug Delivery Systems for Cardiovascular Therapeutics: Formulation Strategies, Pharmacokinetic Enhancement and Precision-Oriented Translation.

Cardiovascular drugs face many barriers to oral delivery, and self-nanoemulsifying drug delivery systems (SNEDDS) are becoming a promising nano-enabled drug delivery vehicle to address these challenges and aid precision cardiovascular delivery. The poor aqueous solubility and/or intestinal permeability, combined with high intestinal and hepatic clearance and, in some cases, intestinal P-glycoprotein (P-gp) efflux, leads to variable systemic exposure and inconsistent therapeutic activity of many antihypertensive drugs, antianginal drugs, and cardioprotective agents. Under gastrointestinal dilut…

Combined Targeted Drug Delivery Systems for Mineral Metabolism Disorders in Chronic Kidney Disease: A Narrative Review.

BACKGROUND: The cardio-renal-bone axis in chronic kidney disease-mineral and bone disorder (CKD-MBD) couples vascular calcification with osteoporosis. Non-specific systemic drug distribution creates a "calcium paradox", where treating one compartment worsens the other. OBJECTIVE: This narrative review evaluates combined targeted drug-delivery systems for CKD-MBD, focusing on mineral metabolism outcomes in vascular and skeletal tissues. METHODS: We searched PubMed, Embase, Web of Science, Cochrane Library, and ClinicalTrials.gov from inception to May 2026. Search terms combined MeSH headings

Advances in Novel Drug Delivery Systems for Targeting the Brain: Overcoming Barriers and Enhancing Therapeutic Efficacy.

The blood-brain barrier (BBB) is critical in CNS pharmacotherapy; it blocks the entry of 98 per cent of small-molecule compounds and almost all macromolecules into the brain parenchyma via tight junction complexes, efflux transporter action, and enzymatic breakdown. This review focuses on the mechanistic basis of BBB resistance and analyses the current landscape of new drug delivery systems developed to overcome this barrier. We elaborate on polymeric nanocarriers, lipid-based nanocarriers, exosomes, dendrimers, and metallic nanoparticles, including surface engineering, the possibility of rece…

Advances in nano-drug delivery systems for metallic compounds in cancer therapy: challenges and future perspectives.

Metallic compounds have shown great promise as anticancer treatments because of their varied mechanisms of action, decreased side effects, and ability to overcome drug resistance. The search for alternative metal-based therapies has been driven by the severe toxicity, drug resistance, and poor selectivity of platinum-based complexes like cisplatin, carboplatin, and oxaliplatin, despite their notable clinical effectiveness. Their clinical translation is made difficult by issues such as off-target toxicity, low absorption, and poor solubility. These findings highlight the potential of nanomedici

Excipients in drug delivery systems: A comprehensive review of approved inactive ingredients for human ophthalmic formulations.

Pharmaceutical excipients, commonly known as inactive ingredients, encompass any substance aside from the active ingredient that fulfills a distinct and vital role in a formulation. Their purpose is to enhance specific characteristics, whether associated with the performance of the formulation or aspects related to patient comfort, safety, and acceptability. Because of the limited toxicity studies provided, and the several allergic and toxic side effects that have been reported throughout the years, it is not trivial for the regulatory agencies to approve inactive ingredients for human use.
