Enhancing Maturation of Human Neuromuscular Organoids via Electrical Stimulation.

Organoids derived from human pluripotent stem cells (hPSCs) are emerging as powerful models for studying development and disease. Despite their physiological relevance, the predictive power of organoids remains limited by the immature state of the constituent cells, posing a major challenge for mechanistic studies of adult physiology and late-onset disorders. Here, we establish a strategy for enhancing the maturation status of human neuromuscular organoids (NMOs) through chronic Electrical Pulse Stimulation (EPS). We demonstrate that low-frequency EPS, applied during the early stages of NMO de

Pediatric considerations in methodology of electrical brain stimulation during SEEG.

OBJECTIVES: Electrical stimulation (ES) during stereoelectroencephalography (SEEG) plays a key role in functional mapping in drug-resistant epilepsy. In pediatrics, protocols must account for neurodevelopmental stage. Moreover, pediatric data remain scarce and protocols vary substantially across centers. Such heterogeneity limits comparability between studies and hinders the evaluation of stimulation efficacy and its prognostic value for postoperative functional deficits. This work aims to provide expert consensus to harmonize SEEG stimulation protocols in the pediatric population. METHODS: A

Exploring Sensorimotor Physiology and Cortical Mapping With Stereoelectroencephalography Electrical Stimulation.

Stereoelectroencephalography (SEEG) has materially broadened the landscape of sensorimotor mapping by enabling access to sulcal, medial, and subcortical structures that remain largely beyond the reach of subdural electrodes (SDE). The inherent sampling bias of SDE toward gyral crests and dorsolateral cortices constrains the exploration of the central sulcus-where primary motor and sensory cortices predominantly reside-and limits insight into deeper epileptogenic networks. In routine practice, this topographic restriction is not trivial; it shapes both our hypotheses and our blind spots. SEEG a

Development and Testing of a Transcutaneous Electrical Nerve Stimulation Device for Pilots.

INTRODUCTION: Neck/back pain is a significant problem for long-haul pilots. It accounts for an extensive burden in time and resources to the U.S. and international partners. Transcutaneous electrical nerve stimulation (TENS) is an established intervention for treating pain. However, existing solutions and form factors preclude use of such devices while flying. MATERIALS AND METHODS: We developed an innovative patented wrist-worn TENS device with large tactile buttons for operation through the flight suit. We assessed range of motion (ROM) in 26 participants using the device with either a neck

Residual photoreceptors affect the response of a degenerate retina to electrical stimulation.

Photovoltaic subretinal prosthesis can restore central vision in patients blinded by age-related macular degeneration with letter acuity matching its 100 µm pixel size. Improving resolution requires smaller pixels, but to still reach the target neurons, electric field should be less confined. However, wide-spreading field may engage adjacent photoreceptors and alter the visual perception. We studied the effects of residual photoreceptors on retinal responses to electrical stimulation using monopolar and bipolar photovoltaic arrays implanted subretinally in Long Evans rats with local photorecep

Extraocular Electrical Stimulation Activates Retinal Ganglion Cells In Vivo.

OBJECTIVE: Here, we directly demonstrate that extraocular electrical stimulation can reliably activate retinal ganglion cells (RGCs) in vivo and systematically identify optimal stimulation waveforms that maximize RGC activation at tolerable amplitudes. APPROACH: Using transpupillary two-photon calcium imaging in Thy1-GCaMP6f rats, we directly visualized RGC activation during extraocular electrical stimulation with single-cell resolution. We tested symmetric (SCB 1:1) and asymmetric (ACB 1:4) charge-balanced waveforms across frequencies ranging from 20 to 5,000 Hz and amplitudes from 1 to 300

Spinal excitability following sensory electrical stimulation of the upper limb.

The purpose of the study was to determine if sensory electrical stimulation (SES), below motor threshold, would reduce spinal excitability via reciprocal inhibition (RI) and determine if any changes were sex-related. Eighteen healthy participants (11 males and 7 females) participated in a pre-post comparison study. The Hoffmann (H-) reflex was elicited to assess the spinal excitability of Flexor Carpi Radialis (FCR) and the influence of RI from Extensor Carpi Radialis Longus (ECRL) on FCR using a paired conditioning pulse paradigm. A 15-min bout of SES (4 pulse bursts at 100 Hz) was applied to

Cross-modal spicy enhancement with odour and electrical tongue stimulation.

Spicy substances like chili pepper and wasabi can have both beneficial and adverse effects on human health. In particular, they may help to reduce salt intake and limit fat absorption, when consumed in appropriate amounts. However, excessive consumption of spicy foods could increase the desire for more intense spiciness, potentially leading to overconsumption. To address the challenge of balancing the amount of spicy substances in food and the desired level of perceived spiciness, electrical tongue stimulation (ETS) has been proposed as a method of sensory enhancement. Although promising, the

Transcorneal Electrical Stimulation Modulates Visual Pathway Function in Mice.

Due to its ability to modulate neuronal activity, electrical stimulation of the eye may be a promising therapy for preserving or restoring vision. To investigate how electrical currents can influence visual function, Transcorneal Electrical Stimulation (TES) was tested on both female and male C57BL/6 mice to evaluate its neuromodulatory effect from the retina to the cerebral cortex through visual evoked potential (VEP) and electroretinogram (ERG) recording. VEP or ERG was acquired before (baseline), immediately (t0), after 5 min (t5), and 10 min (t10) of sham (i.e., no stimulation) or TES appl

Modeling and control of functional electrical stimulation cycling training system.

Functional electrical stimulation (FES), a rehabilitation technique, typically relies on physiotherapists using trial-and-error tests to determine effective stimulation patterns. Therefore, this study proposed a kind of pedal hill modeling to establish an optimal stimulus mode with the maximum torque efficiency optimization objective. This study also proposed a new model based on the particle swarm optimization (PSO) algorithm, the back propagation (BP) neural network algorithm, and the proportional integral derivative (PID) control composite algorithm. Six participants were recruited for the

Improving working memory by electrical stimulation and cross-frequency coupling.

Working memory (WM) is essential for the temporary storage and processing of information required for complex cognitive tasks and relies on neuronal theta and gamma oscillations. Given the limited capacity of WM, researchers have investigated various methods to improve it, including transcranial alternating current stimulation (tACS), which modulates brain activity at specific frequencies. One particularly promising approach is theta-gamma peak-coupled-tACS (TGCp-tACS), which simulates the natural interaction between theta and gamma oscillations that occurs during cognitive control in the brai

The Effect of Cranial Sutures Should Be Considered in Transcranial Electrical Stimulation.

BACKGROUND: Computational modeling is used to optimize transcranial electrical stimulation (tES) approaches, and the precision of these models is dependent on their anatomical accuracy. We are unaware of any computational modeling of tES that has included cranial sutures. OBJECTIVES: The aims of the study were to review the literature on the timing of closure of the coronal and squamous sutures, which are situated under electrode placements used in tES; to review the literature regarding differences in skull and suture conductivity and to determine a more accurate conductivity for sutures; an

Electrical stimulation of injured nerves promotes recovery in animals and humans.

The frequent poor functional outcomes after delayed surgical repair of injured human peripheral nerves results in progressive downregulation of growth-associated genes in parallel with reduced neuronal regenerative capacity under each of the experimental conditions of chronic axotomy of neurones that remain without target contact, chronic distal nerve stump denervation, and chronic muscle denervation. Brief (1 h) low-frequency (20 Hz) electrical stimulation (ES) accelerates the outgrowth of regenerating axons across the surgical site of microsurgical repair of a transected nerve. Exercise prog

The effect of neuromuscular electrical stimulation on the human skeletal muscle transcriptome.

AIM: The influence on acute skeletal muscle transcriptomics of neuromuscular electrical stimulation (NMES), as compared to established exercises, is poorly understood. We aimed to investigate the effects on global mRNA-expression in the quadriceps muscle early after a single NMES-session, compared to the effects of voluntary knee extension exercise (EX), and to explore the discomfort level. METHODS: Global vastus lateralis muscle gene expression was assessed (RNA-sequencing) in 30 healthy participants, before and 3 h after a 30-min session of NMES and/or EX. The NMES-treatment was applied usi
