Variations in enzyme activity in stomach and pancreatic tissue and digesta in piglets around weaning.

A study was performed to investigate the effect of weaning at 4 weeks of age on the activity of digestive enzymes in the stomach and pancreatic tissue and in digesta from 3 days prior to weaning to 9 days postweaning in 64 piglets. In stomach tissue the activity of pepsin and gastric lipase was determined. Pepsin activity declined abruptly after weaning but 5 days postweaning the weaning level was regained and in the gastric contents no change in pepsin activity was observed. Weaning did not influence the activity of gastric lipase. The activity of eight enzymes and a cofactor was measured in

Poloxamer 407-mediated alterations in the activities of enzymes regulating lipid metabolism in rats.

PURPOSE: Recently, the P-407-treated mouse was established as a useful animal model of hyperlipidemia and atherosclerosis. The present study was aimed to determine whether P-407-induced hyperlipidemia in the rat is associated with alterations in the activities of enzymes responsible for lipid metabolism. METHODS AND RESULTS: Rats were made hyperlipidemic by i.p. injection of 1.0 g/kg P-407 and blood samples collected 24 h after administration of P-407. Plasma from P-407-treated rats demonstrated 7- and 13-fold increases in cholesterol and triglycerides, respectively (p < 0.001). The plasma le

Regulation of the angiotensin-converting enzyme activity by a time-course hypoxia in the carotid body.

Chronic hypoxia activates a local angiotensin-generating system in the carotid body. Here, we test the hypothesis that the activity of the critical enzyme for this system, angiotensin-converting enzyme (ACE), in the carotid body is subject to regulation by a time-course hypoxia. Results from the carotid body assays showed that ACE activity was markedly increased under the hypoxic stress of 7-, 14-, 21-, and 28-day exposures. The changes in ACE activity of 7-day (15.00 vs. 30.95 x 10(-5) nmol.microg(-1).min(-1)), 14-day (8.73 vs. 30.25 x 10(-5) nmol.microg(-1).min(-1)), and 21-day (11.41 vs. 31

Propylthiouracil alters the expression and activity of glutathione-dependent enzymes in the mouse olfactory mucosa.

Propylthiouracil (PTU) is an anti-thyroid drug that reportedly can impair olfactory function in humans and mice. In the mouse, PTU treatment disrupts survival and differentiation of olfactory precursor neurons. While the mechanism responsible for this effect is not understood, it is suspected that these changes are consequent to localized toxicity due to PTU metabolism. In vitro and in vivo studies in other biological systems demonstrate that PTU can significantly alter glutathione S-transferase (GST) enzyme expression and activity. The localization of GST biotransformation enzymes in basal ce

Decreased hepatic drug metabolising enzyme activity in rats with nitrosamine-induced tumours.

N-Methyl N-benzyl nitrosamine (MBNA), which requires P450-dependant activation to be mutagenic, has been shown to produce squamous cell carcinoma of rat oesophagus. The aim of this study was to determine the effects of tumour induction on hepatic cytochrome P450 (CYP) and phase II enzyme activity. Female Wistar rats were given MBNA (2.5 mg/kg) by gavage, twice weekly for 12 weeks. At the end of 12 weeks they were sacrificed; livers and oesophagi were removed. The activity of hepatic CYP and phase II enzymes was determined by incubation of liver microsomes with appropriate CYP substrates. All r

Lactulose-induced diarrhoea in rats: effects on caecal development and activities of microbial enzymes.

The intake of large amounts of lactulose and other non-digestible oligosaccharides can cause diarrhoea in rats and humans. The purpose of our study was to estimate tendency and scope of changes in caecum development, amount and composition of caecal digesta and activity of caecal microbial enzymes under the influence of lactulose-rich diet evoking or not evoking diarrhoea. Male Wistar rats were fed on 8%-lactulose diet for 4 weeks. Feeding with lactulose induced enlargement of the caecum (digesta and wall) compared to the control group. However, the hypertrophy of the caecal wall in rats with

[Correction with zaditen of impaired activity of pancreatic digestive enzymes in sensitization and anaphylactic shock].

The effect of the antihistaminic drug zaditen on the activity of pancreatic digestive enzymes was studied in white rats under sensitization and anaphylactic shock conditions. The sensitization and anaphylaxis are accompanied by significant changes in the spectrum and activity of pancreatic enzymes (alpha amylase, protease complex, lipase), leading to disorders in the digestion and assimilation of proteins, fats, and carbohydrates. Zaditen reduced negative changes in the enzyme secretion system of pancreas under conditions of this pathology.

Antioxidative enzyme activities and lipid peroxidation in major depression: alterations by antidepressant treatments.

BACKGROUND: Reactive oxygen species (ROS) may play a role in some neuropsychiatric disorders. There is some evidence that the activation of immune-inflammatory process, increase of monoamines catabolism, and abnormalities in lipid compounds may cause overproduction of ROS and, in turn, antioxidative enzyme activities (AEAs) and lipid peroxidation (LP), and that these phenomena may be related to pathophysiology of major depression. METHODS: The aims of this study were (i) to examine the AEAs and LP levels of the major depressed (MD) patients, and to compare these with healthy controls; and (ii

The influence of intravenous anaesthetics on the activity of enzymes released from polymorphonuclear leucocytes in vitro.

BACKGROUND AND OBJECTIVE: Polymorphonuclear leucocytes make a decisive contribution to defence against bacterial infections. In particular, the effects of anaesthetics on non-oxidative bactericidal mechanisms have previously only been superficially examined. Although the influence of anaesthetic agents on oxidative bactericidal activity has been thoroughly examined, our study concentrated on the effect on non-oxidative processes, which appears to have been a neglected field of research. METHODS: The effects of methohexital, etomidate, ketamine, fentanyl and morphine on the activity of lysozym

Thyroid function and serum hepatic enzyme activity in dogs after phenobarbital administration.

Phenobarbital is the drug of choice for control of canine epilepsy. Phenobarbital induces hepatic enzyme activity, can be hepatotoxic, and decreases serum thyroxine (T4) concentrations in some dogs. The duration of liver enzyme induction and T4 concentration decreases after discontinuation of phenobarbital is unknown. The purpose of this study was to characterize the changes in serum total T4 (TT4), free T4 (FT4), thyroid-stimulating hormone (TSH), cholesterol and albumin concentrations, and activities in serum of alanine aminotransferase (ALT), alkaline phosphatase (ALP), and gamma-glutamyl t

Erythrocyte antioxidant enzyme activities in coal miners from three French regions.

This study investigated whether differences in the prevalence and severity of coal workers' pneumoconiosis (CWP) between three coal mines could be related to differences in oxidative stress exposure as evaluated in vivo through red-blood-cell antioxidant enzyme activities. Blood samples were obtained from 229 miners selected according to their occupation and their pneumoconiotic status. The following biomarkers were evaluated: erythrocyte catalase, Cu2+/Zn2+ superoxide dismutase (Cu2+/Zn2+ SOD), and glutathione peroxidase activities. Antioxidant enzyme activities did not differ significantly b

Activity of mitochondrial respiratory chain enzymes after transient focal ischemia in the rat.

Previous results demonstrated that after 2-hour middle cerebral artery occlusion (MCAO) in the rat, 1- to 2-hour recirculation temporarily restored the bioenergetic state and mitochondrial function, but secondary deterioration took place after 4 hours. The authors measured the activity of mitochondrial respiratory chain complexes, citrate synthase, and glutamate dehydrogenase as possible targets of secondary damage. Focal and penumbral tissues were sampled in the control condition, after 2 hours of MCAO, and after 1, 2, or 4 hours of postischemic recirculation; two groups were treated with alp

An extracellular enzyme with hyaluronidase and chondroitinase activities from some oral anaerobic spirochaetes.

Treponema denticola, Treponema vincentii and Treponema socranskii produce an enzyme that hydrolyses hyaluronic acid (HA) and chondroitin sulphate (CS). The secreted enzyme is specifically inhibited by gold sodium thiomalate and anti-bee-venom antibodies. The use of saturated substrate (HA or CS) transblots allowed the visualization of active enzyme directly from culture supernatants and is a useful tool in clarification of complex polysaccharide-degrading enzyme specificities. The affinity-purified extracellular enzyme of T. denticola contains a single molecular species with a molecular mass o

Modulation of carcinogen-induced DNA damage and repair enzyme activity by dietary riboflavin.

Formation of single strand breaks in nuclear DNA induced by hepatocarcinogens aflatoxin B1 and N-nitrosodimethylamine was observed to be more pronounced in rats maintained on a riboflavin-deficient diet compared to that on a normal diet. This increased damage was reversed on riboflavin supplementation. The induction of repair enzymes poly(ADP-ribose) polymerase, DNA polymerase beta and DNA ligase was significantly higher in riboflavin-deficient rats following DNA damage caused by the administration of carcinogens. Riboflavin supplementation brought down the induction to the levels found in rat

Melatonin stimulates the activity of the detoxifying enzyme glutathione peroxidase in several tissues of chicks.

The pineal hormone melatonin has been shown to directly scavenge free radicals and to stimulate, in the mammalian brain, at least one enzyme, glutathione peroxidase, which reduces free radical generation. In the present studies, we examined the effect of melatonin on glutathione peroxidase activity in several tissues of an avian species. Melatonin (500 micrograms/kg), when injected into chicks, increased glutathione peroxidase activity within 90 min in every tissue examined. Tissue melatonin levels, measured by radioimmunoassay, also increased following its peripheral administration. Depending

Involvement of proteolytic enzymes--plasminogen activators and matrix metalloproteinases--in the pathophysiology of pressure ulcers.

The role of matrix-degrading enzymes, particularly plasminogen activators and matrix metalloproteinases, in the acute wound healing response has been the focus of many scientific studies. Only recently have these classes of endogenously produced proteinases been studied with regard to their involvement in the chronic wound environment. Using both in situ histologic zymography and immunohistochemical techniques, we examined the distribution of plasminogen activators and matrix metalloproteinase in the granulation tissue of pressure ulcers. Using in situ histologic zymography, urokinase was foun

[The effect of methotrexate on activity of T-lymphocyte marker enzymes in patients with psoriasis vulgaris].

Radiochromatographic methods were used in order to ascertain the activity of adenosinedeaminase and purinnucleoside phosphorylase in lymphocytes isolated from the venous blood of 32 patients with psoriasis vulgaris in comparison with enzyme activities in 28 clinically healthy subjects. 11 selected patients with a severe form of psoriasis were subdued to therapy by means of methotrexate and its effect on the activity of these enzymes was investigated. Untreated patients yielded a statistically significant increase in the activities of adenosinedeaminase and purinnucleoside phosphorylase in comp
