Hypophosphatasia and the importance of the general dental practitioner - a case series and discussion of upcoming treatments.

Hypophosphatasia (HPP) is an inherited metabolic disorder that results in poorly mineralised bones and teeth. Clinical symptoms vary widely from mild dental anomalies to fatal fetal defects. The most common dental symptoms include exfoliation of the primary incisors before the age of three with little or no root resorption, large pulp chambers, alveolar bone loss and thin dentinal walls. There is generally minimal periodontal inflammation associated with the bony destruction and tooth loss. The general dental practitioner is usually the first clinician to spot signs of the milder forms of HPP.

Teratological study of the herbicide 4-chloro-2-methylphenoxyacetic acid in rabbits.

4-Chloro-2-methylphenoxyacetic acid (MCPA) is an aryloxyacetic acid derivative categorised as a plant hormone herbicide. The aim of the study was to evaluate the effect of MCPA on pregnant females and the prenatal development of rabbits. The substance tested was administered orally to pregnant New Zealand White rabbits from day 6 to day 27 of gestation at doses of 5, 10 and 25 mg kg(-1) day(-1). The animals were killed on day 28 of gestation and live fetuses were examined for gross, skeletal and visceral anomalies. Administration of MPCA did not induce any signs of maternal toxicity. There was

Toxicological consequences of chloroquine and ethanol on the developing fetus.

In the present study, the effects of chloroquine and ethanol administration during gestation have been investigated on the developing rat fetus. Intragastric administration of chloroquine (700 mg/kg body weight) resulted in several structural abnormalities. The incidence of hepatomegaly was increased by 30%, the liquification of visceral organs was increased by 15% and a 9% higher incidence of cleft palate, wrist drop, clubbed foot and brain liquification was observed in the fetuses from the chloroquine-treated group compared to the corresponding controls. Fetuses from the chloroquine-treated

Effects of low medium magnesium concentration on bone resorption in response to parathyroid hormone and 1,25-dihydroxyvitamin D in organ culture.

We have examined the effects of decreasing the magnesium concentration in the medium on bone resorption in organ culture. During a 5-day culture, low magnesium (0.25 vs. 0.8 mM) caused a decrease in unstimulated 45Ca release from live and killed fetal rat bones for the first 2 days but not thereafter, suggesting an early and transient alteration in physicochemical equilibrium for calcium between bone and extracellular fluid in magnesium deficiency. In addition, 45Ca release in response to low and intermediate doses of PTH and 1,25-dihydroxyvitamin D3 was significantly impaired throughout the 5

Maternal hyperthyroidism increases the prevalence of foregut atresias in fetal rats exposed to adriamycin.

PURPOSE: Gastrointestinal malformations such as esophageal atresia with tracheoesophageal fistula (EA/TEF) and duodenal atresia (DA) have been reported in infants born to hyperthyroid mothers or with congenital hypothyroidism. The present study aimed to test whether maternal thyroid status during embryonic foregut division has any influence on the prevalence of EA/TEF and DA in an accepted rat model of these malformations. METHODS: Pregnant rats received either vehicle or 1.75 mg/kg i.p. adriamycin on gestational days 7, 8 and 9. Transient maternal hyper or hypothyroidism was induced by oral

Bone marker status in mothers and their newborns in Iran.

OBJECTIVES: The contribution of maternal skeletal calcium metabolism in pregnancy is evidenced in changes in the markers of bone formation and bone resorption. Changes in maternal bone markers could affect fetal bone mineralization. The aim of this study was to determine the association between maternal and cord blood bone markers. METHODS: Five hundred and fifty-two pregnant women were recruited from Tehran University educating hospitals in the winter of 2002. Maternal and cord blood samples were taken at delivery. The serum was assayed for osteocalcin and crosslaps, calcium, and parathyroid

The vanishing twin.

In general, it has not been recognized that many twin or multiple gestations are lost in utero early in pregnancy. Until the advent of ultrasound, the ability to document early human fetal loss in multiple gestation was difficult. However, recent reports of serial ultrasound examinations of pregnant women have documented the "disappearance" of at least one of two gestational rings. Furthermore, the number of twins observed at delivery was significantly less than the number of twin conceptions originally identified by ultrasound during the first trimester. These observations led to the concept

Fetal alcohol syndrome (FAS) in C57BL/6 mice detected through proteomics screening of the amniotic fluid.

BACKGROUND: Fetal Alcohol Syndrome (FAS), a severe consequence of the Fetal Alcohol Spectrum Disorders, is associated with craniofacial defects, mental retardation, and stunted growth. Previous studies in C57BL/6J and C57BL/6N mice provide evidence that alcohol-induced pathogenesis follows early changes in gene expression within specific molecular pathways in the embryonic headfold. Whereas the former (B6J) pregnancies carry a high-risk for dysmorphogenesis following maternal exposure to 2.9 g/kg alcohol (two injections spaced 4.0 h apart on gestation day 8), the latter (B6N) pregnancies carry

[Disorders of calcium metabolism during pregnancy and breast feeding].

Hypoparathyroidism following thyroidectomy or due to autoimmune process is the most common cause of hypocalcaemia. If untreated, maternal hypocalcaemia may stimulate fetal parathyroids which results in bone demineralization. Calcium supplementation may also reduce the risk of hypertension. Moreover, hypocalcaemia may inhibit the synthesis of calcium gene-related peptide (CGRP), which, in turn, decreases blood pressure and uterus contractions. In women of reproductive age hypercalcaemia is mostly due to hyperparathyroidism. The complications include toxemia, vomiting, and hypercalcaemic crisis.

Sensitive stages and dose-response analyses of palatal slit and cleft palate in C57BL/6 mice treated with a glucocorticoid.

C57BL/6 mice were treated subcutaneously with triamcinolone acetonide in a single dose of 2.5-15.0 mg/kg once on days 6-15 of pregnancy (vaginal plug = day 0) and the palate of their fetuses was examined at term. The sensitive stages of palatal slit and cleft palate induction were studied and dose-response relations on days 9 and 12 of pregnancy were analyzed by the log-probit transformation method and compared. The test of significant increase in the frequencies of palatal slit and cleft palate showed that the sensitive period of palatal slit began earlier and ended later than that of cleft p

Fetal toxicity of benzodiazepines in rats.

This study was undertaken to evaluate fetal toxicity of benzodiazepine derivatives, nitrazepam, oxazepam, nimetazepam, clonazepam, diazepam and chlordiazepoxide. In fetotoxicities induced by the oral administration of these six drugs (100 mg/kg), nitrazepam (NTZ) and nimetazepam caused a great damage to the fetus compared with other analogues. The fetotoxicity of NTZ was dose-dependent at a dose of 50 to 100 mg/kg p.o. and its peak toxicity was observed in the group treated from the 8th to 10th day of gestation. Maternal serum and whole fetus concentration of NTZ were higher than that of diaze

Distribution and effects of intravenous lead in the fetoplacental unit of the rat.

Lead metabolism was studied in the fetoplacental unit (FPU) of Wistar rats during the genesis of developmental abnormalities and embryonic death. Female rats were injected iv with tracer 210Pb(NO3)2, alone or in combination with 5 or 25 mg Pb(NO3)2/kg, at 9 or 15 days of gestation (dg). The distribution of lead and its effects were determined in the FPUs during the ensuing 30-h period and at 20 dg. Hemorrhage of the egg cylinder was noted as early as 6 h postinjection of 25 mg/kg at 9 dg. By 20 dg, fetuses exhibited characteristic stunting and external malformations (gastroschisis and severe s

The effect of progesterone on fetal and placental development in normal and protein-energy-restricted rats.

Maternal protein-energy restriction (25% of the ad libitum intake) during the first 10 days of pregnancy resulted in severely altered fetal growth rates. Fetal development was assessed by body weight, brain weight, brain DNA, and brain protein content on fetal days 16, 18, 20 and at term. The individual placentas were also examined (weight, DNA and protein content) on each of these fetal days. Progesterone was administered commencing with day 3 of pregnancy until the day Caesarian section was done, in an attempt to rehabilitate placental development. This treatment did not improve placental de

Fetal and placental responses to artificially induced hyperthermia in rats.

Pregnant rats were utilized to study the effect of maternal hyperthermia on fetal development. Eight groups of six to eight rats were exposed to ambient temperatures of 43-44 degrees C at various stages of pregnancy. All rats were killed on day 20 of gestation. Edema, microencephaly and microphthalmia followed heat treatment on day 4, 6, or 8 and skeletal defects occurred on day 10 of gestation. Apparently heat stress of dams after day 14 of gestation had little or no effect on embryos. Most placentas from day 6-10 treatment groups were significantly heavier than control and exhibited extensiv

Loss of function of the tuberous sclerosis 2 tumor suppressor gene results in embryonic lethality characterized by disrupted neuroepithelial growth and development.

Germline defects in the tuberous sclerosis 2 (TSC2) tumor suppressor gene predispose humans and rats to benign and malignant lesions in a variety of tissues. The brain is among the most profoundly affected organs in tuberous sclerosis (TSC) patients and is the site of development of the cortical tubers for which the hereditary syndrome is named. A spontaneous germline inactivation of the Tsc2 locus has been described in an animal model, the Eker rat. We report that the homozygous state of this mutation (Tsc2(Ek/Ek)) was lethal in mid-gestation (the equivalent of mouse E9.5-E13.5), when Tsc2 mR

Pre-clinical validation of a vaginal cream containing copaiba oil (reproductive toxicology study).

The aims of this study was to evaluate the effects of oil-resin of Copaiba (Copaifera duckei Dwyer), aired in vaginal cream on the reproductive performance of female rats (Rattus norvegicus). To determine the components of the C. duckei oleoresin, gas chromatography coupled with mass spectrometry (CG-MS) was used, and considering the trans-caryophyllene sesquiterpene as a phytochemical marker in the oleoresin. Due to the extensive use of copaiba oleoresin in the suppository form for gynecological infections, an evaluation was carried out on the effects of copaiba oleoresin (Copaifera duckei Dw
