Synthesis of and in vitro and in vivo evaluation of a novel TGF-β1-SF-CS three-dimensional scaffold for bone tissue engineering.

The role of transforming growth factor-β1 (TGF-β1) in normal human fracture healing has been previously demonstrated. The objective of the present study was to examine the biocompatibility of TGF-β1-silk fibroin-chitosan (TGF-β1-SF-CS) three-dimensional (3D) scaffolds in order to construct an ideal scaffold for bone tissue engineering. We added TGF-β1 directly to the SF-CS scaffold to construct a 3D scaffold for the first time, to the best of our knowledge, and performed evaluations to determine whether it may have potential applications as a growth factor delivery device. Bone marrow-derived

Trilayered Self-Pumping Dressing with Micropore Array for Drug Backflow Release in Diabetic Wound Healing.

Excessive exudate in chronic wounds increases the risk associated with tissue hydration, bacterial infection, and increased inflammation. While Janus dressings enable unidirectional exudate transport, current designs overlook the critical need for maintaining optimal wound humidity and providing on-demand antibacterial/anti-inflammatory treatment to support repair. To address this, a self-pumping trilayered dressing (GCSIP) is developed, integrating polyhexamethylene guanidine-grafted triglycidyl glycerol ether (PHMG-GTE)-modified cotton, ibuprofen-loaded silk fibroin (IBU/SF), and polyurethan

Natural Underwater Bioadhesive Offering Cohesion Modulation via Hydrogen Bond Disruptor: A Highly Injectable and in Vivo Stable Remedy for Gastric Ulcer Resolution.

Injectable bioadhesives are attractive for managing gastric ulcers through minimally invasive procedures. However, the formidable challenge is to develop bioadhesives that exhibit high injectability, rapidly adhere to lesion tissues with fast gelation, provide reliable protection in the harsh gastric environment, and simultaneously ensure stringent standards of biocompatibility. Here, a natural bioadhesive with tunable cohesion is developed based on the facile and controllable gelation between silk fibroin and tannic acid. By incorporating a hydrogen bond disruptor (urea or guanidine hydrochlo

In Situ Forming Hydrogel as a Tracer and Degradable Lacrimal Plug for Dry Eye Treatment.

Lacrimal plug is an effective and widely therapeutic strategy to treat dry eye. However, almost all commercialized plugs are fixed in a certain design and associated with many complications, such as spontaneous plug extrusion, epiphora, and granuloma and cannot be traced in the long-term. Herein, a simple in situ forming hydrogel is developed as a tracer and degradable lacrimal plug to achieve the best match with the irregular lacrimal passages. In this strategy, methacrylate-modified silk fibroin (SFMA) is served as a network, and a self-assembled indocyanine green fluorescence tracer nanopar

Cur@SF NPs alleviate Friedreich's ataxia in a mouse model through synergistic iron chelation and antioxidation.

Abnormal iron metabolism, mitochondrial dysfunction and the derived oxidative damage are the main pathogeneses of Friedrich's ataxia (FRDA), a single-gene inherited recessive neurodegenerative disease characterized by progressive cerebellar and sensory ataxia. This disease is caused by frataxin (FXN) mutation, which reduces FXN expression and impairs iron sulfur cluster biogenesis. To date, there is no effective therapy to treat this condition. Curcumin is proposed harboring excellent ability to resist oxidative stress through Nrf2 activation and its newly found ability to chelate iron. Howeve

Preparation of TEMPO-partially oxidized sodium alginate/SF scaffolds with degradation compatibility for skin defect repair.

This study aimed to curtail the secondary damage caused by traditional dressings by cross-linking TEMPO-mediated oxidized sodium alginate (TOSA) with silk fibroin (SF) through the EDC system and preparing the scaffolds through freeze-drying and anhydrous ethanol immersion. The good adsorption, mechanical properties, high porosity, and good in vitro and in vivo biocompatibility of the scaffold can improve the healing efficiency of the wound. Additionally, sodium alginate was oxidized by the TEMPO system to lower its molecular weight and hence increase the degradation rate of the scaffold. Conse

Harnessing decellularized amniotic membrane properties for ureteral repair: A fibromodulin-driven approach.

Ureteral reconstruction remains a significant challenge due to the risk of fibrosis and insufficient epithelial regeneration, leading to suboptimal outcomes with current surgical techniques. To address these issues, a tubular porous scaffold was developed by integrating enzymatically digested decellularized amniotic membrane (dAM) with silk fibroin and gelatin solutions, utilizing freeze-drying, compression, and cross-linking techniques. Proteomic analysis revealed that the scaffold retained key bioactive properties of dAM, promoting epithelial regeneration while inhibiting fibrosis in vitro a

Hagfish-inspired hydrogel for root caries: A multifunctional approach including immediate protection, antimicrobial phototherapy, and remineralization.

Root caries is the main cause of oral pain and tooth loss in the elderly. Protecting root lesions from environmental disturbances, resisting pathogens, and facilitating remineralization over time are essential for addressing root caries, but are challenging due to the irregular root surface and the complex oral environment. Hagfish secretes slime when facing danger, which converts into gels upon contact with seawater, suffocating the predators. Inspired by hagfish's defense mechanism, a fluid-hydrogel conversion strategy is proposed to establish a mechanical self-regulating multifunctional pla

Molecular mechanisms governing antibiotic transport in spider silk-riboflavin hydrogels.

Silk hydrogels are emerging as versatile biomaterials for drug delivery owing to their biocompatibility, biodegradability, and tunable hierarchical structure. Their trans-port properties are governed by the interplay between peptide secondary structure, hydration, and intermolecular interactions within the network. Understanding how small drug molecules, penicillin, diffuse through silk-based matrices at the molecular level is therefore critical for rational material design. Here, we studied the molecular mechanisms governing antibiotic, penicillin, transport in a spider silk-riboflavin hydrog

Suture-Free Piezoelectric Band-Aid Membrane for Complex Peripheral Nerve Defects.

The regeneration and functional rehabilitation of peripheral nerve defects remain major clinical challenges. Piezoelectric materials have the potential to restore both nerve morphology and function. Conventional grafts struggle to repair nerves with complex structures. To address this issue, band-aid-inspired tissue-adhesive membranes were designed to accommodate diverse nerve diameters and branching structures. Electrospinning was used to prepare a Janus membrane that served as a suture-free piezoelectric nerve repair membrane. The inner layer comprised oriented adhesive hydrogel fibers fabri

Body-temperature responsive hydrogel adhesive for active wound closure.

Hydrogels have extensive application in sutureless wound closure and healing. For relying on the natural recovery rate of the skin or tissue often results in prolonged healing periods due to the gradual process of wound contraction facilitated by cell migration, our approach involves the integration of two distinct layers to create a cohesive and innovative body-temperature responsive hydrogel tape (TRAT). The shape memory layer of P(PTHF-co-PEG-co-PETMP) possesses excellent mechanical properties and adjustable temperature-responsive characteristics. Upon integration with the adhesive layer co

Influence of the Sequence Motive Repeating Number on Protein Folding in Spider Silk Protein Films.

Like multiblock copolymers, spider silk proteins are built of repetitive sequence motives. One prominent repetitive motif is based on the consensus sequence of spidroin 4 of the spider Araneus diadematus ADF4. The number x of the repeating sequence motives (C) determines the molecular weight of the recombinant ADF4-based, engineered spider silk protein denoted as eADF4(Cx). eADF4(Cx) can be used as a model for intrinsically disordered proteins (IDP) and to elucidate their folding. Herein, the influence of the variation of the sequence motive repeating number x (x = 1, 2, 4, 8, 16) on the prote

Biofunctionalization of electrospun silk scaffolds with perlecan for vascular tissue engineering.

Electrospun silk fibroin scaffolds have garnered significant interest in vascular tissue engineering due to their biocompatibility, mechanical strength, and tunable degradation. However, their lack of intrinsic cell-binding domains limits endothelialization, a critical factor for vascular graft success. This study explores the biofunctionalization of electrospun silk scaffolds with recombinant perlecan domain V (rDV) using plasma immersion ion implantation (PIII) treatment, a surface modification method enabling robust covalent immobilization without the use of reagents. The biofunctionalized

Production of scFv-conjugated affinity silk film and its application to a novel enzyme-linked immunosorbent assay.

Bombyx mori (silkworm) silk proteins have been utilized as unique biomaterials for various medical applications. To develop a novel affinity silk material, we generated a transgenic silkworm that spins silk protein containing the fibroin L-chain linked with the single-chain variable fragment (scFv) as a fusion protein. Previously, the scFv-conjugated "affinity" silk powder specifically immunoprecipitated its target protein, Wiskott-Aldrich syndrome protein. To expand the applicability of affinity silk materials, we processed the scFv-conjugated silk protein into a thin film by dissolving it in

Skeletal muscle regeneration on protein-grafted and microchannel-patterned scaffold for hypopharyngeal tissue engineering.

In the field of tissue engineering, polymeric materials with high biocompatibility like polylactic acid and polyglycolic acid have been widely used for fabricating living constructs. For hypopharynx tissue engineering, skeletal muscle is one important functional part of the whole organ, which assembles the unidirectionally aligned myotubes. In this study, a polyurethane (PU) scaffold with microchannel patterns was used to provide aligning guidance for the seeded human myoblasts. Due to the low hydrophilicity of PU, the scaffold was grafted with silk fibroin (PU-SF) or gelatin (PU-Gel) to impro

Puerarin-Loaded Radially Aligned Tussah Silk/Chitosan/Graphene Oxide Nanofibrous Scaffolds for Enhanced Wound Healing.

Wound healing remains a critical clinical challenge due to uncontrolled bleeding, bacterial infections, poor vascularization, and cell migration at damaged tissue sites. To address this challenge, we developed a radially structured, multifunctional biomimetic scaffold. Tussah silk was subjected to physical shearing and alkaline hydrolysis treatment to obtain tussah silk nanofibers (TSn). The TSn, chitosan (CS), and graphene oxide (GO) were then processed into radially aligned TSn/CS/GO scaffolds. These anisotropic composite scaffolds demonstrated not only antibacterial activity and rapid hemos

Nerve conduits with multiple physical and biochemical guiding factors for peripheral nerve defects.

Peripheral nerve injury is a common complication in patients with trauma, and severe cases can result in sensory and motor function loss, affecting the quality of life. Although the peripheral nervous system has spontaneous regenerative ability, bridging long-distance nerve gaps remains a clinical challenge. Therefore, we innovatively constructed a multi-guided nerve conduit to address this issue. Using gelatin methacryloyl as the raw material, directional micropores were created through casting, demolding, and directional freezing to achieve topographical guidance. A concentration gradient of

Biomimetic multilayered hydrogel scaffold with dual cell types and mesh support for urethral reconstruction.

Urethral strictures and hypospadias remain major challenges in urology, with current treatments limited by donor-site morbidity, graft shrinkage, and poor long-term outcomes. To address these limitations, we developed a biomimetic multilayered hydrogel scaffold with dual cell types and mesh reinforcement for urethral reconstruction. The construct combined predifferentiated buccal mucosa cells and adipose-derived mesenchymal stem cells within a gelatin methacrylate-silk fibroin hydrogel supported by electrowritten and electrospun polymeric meshes, designed to reproduce both epithelial and smoot
