Benzodiazepine receptors in the medial-posterior hypothalamus mediate the reduction of hippocampal theta frequency by chlordiazepoxide.

Separate sub-populations of benzodiazepine receptors are involved in determining hippocampal theta frequency in freely moving rats. Systemic injection of the benzodiazepine chlordiazepoxide (CDP) reduced the frequency of theta elicited by stimulation of the brainstem reticular formation, and theta recorded during fixed interval (FI) bar pressing. Infusion of the benzodiazepine receptor antagonist flumazenil into the supramammillary nucleus (mSuM) or the posterior hypothalamic nucleus (PH) antagonised CDP in both cases. Flumazenil was similarly effective in mSuM and PH, suggesting that these nu

Acetazolamide and amiloride inhibit pentobarbital-induced facilitation of nocifensive reflexes.

BACKGROUND: Neuronal excitation may result from stimulation of gamma-aminobutyric acid A (GABA(A)) receptors that prolong the channel opening, depolarizing the postsynaptic membrane. Drugs such as acetazolamide or amiloride can block GABA depolarization. Barbiturates facilitate nociceptive reflexes and also prolong the GABA(A) channel open-time. To evaluate the possible mechanism, the authors studied the impact of acetazolamide and amiloride on pentobarbital-induced nocifensive reflex facilitation. Because nitric oxide (NO) is a mediator of reflex facilitation, the authors evaluated the effect

[The effects of flumazenil or bicuculline on the respiratory depression by morphine].

The effects of intravenous administration of flumazenil (n = 6) or bicuculline (n = 6) on the discharge of the phrenic nerve were studied following vagotomy in pentobarbital anesthetized mechanically ventilated rats. Morphine (0.4 mg.kg-1.min-1) was administrated until the respiratory rate decreased to about a half of the baseline respiratory rate. In this state, we first administered flumazenil (0.25 mg.kg-1) or bicuculline (0.4 mg.kg-1), intravenously and then administered naloxone (0.02 mg) intravenously in the two groups. The increase of inspiratory time from 0.7 +/- 0.1 to 2.0 +/- 0.5 s b

The GABAA receptor: new insights from single-channel recording.

The GABAA receptor of mammalian neurons exists as a macromolecular complex incorporating several interacting components. These include a chloride-permeable ion channel and a recognition site for GABA. The binding of GABA molecules at the latter site triggers the transient opening of the chloride ion channel, resulting in a flow of charge which inhibits the generation of action potentials in the cell. The precise amount of charge passed during this event is modulated by ligand binding at at least three regulatory sites. These sites act as receptors for barbiturates, benzodiazepines, and certain

From bugs to brain: unravelling the GABA signalling networks in the brain-gut-microbiome axis.

Convergent data across species paint a compelling picture of the critical role of the gut and its resident microbiota in several brain functions and disorders. The chemicals mediating communication along these sophisticated highways of the brain-gut-microbiome (BGM) axis include both microbiota metabolites and classical neurotransmitters. Amongst the latter, GABA is fundamental to brain function, mediating most neuronal inhibition. Until recently, GABA's role and specific molecular targets in the periphery within the BGM axis had received limited attention. Yet, GABA is produced by neuronal an

Hops compounds modulatory effects and 6-prenylnaringenin dual mode of action on GABAA receptors.

Hops (Humulus lupulus L.), a major component of beer, contain potentially neuroactive compounds that made it useful in traditional medicine as a sleeping aid. The present study aims to investigate the individual components in hops acting as allosteric modulators in GABAA receptors and bring further insight into the mode of action behind the sedative properties of hops. GABA-potentiating effects were measured using [3H]ethynylbicycloorthobenzoate (EBOB) radioligand binding assay in native GABAA receptors. Flumazenil sensitivity of GABA-potentiating effects, [3H]Ro 15-4513, and [3H]flunitrazepam

DMCM, a benzodiazepine site inverse agonist, improves active avoidance and motivation in the rat.

There are several modulatory sites at GABA(A) receptors, which mediate the actions of many drugs, among them benzodiazepine. Three kinds of allosteric modulators act through the benzodiazepine binding site: positive (agonist), neutral (antagonist), and negative (inverse agonist). The goal of the present study was to examine the influence of the inverse agonist methyl 6,7-dimethoxy-4-ethyl-beta-carboline-3-carboxylate (DMCM) acting on α GABA(A) receptor and compare its dose-response effects on memory and depression-like behavior. We independently studied the effects of DMCM (0.05-1.0 mg/kg) on

Antihyperalgesic effect of the GABA(A) ligand clobazam in a neuropathic pain model in mice: a pharmacokinetic-pharmacodynamic study.

Facilitation of spinal GABAergic inhibition with benzodiazepines (BZDs) reverses pain sensitization in animals; however, the use of BZDs in man is limited by their sedative effect. The antihyperalgesic effects of GABA(A) agonists are mediated by GABA(A) receptors containing α2 subunits, whereas sedation is linked to α1 subunit-containing receptors. α2 and α3 selective GABA(A) receptor modulators have been tested in animals but are not yet available for use in human beings. Clobazam is a 1,5-BZD, which exhibits less cognitive side effects than other benzodiazepines. Here, we studied its antihyp

[How do antiepileptic drugs work?].

BACKGROUND: There are currently around 25 antiepileptic drugs in use in Norway, of which 15 have entered the market in the last 20 years. All have somewhat different effect- and adverse effect profiles and mechanisms of action. Here we present a brief overview of current knowledge regarding the basic mechanisms of action of these drugs. METHOD: The review is based on a discretionary selection of relevant articles found through a literature search in PubMed and our own clinical and research experience. RESULTS: There are, roughly speaking, four main mechanisms; 1) modulation of ion channels (

Effects of triazolam and diazepam on learning and memory as assessed using a water maze.

One of the reported adverse side effects of the frequently prescribed benzodiazepines diazepam (Valium) and triazolam (Halcion) is an impairment of anterograde memory in humans. The experiments described in this article compared the effects of triazolam and diazepam on performance in a water maze task that is sensitive to drugs that affect learning and memory. The water maze utilized is a traditional type of maze with alleyways and door choices, unlike the Morris open water maze. Time required to find an out-of-the-water platform and errors committed during the swim are used as performance mea

GABA-mediated regulation of lipid metabolism: a conserved relation with diverse physiological significances.

Gamma-aminobutyric acid (GABA) is a four-carbon non-proteinogenic amino acid that is primarily known for its role as an inhibitory neurotransmitter in mammals. It is synthesized from glutamate by the enzyme L-glutamic acid decarboxylase and metabolized via the GABA shunt pathways. GABA is known to influence various metabolic processes, and recent studies have shown its influence on lipid metabolism. This review explores the distinct relationship between GABA and lipid metabolism by highlighting their role in physiological processes in animals, plants, and microalgae. In animals, GABA acts as a

Allosteric modulation of GABAA receptors in acutely dissociated neurons of the suprachiasmatic nucleus.

The gamma-aminobutyric acid (GABA)-induced response was investigated in acutely dissociated suprachiasmatic nucleus (SCN) neurons of 11- to 14-day-old rats, under the voltage-clamp condition of nystatin-perforated patch recording. At a holding potential of -40 mV, application of GABA induced inward currents in a concentration-dependent manner. Pentobarbital and 5 beta-pregnan-3 alpha-ol-20-one (pregnanolone) similarly induced inward currents. GABA-induced inward currents were suppressed in a concentration-dependent manner by pretreating neurons with a GABAA receptor antagonist, bicuculline. Bi

Glial-neuronal crosstalk via GABA signaling: mechanistic insights into neuropsychiatric and neurological pathophysiology.

Glial cells-including astrocytes, microglia, oligodendrocytes (OLs), and oligodendrocyte progenitor cells (OPCs)- are essential for maintaining central nervous system (CNS) homeostasis, shaping neural circuits, supporting neuronal function, and regulating immune responses and myelination. These cells engage in dynamic and reciprocal interactions with neurons, which are frequently disrupted under pathological conditions. Emerging evidence identifies γ-aminobutyric acid (GABA), classically known as the principal inhibitory neurotransmitter, as a key mediator of glia-neuron communication. Glial c

GABA(A) receptor diversity and pharmacology.

Because of its control of spike-timing and oscillatory network activity, gamma-aminobutyric acid (GABA)-ergic inhibition is a key element in the central regulation of somatic and mental functions. The recognition of GABA(A) receptor diversity has provided molecular tags for the analysis of distinct neuronal networks in the control of specific pharmacological and physiological brain functions. Neurons expressing alpha(1)GABA(A) receptors have been found to mediate sedation, whereas those expressing alpha(2)GABA(A) receptors mediate anxiolysis. Furthermore, associative temporal and spatial memor

Measuring T2 in vivo with J-difference editing: application to GABA at 3 Tesla.

PURPOSE: To develop an experimental approach for determining in vivo transverse relaxation rates (T(2)) of metabolites that are detected by spectral editing without using simulations, and to demonstrate this approach to measure the T(2) of γ-aminobutyric acid (GABA). MATERIALS AND METHODS: The proposed method first determines the TE-dependence of the edited signals using measurements in a pure phantom solution (10 mM γ-aminobutyric acid; GABA); the phantom T(2) is also determined. Once the editing echo time (TE)-modulation pattern is known, it can then be used to determine T(2) in vivo. The m

Ro 15-4513 binding to GABAA receptors: subunit composition determines ligand efficacy.

The bidirectional modulation of ligand binding to benzodiazepine receptors (BzR) by GABA (the "GABA shift") has been widely used to predict ligand efficacy. The present study examined the effects of GABA and muscimol on [3H]Ro 15-4513 binding to "diazepam-insensitive" (DI) and "diazepam-sensitive" (DS) BzR. Neither GABA nor muscimol significantly altered [3H]Ro 15-4513 binding to DI in cerebellum, while both compounds inhibit [3H]Ro 15-4513 binding to cerebellar DS in a concentration-dependent fashion. The maximum reductions in [3H]Ro 15-4513 binding to cerebral cortical and hippocampal membra
