Maternal high sugar and fat diet benefits offspring brain function via targeting on the gut-brain axis.

A recent study showed that a gestational high fat diet protects 3xTg-AD offspring from memory impairments, synaptic dysfunction, and brain p … e HSHF diet offspring, there were influenced on various types of neurons, including cholinergic and GABAergic neurons, on autophagy levels in the brain, and on inflammation levels in the intestinal tract. When the offspring grew older (16 months), we found that some genes of benefit against nervous system disease were activated, such as Lhx8, GPR88, RGS9, CD4, DRD2 …

The Bed Nucleus of the Stria Terminalis-Deep Mesencephalic Nucleus Circuit Linking Emotion and Wakefulness.

The bed nucleus of the stria terminalis (BNST), a part of the extended amygdala, integrates emotional and arousal-related signals. While GABAergic BNST (GABABNST) neurons have been implicated in promoting transitions from non-rapid eye movement (NREM) sleep to wakefulness, their downstream mechanisms remain unclear. Here, we identify a neuronal circuit through which GABABNST neurons promote arousal via projections to a midbrain region known as the deep mesencephalic nucleus (DpMe), located within the broader mesencephalic reticular formation. In male mice, we used a combination of optogenetics

Upper gastrointestinal dysmotility after spinal cord injury: is diminished vagal sensory processing one culprit?

Despite the widely recognized prevalence of gastric, colonic, and anorectal dysfunction after spinal cord injury (SCI), significant knowledge gaps persist regarding the mechanisms leading to post-SCI gastrointestinal (GI) impairments. Briefly, the regulation of GI function is governed by a mix of parasympathetic, sympathetic, and enteric neurocircuitry. Unlike the intestines, the stomach is dominated by parasympathetic (vagal) control whereby gastric sensory information is transmitted via the afferent vagus nerve to neurons of the nucleus tractus solitarius (NTS). The NTS integrates this senso

The effect of bicuculline application on auditory response properties of inferior collicular neurons of mice with or without monaural middle ear destruction in early age.

Previous studies have demonstrated that abnormal auditory stimulation during early postnatal development can be manifested through physiological changes that occur in the inferior colliculus (IC) of mammals. To determine the contribution of the GABAergic transmitter systems to the development of response properties of IC neurons, we examined the effect of application of bicuculline (which is an antagonist for the GABA(A) receptors) on response properties of IC neurons of the laboratory mice, Mus musculus, with or without early monaural middle ear destruction. Monaural middle ear destruction wa

Prenatal Ethanol Exposure Results in Cell Type, Age, and Sex-Dependent Differences in the Neonatal Striatum That Coincide with Early Motor Deficits.

Delayed motor development is an early clinical sign of fetal alcohol spectrum disorders. However, changes at the neural circuit level that underlie early motor differences are underexplored. The striatum, the principal input nucleus of the basal ganglia, plays an important role in motor learning in adult animals, and the maturation of the striatal circuit has been associated with the development of early motor behaviors. Here, we briefly exposed pregnant C57BL/6 dams to ethanol (5% w/w) in a liquid diet on embryonic days 13.5-16.5 and assessed the mouse progeny using a series of nine brief mot

Heterogeneous subpopulations of GABAAR-responding neurons coexist across neuronal network scales and developmental stages in health and disease.

Gamma-aminobutyric acid (GABA) is the main inhibitory neurotransmitter in adults. Depolarizing GABA responses have been well characterized at neuronal-population average level during typical neurodevelopment and partially in brain disorders. However, no investigation has specifically assessed whether a mosaicism of cells with either depolarizing or hyperpolarizing/inhibitory GABAergic responses exists in animals in health/disease at diverse developmental stages, including adulthood. Here, we showed that such mosaicism is present in wild-type (WT) and down syndrome (DS) neuronal networks, as as

Motor learning deficits in aged rats are correlated with loss of cerebellar noradrenergic function.

We have demonstrated that aged rats show impairments in learning patterned motor movements. Similar behavioral impairment is observed in rats with noradrenergic lesions. Norepinephrine is known to act as a neuromodulator in the cerebellar cortex because it can augment the action of GABA and other neurotransmitters. This effect of NE to augment the signal to noise ratio of GABAergic inputs to cerebellar Purkinje neurons is a possible substrate for NE's effect on motor learning. Aged rats demonstrate deficits in the modulatory actions of NE to augment GABAergic inhibitions when both substances a

Characterisation of hyperpolarization-activated currents (I(h)) in the medial septum/diagonal band complex in the mouse.

Hyperpolarization-activated cyclic nucleotide gated (HCN) channel subunits are distributed widely, but selectively, in the central nervous system, and underlie hyperpolarization-activated currents (I(h)) that contribute to rhythmicity in a variety of neurons. This study investigates, using current and voltage-clamp techniques in brain slices from young mice, the properties of I(h) currents in medial septum/diagonal band (MS/DB) neurons. Subsets of neurons in this complex, including GABAergic and cholinergic neurons, innervate the hippocampal formation, and play a role in modulating hippocampal

Mechanisms and Cell Type-Dependence of Non-Hebbian Long-Term Potentiation at Primary Afferent Synapses in the Mouse Spinal Superficial Dorsal Horn.

Sensory synapses onto ascending spinal projection neurons in the superficial dorsal horn (SDH) can exhibit non-Hebbian plasticity, such as when the firing of primary afferent neurons alone is sufficient to induce long-term potentiation (LTP), following neonatal tissue damage or D1/D5 dopamine receptor activation. However, whether non-Hebbian LTP can be induced at sensory synapses in the absence of tissue injury or exogenous neuromodulatory signaling, the molecular mechanisms underlying this form of synaptic plasticity, and the degree to which non-Hebbian LTP occurs in a cell type-dependent man

Microglial activation drives neuronal dysregulation in alcohol-induced seizure susceptibility.

Excessive alcohol drinking can induce seizures, but the mechanisms underlying acute alcohol-induced seizures remained to be extensively investigated. To investigate this, we established an acute alcohol-treated model and observed a microglial response in the hippocampal CA1 region of mice, which was associated with the enhancement of seizure susceptibility. Furthermore, we found an increased abundance of GABAergic interneurons and a decrease in the activity of CaMKII in the hippocampal CA1 region. Minocycline-mediated microglial depletion fully inhibited the increase in GABAergic interneurons

Ethanol and the benzodiazepine-GABA receptor-ionophore complex.

Ethanol has a pharmacological profile similar to that of classes of drugs like benzodiazepines and barbiturates, which enhance GABAergic transmission in the mammalian CNS. Several lines of behavioral, electrophysiological and biochemical studies suggest that ethanol may bring about most of its effects by enhancing GABAergic transmission. Recently, ethanol at relevant pharmacological concentrations has been shown to enhance GABA-induced 36Cl-fluxes in cultured spinal cord neurons, synaptoneurosomes and microsacs. These enhancing effects of ethanol were blocked by GABA antagonists. Ro15-4513, an

Interactions between Brainstem Neurons That Regulate the Motility to the Stomach.

Activity in the dorsal vagal complex (DVC) is essential to gastric motility regulation. We and others have previously shown that this activity is greatly influenced by local GABAergic signaling, primarily because of somatostatin (SST)-expressing GABAergic neurons. To further understand the network dynamics associated with gastric motility control in the DVC, we focused on another neuron prominently distributed in this complex, neuropeptide-Y (NPY) neurons. However, the effect of these neurons on gastric motility remains unknown. Here, we investigate the anatomic and functional characteristics …

Chronic administration of DL-allyl-glycine into the neostriatum, disorganises the firing modes of the nigral dopaminergic neurons in the rat.

Nigral dopaminergic (DA) neurons have been reported to fire according to three modes: very regular (pacemaker 42%) irregular (random 46%) and bursty (12%). The switch from simple spiking mode (pacemaker or random) to bursty firing would correspond to an increase in DA release necessary for the performance of a new motor act. As nigral DA cells are impinged upon by a high percentage of GABAergic afferents we blocked striatal GABAergic output neurons by chronic administration into the neostriatum of allyl-glycine, a glutamic acid decarboxylase (GAD) inhibitor. After treatment, rats presented hyp

Preictal dysfunctions of inhibitory interneurons paradoxically lead to their rebound hyperactivity and to low-voltage-fast onset seizures in Dravet syndrome.

Epilepsies have numerous specific mechanisms. The understanding of neural dynamics leading to seizures is important for disclosing pathological mechanisms and developing therapeutic approaches. We investigated electrographic activities and neural dynamics leading to convulsive seizures in patients and mouse models of Dravet syndrome (DS), a developmental and epileptic encephalopathy in which hypoexcitability of GABAergic neurons is considered to be the main dysfunction. We analyzed EEGs from DS patients carrying a SCN1A pathogenic variant, as well as epidural electrocorticograms, hippocampal l…

Vagal GABAergic signaling in autonomic control of cardiometabolic function.

Gamma-aminobutyric acid (GABA) and its receptors play a critical role in maintaining the balance between excitatory and inhibitory neurotransmission in the central nervous system, including autonomic regulatory regions that control cardiometabolic function. Vagal circuits facilitate bidirectional communication between peripheral organs and the brain to elicit autonomic reflexes and modulate complex adaptive behavior to preserve cardiometabolic homeostasis. Dampened vagal activity can lead to the development of cardiometabolic diseases. Emerging evidence over the past decade identifies inhibito

Intracerebral Transplantation of BDNF-overexpressing Human Neural Stem Cells (HB1.F3.BDNF) Promotes Migration, Differentiation and Functional Recovery in a Rodent Model of Huntington's Disease.

Huntington's disease (HD) is a dominantly inherited neurodegenerative disorder caused by abnormally expanded CAG repeats in the huntingtin gene. The huntingtin gene mutation leads to the progressive degeneration of striatal GABAergic medium spiny neurons (MSN) and reduces the level of brain-derived neurotrophic factor (BDNF) in HD patient's brain. BDNF is an essential neurotrophic factor for the cortico-striatal synaptic activity and the survival of GABAergic neurons. In this study, we transplanted BDNF-overexpressing human neural stem cells (HB1.F3.BDNF) into the contra-lateral side of unilat…

Dysfunction of Orbitofrontal GABAergic Interneurons Leads to Impaired Reversal Learning in a Mouse Model of Obsessive-Compulsive Disorder.

Cognitive inflexibility is a cardinal symptom of obsessive-compulsive disorder (OCD) and often manifests as impaired reversal learning. Abnormal recruitment of the orbitofrontal cortex (OFC)-striatal circuit is implicated in reversal learning deficits in patients with OCD. However, the precise circuitry mechanism underlying normal and impaired reversal learning remains elusive. Using fiber photometry and optogenetics, we demonstrated cell-type-specific activity dynamics in the OFC-striatal circuit underlying normal reversal learning and cell-type-specific dysfunctions that causally lead to imp
