NPR1 Promotes Its Own and Target Gene Expression in Plant Defense by Recruiting CDK8.

NPR1 (NONEXPRESSER OF PR GENES1) functions as a master regulator of the plant hormone salicylic acid (SA) signaling and plays an essential role in plant immunity. In the nucleus, NPR1 interacts with transcription factors to induce the expression of PR (PATHOGENESIS-RELATED) genes and thereby promote defense responses. However, the underlying molecular mechanism of PR gene activation is poorly understood. Furthermore, despite the importance of NPR1 in plant immunity, the regulation of NPR1 expression has not been extensively studied. Here, we show that SA promotes the interaction of NPR1 with b

Plant-specific regulation of replication protein A2 (OsRPA2) from rice during the cell cycle and in response to ultraviolet light exposure.

DNA replication is a process that is highly conserved among eukaryotes. Nonetheless, little is known about the proteins involved in it in plants. Replication protein A (RPA) is a heterotrimeric, single-stranded DNA-binding protein with several functions in DNA metabolism in humans and yeast and supposedly also in plants. Here we report on the regulation of OsRPA2, the 32-kDa subunit of RPA from rice ( Oryza sativa L.). We found conserved regulation mechanisms at the level of gene expression between animal and plant RPA2 genes and distinct features of OsRPA2 regulation at the level of protein e

Virus-induced gene silencing is an effective tool for assaying gene function in the basal eudicot species Papaver somniferum (opium poppy).

Virus-induced gene silencing (VIGS) is an attractive method for assaying gene function in species that are resistant to conventional genetic approaches. However, VIGS has been shown to be effective in only a few, closely related plant species. Tobacco rattle virus (TRV), a bipartite RNA virus, has a wide host range and so in principle could serve as an efficient vector for VIGS in a diverse array of plant species. Here we show that a vector based on TRV sequences is effective at silencing the endogenous phytoene desaturase (PapsPDS) gene in Papaver somniferum (opium poppy). We show that this v

Genome-wide identification and expression analysis of dirigent-jacalin genes from plant chimeric lectins in Moso bamboo (Phyllostachys edulis).

Dirigent-jacalin (D-J) genes belong to the plant chimeric lectin family, and play vital roles in plant growth and resistance to abiotic and biotic stresses. To explore the functions of the D-J family in the growth and development of Moso bamboo (Phyllostachys edulis), their physicochemical properties, phylogenetic relationships, gene and protein structures, and expression patterns were analyzed in detail. Four putative PeD-J genes were identified in the Moso bamboo genome, and microsynteny and phylogenetic analyses indicated that they represent a new branch in the evolution of plant lectins. P

SPOTTED-LEAF7 targets the gene encoding β-galactosidase9, which functions in rice growth and stress responses.

β-Galactosidases (Bgals) remove terminal β-D-galactosyl residues from the nonreducing ends of β-D-galactosidases and oligosaccharides. Bgals are present in bacteria, fungi, animals, and plants and have various functions. Despite the many studies on the evolution of BGALs in plants, their functions remain obscure. Here, we identified rice (Oryza sativa) β-galactosidase9 (OsBGAL9) as a direct target of the heat stress-induced transcription factor SPOTTED-LEAF7 (OsSPL7), as demonstrated by protoplast transactivation analysis and yeast 1-hybrid and electrophoretic mobility shift assays. A wheat-specific proline-rich protein regulates grain weight through a ubiquitin-mediated delay of endosperm cellularization.

Grain weight is a key determinant of wheat yield, yet the developmental mechanisms that define this trait remain poorly understood. Here, we identify TaPGW1-A1, a wheat-specific proline-rich gene that regulates grain weight. TaPGW1-A1 is a recently evolved gene found only in wheat. It is highly expressed in early developing grains, and its overexpression delays endosperm cellularization and prolongs the syncytial phase, resulting in an 8% increase in field-based grain weight. TaPGW1-A1 protein directly interacts with the ubiquitin receptor TaDSK2a (Dominant Suppressor of KAR2 homolog) via its

Meiosis-specific protein co-expression in Arabidopsis thaliana: a protein delivery tool in meiotic cells.

Meiosis is a key process in sexual reproduction, orchestrated by numerous proteins that differ in their spatiotemporal expression level and dynamics. Employing meiotic gene promoters typically leads to varying levels of transgene expression within meiotic and non-meiotic cells in plants. Hence, their application for protein expression during meiosis is limited. Here, we present a tool for meiosis-specific protein co-expression in Arabidopsis thaliana. By utilizing ribosome-skipping peptides, we leverage the distinct expression patterns of the two meiotic genes ASY1 and MTOPVIB to co-express va

ZmSSRP1-mediated chromatin accessibility modulates maize development via regulation of ZmBRD1 transcription.

The FAcilitates Chromatin Transcription (FACT) complex, comprising structure-specific recognition protein 1 (SSRP1) and suppressor of Ty 16 (SPT16), plays a key role in regulating gene transcription by facilitating ATP-independent nucleosome assembly. SSRP1 contains an HMG domain that binds to DNA and regulates cell proliferation, apoptosis, and DNA repair. However, its role in plant development remains poorly understood. In this study, we identified a mutant, small kernel 301 (smk301), characterized by reduced kernel size and semi-dwarfism. Map-based cloning revealed that the mutation affects

Contrasting redox-related physiological responses associated with HaGATA23 and HaGATA36 during Orobanche cumana parasitism in sunflower (Helianthus annuus L.).

Helianthus annuus is an economically important Asteraceae species used for seed oil production and ornamental purposes, but its production is seriously affected by the root-parasitic plant Orobanche cumana. GATA transcription factors are zinc-finger DNA-binding regulators involved in plant development and stress adaptation. However, the molecular characteristics of GATA transcription factors in Helianthus annuus and their contribution to Helianthus annuus -Orobanche cumana interaction remain poorly understood. Here, 36 HaGATA members were retrieved from the Helianthus annuus genome and classif

Molecular insights and translational opportunities to enhance heat tolerance in rice.

Heat stress is an increasingly serious threat to rice (Oryza sativa L.) productivity, yet the genetic and regulatory architecture underlying thermotolerance remain poorly resolved and fragmented across studies. Earlier research focused on individual pathways or specific developmental stages; however, recent advances now support an integrated understanding of heat stress adaptation in rice. This review synthesizes emerging insights into molecular physiology, regulatory signaling, epigenetic memory, and genome-scale variation associated with thermotolerance. We highlight the interconnected roles

Cell-specific-resolved coordination of mitochondrial metabolism and DNA damage responses underpins aluminium responses in plant roots.

Aluminium (Al) toxicity is a major constraint on crop productivity in acidic soils (pH < 5.5), severely impairing root growth and function. While organic acid (OA)-mediated chelation and DNA damage response (DDR)-mediated control of cell division are recognized as key tolerance mechanisms, their integration at the cellular level remains poorly understood. Here, we employed Translating Ribosome Affinity Purification (TRAP) to achieve cell-type-specific gene expression profiling of Arabidopsis thaliana roots, enabling the dissection of molecular responses in whole roots, the quiescent centre, an

Chilling Accumulation Shapes Transcriptomic Responses to Warming Exposure During Grapevine Deacclimation.

During winter, grapevine (Vitis vinifera) bud dormancy and cold hardiness are regulated by complex interactions between chilling accumulation and warm temperature cues. However, the molecular mechanisms underlying physiological transitions during winter remain poorly understood. In this study, we performed time-series RNA-seq on "Cabernet Sauvignon" dormant buds with varying chilling accumulation, followed by warm temperature exposure under a controlled forcing condition. Using weighted gene co-expression network analysis, empirical modeling, and a novel calculation of molecular warm temperatu

Two LEAFY homologs regulate floral patterning and development without affecting flowering time in kiwifruit.

Actinidia chinensis (kiwifruit) are economically important fruit-bearing woody perennial vines but with a long juvenile phase, which slows plant breeding efforts. LEAFY (LFY) encodes a plant-specific transcription factor central to the reproductive pathway in many plants and its overexpression can accelerate flowering, including in some trees. We identified two kiwifruit LFY genes that are expressed in actively growing apical buds. Overexpression of AcLFY1 or AcLFY2 promoted flowering in Arabidopsis, but did not accelerate flowering in kiwifruit. Instead, branching at the lower nodes and chang

Mechanisms and Advances in Plant Lipid Regulatory Responses Under Biotic and Abiotic Stress.

Biotic stresses (pest feeding, pathogenic fungal/bacterial/viral infection) and diverse abiotic stresses (extreme temperature, drought, waterlogging, saline-alkali soil, heavy metal pollution, nutrient deficiency, UV-B, ozone) severely restrict crop growth and global agricultural yield. Lipids act as core membrane structural constituents and vital secondary signaling messengers, executing multi-layered adaptive balancing functions during cell-type interactive stress acclimation, rather than uniform whole-plant lipid responses. They sustain membrane structural integrity across distinct cell pop

Mutations of the brassinosteroid biosynthesis gene HvDWARF5 enable balance between semi-dwarfism and maintenance of grain size in barley.

Brassinosteroids (BRs) are phytohormones which regulate various developmental processes in plants. They are exceptional phytohormones, as they do not undergo long-distance transport between plant organs. However, knowledge about the function of the enzymes that catalyse BR biosynthesis (particularly its early stages) in cereal crops remains limited. Therefore, this study identifies and analyses the function of the HvDWARF5 (HvDWF5) gene, involved in the early stage of BR biosynthesis in barley (Hordeum vulgare), an important cereal crop, using the TILLING (Targeting Induced Local Lesions IN Ge

OsCK2 Phosphorylates OsSIZ1 to Regulate Phosphate Starvation Response and Defence Response in Rice.

Phosphorus is a crucial nutrient for the normal growth and development of rice. Enhancing the efficiency of phosphorus acquisition in plants is vital for optimising soil phosphorus utilisation. The SUMO E3 ligase OsSIZ1 is an important factor for regulating plant phosphorus acquisition and starvation response in rice. However, how its activity responds to Pi-supplied conditions remains unknown. In this study, we discovered that the mutations in OsCK2β3, which encodes a regulatory subunit of CK2, influenced the Pi starvation response. The global level of SUMOylation was found to decrease in Osc

Persulfidation of transcription factor MYB10 inhibits anthocyanin synthesis in red-skinned pear.

Hydrogen sulfide (H2S) is a gaseous signaling molecule that delays color change during fruit ripening. Whether H2S affects anthocyanin biosynthesis in red-skinned pears (Pyrus L.) remains unclear. Here, we found that H2S substantially inhibits anthocyanin accumulation in red-skinned pears and the expression of several genes encoding transcription factors is affected in response to H2S signaling. For example, PyMYB10 and PyMYB73 were down-regulated, whereas PyMYB114 and PyMYB6 were up-regulated. Bioinformatics analysis showed that PyMYB73 and PyMYB6, each containing an EAR motif, may negatively
