Scale insects (Hemiptera: Coccomorpha) are important sap-feeding herbivores that influence plant physiology and contribute to forest ecosystem functioning. Despite their ecological importance, their host specificity and infestation dynamics remain insufficiently documented in Mediterranean semi-arid forests. This study investigated the distribution and ecological specialization of scale insects in Belezma National Park (Algeria), focusing on host tree species, plant communities, and infested plant parts. Significant variations (p = 0.006) in infestation rates were observed among host tree spec…

BACKGROUND: Maternally inherited symbionts can impact the reproduction of their host insects in various ways. The ovary is critical for the successful reproduction of female insects. Such symbionts have strong ovary tropism. Intracellular symbionts depend on host cells for replication and transmission. However, the cellular mechanisms by which symbionts impact insect reproduction through affecting ovary development have seldom been investigated. Our previous work has shown that the facultative symbiont Rickettsia can be vertically transmitted through eggs in the whitefly Bemisia tabaci and Ric

Adult spotted lanternflies (Lycorma delicatula) were differentially susceptible to Beauveria bassiana when inoculated with conidia on the distal ventral abdomen versus distal forewings. More adults inoculated on the abdomens died of B. bassiana infections than those inoculated on the wings. Abdominal inoculants also died more quickly than wing inoculants. Due to the large dorsal forewings of these planthoppers, typically covering abdomen and thorax, we suggest that the wings can at least partially protect from dorsal sprays of an infectious Hypocreales; we hypothesize that spraying surfaces on

BACKGROUND: Entomopathogenic fungi (EPF) develop antagonistic strategies to compete with bacteria on the host cuticle surface, facilitating fungal invasion of their host. However, the presence of fungal species on host cuticles and the mechanisms by which EPF compete against cuticular fungi remain unclear.
RESULTS: Beauveria bassiana is a ubiquitous entomopathogenic fungus with significant potential for biological control of the brown planthopper Nilaparvata lugens. In this study, we demonstrated that three yeast species are dominant on the cuticle surface of N. lugens: Kurtzmaniella quercitr

Begomoviruses are a group of single stranded DNA plant viruses exclusively transmitted by the sweet potato whitefly Bemisia tabaci in a persistent, circulative manner. After acquisition from plant phloem, this group of viruses circulate and are retained within the whitefly, interacting with tissues, cells and molecular pathways for maintaining the safety of the infective intact virions, by exploiting cellular mechanisms and avoiding degradation by the insect immune responses. During retention, the virions are internalized in the midgut cells, exit and spend hours-days in the hemolymph and cros

The lack of sequence similarity among insect effectors necessitates the independent identification of effectors in the Asian citrus psyllid Diaphorina citri, the primary vector of huanglongbing pathogens. In this study, nine candidate effectors were identified from the D. citri salivary gland transcriptome. Among them, DcE13 exhibited the highest FPKM value and was delivered into plants during feeding. Transient expression in tobacco leaves showed that DcE13 is localized in the plasma membrane and around the nuclear envelope of plant cells. It inhibits BAX- and INF1-induced hypersensitive resp

BACKGROUND: Sogatella furcifera, commonly known as white-backed planthopper, is a highly threatening migratory pest that feeds on rice and can transmit phytoviruses. Recently, mesoionic insecticides, exemplified by triflumezopyrim (TFM), have proven effective in managing such pests. Nevertheless, available data indicate that TFM remains highly toxic to ecologically important pollinators. Herein, the development of a class of pyrido[1,2-a]pyrimidine-based cinnamic acid analogs, their insecticidal activity, modes of action, and toxicity to bees are disclosed.
RESULTS: A total of 37 novel pyrido

BACKGROUND: Diaphorina citri is one of the most severe pests affecting citrus production worldwide. Our previous research identified a series of quassinoids that exhibit insecticidal activity against D. citri adults. However, the larvae of D. citri also pose a significant threat to the citrus industry. In an effort to discover novel larvicidal lead compounds, a systematic phytochemical study on the relatively understudied fruits of Picrasma quassioides was conducted, and the insecticidal mechanism was also explored preliminarily.
RESULTS: Ten new (1-10) and 15 known quassinoids (11-25) were i

The leafhopper genus Arboridia includes several species that feed on Vitis vinifera and cause leaf chlorosis. We report the first alien Arboridia infestation in Italy in 2021 in an Apulian vineyard. To confirm the taxonomic status of the species responsible for crop damage, and reconstruct its demographic history, we barcoded individuals from Apulia together with Arboridia spp. from Crete (Greece), A. adanae from Central Turkey and other specimens of the presumed sister species, A. dalmatina from Dalmatia (Croatia). Molecular phylogenies and barcoding gap analysis identified clades not associa

To perform its propagative and circulative cycle into its insect vector, the flavescence dorée phytoplasma invades different cell types. Clathrin-mediated endocytosis is used by a wide range of bacteria to infect eukaryote cells. Among the insect proteins interacting with the phytoplasma adhesin VmpA, we identified the adaptor protein complex AP-1 and AP-2 suggesting that phytoplasmas could enter the insect cells via clathrin-mediated endocytosis. By infection assays of insect cells in culture, we showed that phytoplasmas entry into Drosophila S2 cells was more efficient than infection of the

Many plant viruses are transmitted by insect vectors in a circulative manner. For efficient transmission, the entry of the virus from vector hemolymph into the primary salivary gland (PSG) is a step of paramount importance. Yet, vector components mediating virus entry into PSG remain barely characterized. Here, we demonstrate the role of clathrin-mediated endocytosis and early endosomes in begomovirus entry into whitefly PSG. Our findings unravel the key components involved in begomovirus transport within the whitefly body and transmission by their whitefly vectors and provide novel clues for

BACKGROUND: Parasitoids are natural enemies that can modify their behaviors as they search for hosts based on the characteristics of the sites in which they forage. Theoretical models predict that a parasitoid will stay for longer periods in high-quality sites or patches than in those of low quality. In turn, patch quality may be linked to factors such as the number of hosts and the risk of predation. In the present study, we sought to determine if the factors of the number of hosts, risk of predation, and their interaction influence the foraging behavior of the parasitoid Eretmocerus eremicus

Herbivore-induced defense responses are often specific, whereas plants could induce distinct defense responses corresponding to infestation by different herbivorous insects. Brown plant hopper (BPH) Nilaparvata lugens, a phloem-feeding insect, and rice leaf folder (LF) Cnaphalocrocis medinalis, a chewing insect, are both specialist herbivores on rice. To characterize the distinct resistance primed by prior damage to these two specialist herbivores, we challenged rice plants with two herbivores during vegetative growth of parent plants and assessed plant resistance in subsequent ratoons. Here,

Insect predators are the most important natural enemies of the crapemyrtle bark scale (CMBS) in the USA. Mycopesticides (biocontrol fungi) are considered an IPM tool to increase CMBS mortality; however, their impacts on CMBS predators are unknown. The objectives of this study were to assess the abundance and diversity of CMBS natural enemies in Louisiana; evaluate the impacts of mycopesticides on survival of CMBS predators by life stage; and determine if entomopathogenic spores delivered to crapemyrtles are transferred to predators under field conditions. The mycopesticides Ancora® (Isaria fum

The hemlock woolly adelgid (Hemiptera: Adelgidae Adelges tsugae Annand) poses a serious threat to hemlocks in eastern North America, and ongoing research is focused on the identification and development of biological controls to protect and manage hemlock resources. Three predators native to the Pacific Northwest of North America that have been the focus of much research are Leucopis argenticollis (Zetterstedt), Leucopis piniperda (Malloch) (Diptera: Chamaemyiidae), and Laricobius nigrinus (Fender) (Coleoptera: Derodontidae). This study addresses the knowledge gap of adult Leucopis spp. emerge…

For molecular research, the quality and integrity of DNA obtained will affect the reliability of subsequent results. Extracting quality DNA from scale insects, including mealybugs, can be difficult due to their small body size and waxy coating. In this study, we evaluate eight commonly used DNA extraction methods to determine their efficacy in PCR analysis across life stages and preservation times. We find that fresh samples, immediately upon collection or after 2 wks, resulted in the most effective DNA extraction. Methods using the DNeasy Blood & Tissue kit, NaCl, SDS-RNase A, and SDS isolate

We studied the adult ambulatory response of the predator, Laricobius nigrinus Fender (Coleoptera: Derodontidae), to odors from its prey, Adelges tsugae Annand, the hemlock woolly adelgid, and foliage of hemlock woolly adelgid, host hemlocks (Tsuga spp.), and other conifers. Both the predator and hemlock woolly adelgid are apparently native to western North America, but the predator is being released in the eastern United States, which has different hemlock species, for biological control of a lineage of hemlock woolly adelgid inadvertently introduced from Japan. L. nigrinus responded to odors

The infection process and pathological changes of Japanese wax scale, Ceroplastes japonicus Green, by the hyphomycete Lecanicillium lecanii (Zimmermann) Gams & Zare were investigated by light, scanning and transmission electron microscopy. The results showed that L. lecanii generally infected the wax scale by penetrating the integument. The anal area, the body margin, around the base of mouthparts and legs, over the stigmatic furrow and the area around the vulva were susceptible places, while the wax test had an inhibitory effect on L. lecanii. Within 24h after inoculation, conidia became atta

BACKGROUND: Cashew (Anacardium occidentale L.) has become a very important non-traditional tree crop in Ghana. The crop is, however, attacked by sap-sucking insects, particularly the mosquito bug, Helopeltis schoutedeni Reuter, the leaf-footed bug, Pseudotheraptus devastans (Dist.), and the coreid bug, Anoplocnemis curvipes (F.), which feed on shoots, panicles and fruits. Their damage is characterised by withering of the latter. In Ghana, Oecophylla longinoda Latr. occurs in large numbers on cashew and other native plants, but little is known about its relationship with insect pests. The relat

Invasive, non-indigenous, phytophagous insects have caused widespread declines in several dominant tree species. The decline in dominant tree species may lead to cascading effects on other tree and microbial species and their interactions, affecting forest recovery following the decline. In the eastern USA, eastern hemlock (Tsuga canadensis (L.) Carr) is declining because of infestation by the hemlock woolly adelgid (HWA; Adelges tsugae Annand). Northern red oak (Quercus rubra L.) is a common replacement species in declining hemlock stands, but reduced mycorrhizal inoculum potential in infeste
