The effects of several heparin preparations on rat striatal neurokinin-1 receptor labelling were investigated. A low concentration of heparin dose-dependently stimulated [125I]Bolton-Hunter substance P ([125I]BH-SP) binding to rat striatal membranes. This effect was mainly due to an increase of the association velocity reflected by a decrease of the equilibrium dissociation rate constant. A higher heparin concentration inhibited specific [125I]BH-SP labelling by a negative allosteric mechanism as a result of an increase of the dissociation velocity.

The use of controlled local-release heparin was studied as a means to prevent thrombosis in microvascular surgery, using a rat arterial inversion graft as a model. Polyvinyl alcohol embedded with heparin was placed around the outside of inversion graft anastomoses in 16 animals. Subarachnoid hemorrhage (SAH) was produced in cats by transorbital rupture of the right middle cerebral artery (MCA). In untreated cats, widespread proliferative angiopathy occurred in both MCA's by 16 days after SAH. In cats that received systemic heparin, the pathological events following SAH were clearly reduced in the ruptured artery, and were not present in the contralateral left MCA. Heparin sodium-induced thrombosis is insidious and difficult to diagnose. If untreated, it results in death or major amputation. We have treated seven patients with thromboses resulting from platelet aggregation induced by heparin.
