In vitro and in vivo release of histamine by contrast media in the rat.

Some contrast media are able to release histamine from isolated mast cells of the rat and produce degranulation of these cells. Such properties are fairly correlated with the ability to increase vascular permeability in vivo and to produce a drop in blood pressure. If histamine is similarly being released in humans many untoward effects of these agents referred to clinically might be thus explained.

The role of histamine in narcolepsy type 1: a narrative review.

Narcolepsy type 1 (NT1) is a chronic neurological disorder characterized mainly by impaired wakefulness maintenance and dysregulated rapid eye movement (REM) sleep, which markedly affects patients' daytime functioning and quality of life. The core pathological basis of NT1 is the selective loss of orexinergic neurons. The orexinergic system regulates histaminergic neurons in the tuberomammillary nucleus (TMN) through anatomical projections and functional excitation, while the histaminergic system contributes to cortical activation, arousal maintenance, and attention regulation. Therefore, the …

Effects of histamine on the contractility of the rat distal cauda epididymis.

The motor activity of the epididymis duct is an essential process for male fertility and it is regulated by hormonal, neuronal and epithelial mechanisms. However, although there is evidence for the presence of histamine in the epididymis, its effects on epididymal motor activity are unknown. This study sought to evaluate the contractile effects of histamine on the rat distal cauda epididymis duct. Segments of the distal cauda epididymis duct from male Wistar rats were isolated and used in isolated organ bath experiments to evaluate the contractile effects of histamine in the absence or presenc…

Inhibition of hepatic apolipoprotein A-I gene expression by histamine.

In a recent high throughput analysis to identify drugs that alter hepatic apolipoprotein A-I (apo A-I) expression, histamine receptor one (H1) antagonists emerged as potential apo A-1 inducing drugs. Thus the present study was undertaken to identify some of the underlying molecular mechanisms of the effect of antihistaminic drugs on apo AI production. Apo A-I levels were measured by enzyme immunoassay and Western blots. Apo A-I mRNA levels were measured by reverse transcription real-time PCR using glyceraldehyde-3-phosphate dehydrogenase (GAPDH) mRNA as the internal control. The effects of his…

Elevation in histamine and tryptase following exercise in patients with mastocytosis.

Patients with mastocytosis exhibit an increase in serum levels of mast cell-related mediators with exercise and therefore should be guided regarding management of mast cell-associated symptoms to improve safety and quality of life.

Impact of oral vitamin C on histamine levels and seasickness.

BACKGROUND: Seasickness is a risk aboard a ship. Histamine is postulated as a causative agent, inversely related to the intake of vitamin C. Persons with mastocytosis experienced improvement of nausea after the intake of vitamin C. OBJECTIVE: To determine whether vitamin C suppresses nausea in 70 volunteers who spent 20 minutes in a life raft, exposed to one-meter-high waves in an indoor pool. METHOD: Double-blind placebo-controlled crossover study. Two grams of vitamin C or placebo was taken one hour before exposure. Blood samples were taken one hour before and after exposure to determine h…

Evidence for histamine as a new modulator of carotid body chemoreception.

It has been proposed that histamine is an excitatory transmitter between the glomus cells of the carotid body (CB) and the nerve endings of the petrosal ganglion (PG) neurons. The histamine biosynthetic pathway and the presence of histamine H1, H2 and H3 receptors have been reported in the CB. Thus, histamine meets some of the criteria to be regarded as a transmitter. However, there is no evidence that glomus cells contain histamine, or whether its application produces chemosensory excitation. Therefore, we studied its immunocytochemical localization on cat CB and its effects on chemosensory a…

Effects of meconium on airway reactivity to histamine and acetylcholine in vitro.

To better understand the mechanisms contributing to altered airway reactivity in the meconium aspiration syndrome, in this study we investigated whether there could be a meconium dose-dependent response of tracheal smooth muscle and lung tissue reactivity to histamine and acetylcholine in vitro. Tracheal and lung tissue strips from healthy guinea pigs were incubated for 1 hour in organ chambers with three different concentrations of meconium (1, 2, and 5 mg/ml) or in Krebs-Henseleit solution. Thereafter, the contractile responses to histamine and acetylcholine were recorded. Cumulative doses o…

Release and permeation of histamine in isolated caecum epithelia of pigs.

OBJECTIVE: The objective was to investigate the role of histamine in the porcine caecum with special regard to its release and permeation. MATERIAL AND METHODS: Caecal epithelia were mounted in Ussing chambers. Mast cells were stimulated by A23187 (1 micromol/l). Permeation and changes in short-circuit current (Isc) were assessed after unilateral application of 3H-labelled histamine (100 micromol/l). Mucosal-to-serosal (ms) and serosal- to-mucosal (sm) flux rates were calculated based on the contralateral appearance of radioactive histamine label (hist-rad; representing histamine plus catabol…
