The host interactome of influenza virus presents new potential targets for antiviral drugs.

Increasing antiviral drug resistance is a major concern for treating influenza, especially in a pandemic setting when the availability of a protective vaccine is uncertain. Resistance is often an issue with drugs directed at viral proteins and for small RNA viruses; there are also a limited number of viral proteins that are amenable to inhibition by a small molecule. A new approach that is gaining support is that cellular proteins, which facilitate virus replication, may be used as alternative targets. Whereas drugs directed at viral proteins tend to be virus-specific, drugs directed at host t

Host cell autophagy in immune response to zoonotic infections.

Autophagy is a fundamental homeostatic process in which cytoplasmic targets are sequestered within double-membraned autophagosomes and subsequently delivered to lysosomes for degradation. Accumulating evidence supports the pivotal role of autophagy in host defense against intracellular pathogens implicating both innate and adaptive immunity. Many of these pathogens cause common zoonotic infections worldwide. The induction of the autophagic machinery by innate immune receptors signaling, such as TLRs, NOD1/2, and p62/SQSTM1 in antigen-presenting cells results in inhibition of survival and elimi

Scaling Phage-Bacteria Interaction Contexts: From Pairwise Mechanisms to Community Dynamics.

Bacteriophages (phages), viruses that parasitize bacteria, hold tremendous potential as antimicrobial agents, microbiome modulators, and industrial biocontrol tools; yet clinical and environmental applications remain frustratingly inconsistent. Decades of research on isolated phage-bacteria pairs have revealed fundamental mechanisms governing infection specificity, coevolutionary arms races, and resistance trade-offs. These foundational studies, however, do not help predict outcomes when phages encounter multi-species assemblages characteristic of natural ecosystems. In this review, we integra

Location, Location, Location-Commensalism, Damage and Evolution of the Pathogenic Neisseria.

The 10 human-restricted Neisseria species all colonize mucosal surfaces, but show a spectrum of pathogenicity. The commensal Neisseria do not normally cause pathology, while the two pathogenic species, Neisseria meningitidis and Neisseria gonorrhoeae, straddle the border between commensalism and pathogenicity. Why the pathogenic Neisseria continue to mediate host damage after thousands of years of co-evolution with their human host, and why the commensal species have not acquired the ability to damage the host, if this capability provides a selective advantage, is not understood. One way the p

Torquetenovirus: the human virome from bench to bedside.

Torquetenovirus (TTV) is the most abundant component of human virome. Virologists have long ignored this orphan and highly divergent virus, in part because TTV cannot be cultured and because it lacks serology reagents and animal models. Nevertheless, it is almost endemic worldwide and is insensitive to current antiviral drugs, so its monitoring is useful in various conditions. To date, TTV as a marker has proved useful in at least two circumstances: to identify anthropogenic pollution and to assess functional immune competence in immunosuppressed individuals. This review summarizes recent find

Immune reconstitution syndrome and fungal infections.

PURPOSE OF REVIEW: Fungal infections-related immune reconstitution syndrome (IRS) poses challenging diagnostic and management issues in immunocompromised hosts. This review summarizes the current state of knowledge regarding its pathophysiologic basis, presentation, and treatment strategies. RECENT FINDINGS: Existing evidence suggests that IRS is a state of an imbalance between protective immunity and inflammatory pathology versus anti-inflammatory responses that restrain inflammation. IRS has been observed in diverse hosts including HIV-infected patients initiating potent antiretroviral ther

Re-defining granulomas: bacterial effectors, host circuits, and spatially resolved immunity.

Granulomas are organized immune aggregates of myeloid and lymphoid cells that arise when the host fails to eliminate persistent stimuli; however, their function remains paradoxical, by promoting both protection from systemic dissemination while also promoting a niche for microbial survival. Traditionally defined by histopathology and morphology, granulomas are now being revisited with high-throughput "omics approaches. The findings reveal previously unrecognized cellular diversity and cytokine gradients that together shape granuloma behavior. Across diverse pathogens, including Mycobacterium,

iTRAQ-based phosphoproteomic analysis reveals host cell's specific responses to Toxoplasma gondii at the phases of invasion and prior to egress.

Protein phosphorylation plays a key role in host cell-T. gondii interaction. However, the phosphoproteome data of host cell at various phases of T. gondii infection has not been thoroughly described. In this study, we assessed the host phosphoproteome data with isobaric tags for relative and absolute quantification (iTRAQ) method during the phases of T. gondii invasion (30 min post infection, PI) and prior to egress (28 h PI). Our iTRAQ analysis revealed a total of 665 phosphoproteins, among which the significantly regulated phosphoproteins in different between-group comparisons were further a

Molecules-mediated bidirectional interactions between microbes and human cells.

Complex molecules-mediated interactions, which are based on the bidirectional information exchange between microbes and human cells, enable the defense against diseases and health maintenance. Recently, diverse single-direction interactions based on active metabolites, immunity factors, and quorum sensing signals have largely been summarized separately. In this review, according to a simplified timeline, we proposed the framework of Molecules-mediated Bidirectional Interactions (MBI) between microbe and humans to decipher and understand their intricate interactions systematically. Inflammatory bowel disease through the lens of microbe-host interactions: immunomodulation, metabolic effects, and genetic susceptibility in microbiota dysbiosis.

Inflammatory bowel disease (IBD), including ulcerative colitis, Crohn's disease, and inflammatory bowel disease-unclassified, is a complex intestinal disease influenced by microbial factors, genetic and environmental. IBD has become a global disease with an increasing prevalence, endangering human health worldwide. Through its interactions with host immunity, bacterial metabolites, and genetic components, the intestinal microbiome plays a crucial role in initiating and advancing IBD. Treatment for IBD includes not only corticosteroids, aminosalicylates, antibiotics, TNF-α, α4β7 integrins, IL-1

No effector is an island: joint action of Toxoplasma gondii secreted effectors in host immune subversion.

The molecular dialogue between host and pathogen is defined by competition for the intracellular niche. Toxoplasma gondii exemplifies this mastery by deploying secreted effectors to remodel host cell biology and neutralize immune defenses. Yet a striking paradox has emerged, in which, despite exporting only a limited subset of effectors beyond the parasitophorous vacuole membrane into the host cell nucleus, the parasite achieves extensive transcriptional and functional reprogramming of the host. In this mini-review, we synthesize emerging evidence that Toxoplasma resolves this constraint throu

Interaction between tick and host microbiotas: a four-step waltz.

Tick-borne diseases represent a growing public health concern worldwide, yet the microbial factors that govern pathogen transmission remain incompletely understood. Over the past decade, high-throughput metagenomics and functional studies have revealed that two distinct microbial communities-the vertebrate host's skin microbiota and the tick's own microbiome-act synergistically as key modulators of pathogen acquisition, persistence within the vector, and successful transmission to the vertebrate host. At the feeding site, the skin microbiota orchestrates local cutaneous immunity, influences in

Oncogenic Viruses in Organ Transplantation: Implications of Virus-Host Interactions for Cancer Development.

Organ transplantation significantly enhances the survival and quality of life for recipients. However, multiple dependent and independent variables can adversely affect life expectancy after transplantation. Cancer is one of the most common causes of morbidity and mortality for long-term organ transplant recipients. The incidence of cancer in transplanted tissues can be twice as high in approximately 32 distinct cancer types. Oncogenic viruses present in graft tissues may contribute to the etiology of various cancers in transplant recipients. Such oncogenic viruses include hepatitis viruses, p

Chronic suppurative otitis media: disrupted host-microbial interactions and immune dysregulation.

Recent research has uncovered new mechanisms that disrupt the balance between the host and microbes in the middle ear, potentially leading to dysbiosis and chronic suppurative otitis media (CSOM). Dysbiotic microbial communities, including core pathogens such as persister cells, are recognized for displaying cooperative virulence. These microbial communities not only evade the host's immune defenses but also promote inflammation that leads to tissue damage. This leads to uncontrolled disorder and pathogen proliferation, potentially causing hearing loss and systemic complications. In this discu

The effects of radioactive pollution on the dynamics of infectious diseases in wildlife.

The interactions between infectious diseases and chemical pollution are well known and recognised as important factors in regulating the way wild animals respond to contaminant exposure. However, the impact of ionising radiation and radionuclides has often been overlooked when assessing host-pathogen interactions in polluted habitats, despite often occurring together with chemical contamination. Nevertheless, a comprehensive body of literature exists from laboratory and field studies on host-pathogen relationships under radiation exposure, and with a renewed interest in radioecology developing…
