Symptoms related to sleep, weight, and endocrine dysfunction are common in Alzheimer's disease (AD) and Lewy body dementia (LBD). The cause of these symptoms is not known, but they may be related to hypothalamic neurodegeneration. We performed a systematic search of MEDLINE and EMBASE for studies using MRI or PET imaging to examine the hypothalamus in AD or LBD. The Newcastle-Ottawa scale was used to assess the risk of bias. A random-effects meta-analysis was conducted using the standardised mean difference (SMD) in hypothalamic volume, and a narrative synthesis was used to examine association

Polycystic ovary syndrome (PCOS) is the most common endocrine disorder among women of reproductive age, with a prevalence estimated between 10% and 13%. It is characterized by ovulatory dysfunction, hyperandrogenism, and polycystic ovarian morphology. Antimüllerian hormone (AMH) plays a key role in regulating normal reproductive function in both males and females. Over the past few decades, significant progress has been made in understanding AMH, with numerous studies revealing its unexpected roles throughout the hypothalamic-pituitary-gonadal axis. Antimüllerian hormone and its receptor are a

The anterior and posterior hypophyseal hormones alter glucose metabolism in health and disease. Secondary diabetes may occur due to hypersecretion of anterior pituitary hormones like adrenocorticotrophic hormone in Cushing's disease and growth hormone in acromegaly. Other hormones like prolactin, gonadotropins, oxytocin and vasopressin, though not overtly associated with causation of diabetes, have important physiological role in maintaining glucose homeostasis. Hypoglycaemia is not an unusual occurrence in hypopituitarism. Many of the medications that are used for treatment of hypophyseal dis

BACKGROUND: Hypothalamic gonadotropin-releasing hormone (GnRH) plays a central role in regulating the pituitary-gonadal axis. The pulsatility of GnRH release is critical for maintaining the function of GnRH receptors and the secretion pattern of gonadotropins, namely follicle-stimulating hormone (FSH) and luteinizing hormone (LH), which regulate endocrine function and follicular growth and maturation. During the luteal phase, LH is crucial for supporting a functional corpus luteum and stimulating it to produce progesterone, estradiol and relaxin.Hypothalamic hypogonadotropic hypogonadism origi

Apart from its protective function, the skin is also both hormonally and metabolically active. Many hormones are produced in the skin and the regulation of these changes is controlled, as are other organs, by the pituitary gland and hypothalamus. Under the influence of stress factors such as ultraviolet radiation, many reactions are stimulated. After exposure to light in keratinocytes apart from eumelanin, also proopiomelanocortin, dopamine, and opioid substances - β-endorphins are produced. The skin also has a hypothalamic-pituitary-adrenal axis homolog. A corticotropin-releasing hormone is p

Oscillating levels of adrenal glucocorticoid hormones are essential for optimal gene expression, and for maintaining physiological and behavioural responsiveness to stress. The biological basis for these oscillations is not known, but a neuronal "pulse generator" within the hypothalamus has remained a popular hypothesis. We demonstrate that pulsatile hypothalamic activity is not required for generating ultradian glucocorticoid oscillations. We show that a constant level of corticotrophin-releasing hormone (CRH) can activate a dynamic pituitary-adrenal peripheral network to produce ultradian ad

The secretion of peptide hormone by the pituitary gland is obligatory for controlling a wide-range of downstream physiological processes. To achieve this, endocrine cells must respond co-ordinately to hypothalamic input to release defined pulses of hormone into the bloodstream. The organ context is clearly essential for proper hormone release since enzymatically dispersed cells mount attenuated responses to secretagogue. Yet, scant attention has been paid to whether endocrine cell populations are organized at the tissue level within the pituitary. Recently, using transgenic animals allied to s

Reproduction and metabolism are intricately linked. Gut hormones play key roles in the regulation of body weight and glucose homeostasis, factors that influence the functioning of the hypothalamic-pituitary-gonadal axis and reproductive outcomes. Data from rodent models suggest gut hormones may have direct stimulatory effects on reproductive hormone release. However, the effects of gut hormones on reproductive function in humans are more complex, with possible involvement of direct (e.g. via gut hormone receptor agonism) as well as indirect (e.g. via weight reduction in people with obesity) me

Recent studies have modified remarkably our concepts regarding the hormonal control of the human menstrual cycle. The previously accepted hypothesis was mainly based on rat experiments. It postulated a single integrated hypothalamic-pituitary-ovarian feedback system directed by a "CNS clock". Primate investigations now suggest two interrelated, but separable functional elements, i.e. a hypothalamic-pituitary unit and a gonadal-pituitary unit. The continuous pulsatile, yet unvarying hypothalamic secretion of gonadotropin-releasing hormone (GnRH) facilitates a direct pituitary response to changi

Cytokines may regulate the function of the hypothalamic-pituitary-adrenal axis during schistosomiasis. This possibility was investigated in baboons experimentally infected with Schistosoma mansoni. Serum levels of corticotrophin-releasing hormone, adrenocorticotrophin, cortisol and dehydroepiandrosterone were confirmed to be decreased in infected baboons as previously shown. To explore if this effect is associated with specific expression of cytokines with endocrine activity, and are also associated with the pathology of the disease, Northern blots for interleukin-1beta, interleukin-6, tumor n

OBJECTIVES: Various complaints of patients with fibromyalgia often resemble clinical features observed in patients with hypothalamic-pituitary diseases. The aim of this study was to evaluate whether patients with hypothalamic-pituitary diseases are at increased risk for fibromyalgia syndrome (FMS).
METHODS: A questionnaire for evaluating fibromyalgia-associated symptoms was sent to 121 patients with hypothalamic-pituitary disorders (HPD) (60 women, 61 men; mean age, 55.4 years; range, 21-83 years) of the endocrine outpatient clinic. 115 patients (57 women, 58 men; mean age 56.9 years; range,

Perimenopause is characterized by fluctuating and ultimately declining estrogen and progesterone levels and is associated with an increased risk of depression. Approximately 35% of women experience depressive symptoms, and up to 24% may develop a depression during the perimenopause. The etiology is multifactorial, involving both psychosocial and biological factors such as an individual vulnerability to hormonal fluctuations ('hormonal mood sensitivity'). Neurobiological mechanisms include interactions between the hypothalamic-pituitary-gonadal and hypothalamic-pituitary-adrenal axes, as well a

Deficiency of one or more anterior pituitary hormones may follow treatment with external irradiation when the hypothalamic-pituitary axis falls within the fields of irradiation. Hypopituitarism occurs in patients who receive radiation therapy for pituitary tumours, nasopharyngeal cancer and primary brain tumours, as well as in children who undergo prophylactic cranial irradiation for acute lymphoblastic leukaemia, or total body irradiation for a variety of tumours and other diseases. The degree of pituitary hormonal deficit is related to the radiation dose received by the hypothalamic-pituitar

Selective breeding of broilers has significantly improved growth rates, muscle mass, and feed efficiency and may have influenced the neuroendocrine systems that regulate growth and metabolism. Embryonic development represents one-third of the life of a modern broiler. To assess the impact of genetic selection on the neuroendocrine regulation of growth and metabolism during embryonic development, we examined mRNA expression of growth-related genes in the hypothalamus and anterior pituitary of two chicken breeds: the modern Ross 708 broiler and the Athens Canadian Random Bred (ACRB) line, the ol

Alcoholic liver disease shares with other nonthyroidal illnesses patterns of serum thyroid hormones that usually do not reflect true thyroid function. Although ethanol can affect the thyroid gland and also may directly interfere with thyroid hormone metabolism, most patients with alcoholic liver disease are euthyroid. However, some have mild hypothalamic pituitary axis dysfunction. In persons who chronically and continuously drink alcohol, there are demonstrable biochemical abnormalities similar to the effects of thyroid hormone on the liver. However, a primary pathogenetic role in alcoholic h

Clinically relevant HP dysfunction occurs infrequently in the course of HIV infection and AIDS, and is usually due to destruction or infiltration of the region by opportunistic infections. However, more subtle defects occur with much greater frequency. Since these impairments, especially in the gonadotrophic and corticotrophic axes, are observed in HIV patients before the development of ARC or AIDS, direct involvement of the hypothalamus or pituitary by the neurotropic HIV virus or by other HIV-mediated factors is possible. Further investigation of hypothalamic and pituitary hormones by provoc

Prader-Willi syndrome (PWS) is a rare genetic neurodevelopmental disorder linked to the lack of expression of specific maternally imprinted genes located in the chromosomal region 15q11-q13. Impaired hypothalamic development and function explain most of the phenotype that is characterized by a specific trajectory from anorexia at birth to excessive weight gain at later ages, which is accompanied by hyperphagia and early severe obesity, as well as by other hormonal deficiencies, behavioral deficits, and dysautonomia. In almost all patients, their endocrine dysfunction involves growth hormone de

BACKGROUND: Sepsis-induced acute skeletal muscle wasting would impede critical patients' recovery. The underlying mechanism for this metabolic disorder has not been fully elucidated. Here, we tested the hypothesis that circulating hormone ghrelin could regulate sepsis-induced acute skeletal muscle wasting via hypothalamic Janus Kinase 2 (JAK2), signal transducer and activator of transcription 3 (STAT3) and agouti-related protein (AgRP) pathway.
METHODS: Cecal ligation and puncture (CLP) was used to establish the model of sepsis-induced acute skeletal muscle wasting in rats. Ghrelin was admini
