Effects of calcitonin and CGRP alone or in combination on food intake and forestomach (reticulum) motility in sheep.

The effects of intracerebroventricular (ICV) and intravenous (IV) administration of calcitonin and calcitonin gene-related peptide (CGRP) on feeding behavior and reticular motility were investigated in sheep. ICV calcitonin at a dose of 2 to 200 mU/kg reduced, in a dose-related manner, the immediate (0-60 min) food intake. The daily food intake was also significantly (p less than 0.05) decreased for doses up to 20 mU/kg, and the frequency of reticular contractions during the first hour of eating was decreased by 27.9%. Calcitonin at the highest IV dose (200 mU/kg) did not affect feeding behavi

Endogenous sugar acid derivative acting as a feeding suppressant.

Evidence suggests that endogenous sugar acids 3,4-dihydroxybutanoic acid (2-deoxytetronic acid, 2-DTA) and 2,4,5-trihydroxypentanoic acid (3-deoxypentonic acid, 3-DPA) may participate in the regulation of feeding. To study the effect of 2-buten-4-olide, a 2-DTA synthetic derivative, on food intake, male Wistar rats were subjected to various applications. Intraperitoneal administration of 2-buten-4-olide in doses of 30 to 100 mg/kg, decreased food intake dose-dependently by reducing meal frequency, meal size and eating rate, and prolonging meal duration, latency to eat the first meal after inje

Involvement of hypothalamic somatostatin in the suppression of growth hormone secretion by central corticotropin-releasing factor in conscious male rats.

The role of central corticotropin-releasing factor (CRF) in the regulation growth hormone (GH) secretion was studied in freely moving conscious male rats with indwelling intra-atrial and intracerebroventricular (i.c.v.) cannulae. GH measurements in blood samples obtained every 20 min from 10.00 to 14.00 h in control animals injected with saline either intravenously (i.v.) or into the lateral cerebral ventricle revealed that spontaneous GH secretion was pulsatile, and occurred regularly at around 12.00 h. When ovine CRF (10 micrograms) was injected i.c.v., spontaneous GH secretion was inhibited

Behavioral interaction of adenosine and diazepam in mice.

Mice were implanted with chronic indwelling cannulae in the lateral cerebral ventricle. The behavioral interaction of intraperitoneal (i.p.) injections of diazepam with intracerebroventricular (i.c.v.t.) injections of adenosine or 5'-N-ethylcarboxamidoadenosine (NECA) was examined on spontaneous locomotor activity. Concurrent injections of i.c.v.t. adenosine and i.p. diazepam, at doses which had no significant effect on locomotor activity when given alone, acted synergistically to produce a marked depression of locomotor activity. In contrast, i.p. injections of diazepam did not potentiate the

Inhibitory effect of sulpiride on cinchophen gastric ulceration in dogs.

An intraventricular administration of sulpiride suppressed the experimental cinchophen ulcer, thereby the effect of repeated administration of the material on cinchophen ulcerogenesis is evident, whereas intermittent administration lacked this effect. Such an effect as reducing experimental cinchophen ulceration, which is due to central neurohumoral mechanism, following intraventricular infusion of sulpiride appears to be ascribable to a chemical blockage of the hypothalamic structure. It can thus be concluded that sulpiride acts principally at the level of hypothalamus. Its inhibitory action

Gys1 antisense therapy rescues neuropathological bases of murine Lafora disease.

Lafora disease is a fatal progressive myoclonus epilepsy. At root, it is due to constant acquisition of branches that are too long in a subgroup of glycogen molecules, leading them to precipitate and accumulate into Lafora bodies, which drive a neuroinflammatory response and neurodegeneration. As a potential therapy, we aimed to downregulate glycogen synthase, the enzyme responsible for glycogen branch elongation, in mouse models of the disease. We synthesized an antisense oligonucleotide (Gys1-ASO) that targets the mRNA of the brain-expressed glycogen synthase 1 gene (Gys1). We administered G

Neonatal ventriculitis: a case series and review of literature.

Ventriculitis after meningitis is a serious complication in the neonatal age group. The role of intraventricular antibiotics in treatment is controversial. We present five such cases which were refractory to conventional intravenous antibiotic therapy, had persistent features of ventriculitis and in whom raised intracranial pressure (ICP) necessitated insertion of an external ventricular drain (EVD). Three of the five infants required intraventricular antibiotics but also developed EVD-related complications. Early diagnosis of ventriculitis and treatment is necessary to avoid a fatal outcome.

Behavioral and physiological responses to central administration of corticotropin-releasing factor in the bluebanded goby (Lythrypnus dalli).

Central manipulation of neuromodulators is critical to establishing causal links between brain function and behavioral output. The absence of a rigorous method of evaluating intracerebroventricular (i.c.v.) injection efficacy in small model organisms is one reason why peripheral administration of neuroactive substances is more common. We use the bluebanded goby (Lythrypnus dalli), a small, highly social fish, to 1) validate our method of i.c.v. injection by testing the hypothesis that corticotropin-releasing factor (CRF) elevates ventilation rate (VR) and 2) propose a novel bioassay using basa

Central administration of metastin increases food intake through opioid neurons in chicks.

Metastin, an RFamide peptide, has been isolated from human placenta and possesses several physiological actions in mammals. However, little is known about this bioactive peptide in avian species. This study was conducted to assess the effect of metastin on feeding behavior of chicks (Gallus gallus). The food intake of chicks is significantly increased by the intracerebroventricular injection of metastin. Beta-funaltrexamine, a mu-opioid receptor antagonist, significantly attenuates metastin-induced food intake in chicks. In contrast, delta- and kappa-opioid receptor antagonists did not show an

Intracerebroventricular administration of the angiotensin II receptor antagonist saralasin reduces respiratory rate and tidal volume variability in freely moving Wistar rats.

The possible importance of intra-individual variations in respiratory rate and tidal volume has recently gained interest in psychiatric research, as a result of the observations that patients with panic disorder or premenstrual dysphoric disorder display enhanced respiratory variability as compared to controls. Although the role of brain neurotransmitters in the regulation of breathing has been extensively studied, as yet data on the central regulation of respiratory variability is sparse. Prompted by previous studies indicating that angiotensin II (ANG II) may influence ventilation as well as

Acute third ventricular administration of insulin decreases food intake in two paradigms.

The pancreatic hormone, insulin, has been hypothesized to be an important regulator of food intake. Consistent with this hypothesis is the finding that exogenous insulin, in doses that do not affect blood glucose, reliably suppresses food intake and body weight. However, previous experiments have utilized a long-term delivery paradigm, in which insulin is administered via osmotic minipump and changes in body weight and food intake are measured across days. In separate experiments, we report that acute central injections of insulin can reduce food intake. In Experiment 1, injection of insulin (

The effect of netilmicin and vancomycin on lipid peroxidation processes in cerebrospinal fluid in children with hydrocephalus.

In biological systems, it is difficult to determine free radicals because of their reactivity and their very short time of existence. On the basis of markers, which come into being as a result of radical processes, one might believe that there exist reactive oxygen species. One of the determinants of free radical activity of oxygen is the presence of malondialdehyde (MDA), a final product of lipid peroxidation. This study aimed at finding the answer to the question whether the concentration of netylmicin and vancomycin influences the amount of substances reacting with thiobarbituric acid (TBA)

Intracerebroventricular administration of copper-zinc superoxide dismutase inhibits pulsatile luteinizing hormone secretion in ovariectomized ewes.

Intracerebroventricular (i.c.v.) injection of an inhibitor of nitric oxide synthase (NOS) abolishes pulsatile luteinizing hormone (LH) secretion. It has been demonstrated that structural and functional analogs of copper-zinc superoxide dismutase (Cu,Zn-SOD) inhibit neuronal NOS. The present study examined the ability of Cu,Zn-SOD to affect pulsatile LH release in the ewe. Bovine Cu,Zn-SOD was administrated into the third cerebral ventricle of unanesthetized, freely moving, ovariectomized (OVX) ewes. Jugular blood samples were taken every 15 min for 5 h before and 8 h after i.c.v. injections. I

Activation of GABAA receptors in hypothalamus modulates PGF2 alpha- or PGE2-induced catecholamine secretion in rats.

We previously reported that injection of PGF2 alpha into the third cerebral ventricle produces hyperglycemia and hyperthermia associated with catecholamine secretion in anesthetized rats. We have also studied the potency of catecholamine secretion induced by injecting PGE2 or PGF2 alpha into the third cerebral ventricle and the effect of the GABA-selective agonist, muscimol, on the catecholamine secretion induced by PGE2 or PGF2 alpha. Administration of 50 micrograms of PGE2 into the third cerebral ventricle increased norepinephrine secretion to a greater extent than the same dose of PGF2 alph

Centrally acting endogenous hypotensive substances in rats subjected to endotoxic shock.

Cerebroventricular perfusate (CVP) from rats subjected to endotoxic shock was infused into the lateral ventricle of the naive rat. This procedure produced a hypotensive response in the recipient rat which could be reversed by the intravenous injection of the opioid antagonist naltrexone. The degree of endotoxemic hypotension in the donor rat was attenuated by perfusing the cerebroventricular system with mock CSF. The results suggest the existence of endogenous hypotensive substances in rat central nervous system, possibly opioid in nature, which may play a critical role in the pathogenesis of

Cholecystokinin octapeptide alters morphine-induced effects on EEG power spectra both quantitatively and qualitatively.

In the present study, EEG analysis was used to determine if cholecystokinin octapeptide (CCK-8) has an effect on the EEG spectral profile associated with morphine-induced bursting. Adult female Sprague-Dawley rats were implanted with cortical electrodes and indwelling i.c.v. and i.v. cannulas. On the day of the experiment, each rat received an i.c.v. injection of either H2O or one of four doses of CCK-8: 8.0, 16.0, 32.0 or 64.0 ng. Ten minutes after receiving the i.c.v. injection, each rat received a 10.0 mg/kg dose of morphine by i.v. injection. Neither the i.c.v. injections of H2O nor CCK-8

Efficacy of lateral ventricular injection of epinephrine, cyproheptadine, or adenosine triphosphate on feed intake in thiamin-deficient turkeys.

In previous work, thiamin deficiency (TD) resulted in decreased brain levels of adenosine triphosphate (ATP) and epinephrine, but serotonin (5-HT) was increased in the turkey. The goal of the present study was to determine whether the introduction of calculated levels of epinephrine, ATP, or cyproheptadine into the lateral ventricle could affect feed intake of TD birds. Adenosine triphosphate, epinephrine, and cyproheptadine (5-HT antagonist) were injected into the lateral ventricle of TD and control birds. These injections did not alter feed intake in the control birds. Cyproheptadine and ATP

Effect of centrally administered atropine and pirenzepine on radial arm maze performance in the rat.

The effect of two anticholinergic drugs administered intracerebroventricularly on acquisition of an 8-arm radial maze task was examined in the rat. Increasing doses of atropine (1, 4.5, 22.5, 45 micrograms/rat) and pirenzepine (4.5, 15, 60, 90 micrograms/rat) significantly impaired performance in the working-memory components of the task. For both drugs this impairment was linearly related to the log of the administered doses and log-dose-response relationship were parallel. The regression lines calculated for each parameter for both drugs were parallel to each other, thus allowing us to calcu

[Inhibitory effects of gamma-aminobutyric acid and diazepam on ventricular arrhythmias induced by hypothalamic electric stimulation].

The experiments were carried out on rabbits with ventricular arrhythmias (VA) induced by hypothalamic electric stimulation. The effects of GABA or diazepam (Dz) and picrotoxin on VA were observed. After GABA 0.25, 0.5, 1 mg/rabbit were injected into cerebral ventricle (icv) or cisterna magna (icm), the VA was dose-dependently reduced, the similar effect was seen after Dz 0.1 mg was injected into subarachnoid cavity (sac) or icm. The effect was not induced by GABA sac or Dz icv. Picrotoxin 20 micrograms icv or 30 micrograms icm obviously increased VA, and this effect was partly antagonized by p
