Zinc deficiency is commonly attributed to inadequate absorption of the metal. Instead, we show that body zinc stores in Drosophila melanogaster depend on tryptophan consumption. Hence, a dietary amino acid regulates zinc status of the whole insect—a finding consistent with the widespread requirement of zinc as a protein cofactor. Specifically, the tryptophan metabolite kynurenine is released from insect fat bodies and induces the formation of zinc storage granules in Malpighian tubules, where 3-hydroxykynurenine and xanthurenic acid act as endogenous zinc chelators. Kynurenine functions as a p

The life cycle of African trypanosomes involves adaptations to the defense mechanisms of two completely different hosts, the insect vector Glossina and the mammalian host. This interplay ultimately determines host resistance and/or tolerance to parasite infection. In the tsetse fly, the immune deficiency (IMD)-regulated pathway, the scavenger receptor peptidoglycan-recognition protein LB (PGRP-LB), and the reactive oxygen species (ROS)-mediated response modulate the insect's capacity to transmit the parasite. In experimental mice, control of parasite burden and tissue pathogenicity relies on t

Insects of the order Diptera are vectors for parasitic diseases such as malaria, sleeping sickness and leishmania. In the search for genes encoding proteins involved in the antiparasitic response, we have used the protozoan parasite Octosporea muscaedomesticae for oral infections of adult Drosophila melanogaster. To identify parasite-specific response molecules, other flies were exposed to virus, bacteria or fungi in parallel. Analysis of gene expression patterns after 24 h of microbial challenge, using Affymetrix oligonucleotide microarrays, revealed a high degree of microbe specificity. Many

Diet composition significantly influences the chemical quality of edible insect ingredients. This study compared Tenebrio molitor larvae reared on wheat bran (WB) and tuna by-products (TB) using proximate analysis and metabolomics. On a dry matter basis, crude protein was similar at about 45 g per 100 g, whereas TB showed higher lipid and lower carbohydrate contents. Fatty acid methyl ester analysis revealed higher oleic and linoleic acids and a shift toward C18 unsaturated fatty acids in TB. Total hydrolyzable amino acids and essential amino acid proportion were conserved, although proline wa

Centromere protein J (CENPJ) is a core centrosomal protein essential for centriole duplication and microtubule organization, and mutations in the gene cause aberrant neuronal morphology and primary microcephaly in mammals. However, its functions in insect development remain poorly understood. In this study, we investigated the role of CENPJ in the silkworm Bombyx mori, a lepidopteran model featuring unique holocentric chromosomes. We found that CENPJ is evolutionarily conserved and predominantly expressed in silkworm gonads. Cellular assays demonstrated that CENPJ exhibits cytoplasmic localiza

The aim of this study was to generate a recombinant protein to represent the entire extracellular domain of canine desmoglein 1 (Dsg1), a desmosomal cell-cell adhesion molecule, by the baculovirus expression system. Cotransfection of a baculovirus transfer vector containing the cDNA for the entire extracellular domain of canine Dsg1 with baculovirus DNA into insect cells resulted in the secretion of soluble canine Dsg1 into insect culture supernatants. Immunoreactivity of 11 human pemphigus foliaceus (PF) sera against the cell surface of canine keratinocytes was completely removed when the ser

Proteinase 3 (PR3) is a multifunctional serine protease found in myeloperoxidase-positive granula of PMN. It is also the main target antigen for antineutrophil cytoplasmic antibodies (ANCA) found in Wegener's granulomatosis (WG). There is ample evidence that these autoantibodies play an active role in the pathogenesis of this disease. We have established the expression of the autoantigen (PR3) in a baculovirus expression system using insect cells. Recombinant PR3 was identified by IIF, direct ELISA using 3 different MoAbs and by catching ELISA using a MoAb and a rat polyclonal Ab or cANCA-posi

A approximately 370 base pair cDNA encoding the alpha insect toxin Lqh alpha IT of the scorpion Leiurus quinquestriatus hebraeus was cloned and sequenced. The deduced amino acid sequence for the putative mature polypeptide is identical to the protein sequence determined chemically (Eitan et al., Biochemistry 29, 5941, 1990). A 19 amino acid signal peptide precedes the 64 amino acid long toxin. Two additional amino acid residues that do not correspond to the purified toxin are found at the COOH-terminus and may imply post-translational modification. The signal peptide region in the present clon

BACKGROUND: Iron metabolism and regulation is an indispensable part of species survival, most importantly for blood feeding insects. Iron regulatory proteins are central regulators of iron homeostasis, whose binding to iron response element (IRE) stem-loop structures within the UTRs of genes regulate expression at the post-transcriptional level. Despite the extensive literature on the mechanism of iron regulation in human, less attention has been given to insect and more specifically the blood feeding insects, where research has mainly focused on the characterization of ferritin and transferri

Insects are important vectors of diseases with remarkable immune defense capabilities. Hymenopteran endoparasitoids are adapted to overcome the host defense system and, therefore, are useful sources of immune-suppressing proteins. Not much is known about venom proteins in endoparasitoids, especially those that have a functional relationship with polydnaviruses (PDVs). Here, we describe the isolation and characterization of a small venom protein (Vn4.6) from an endoparasitoid, Cotesia rubecula, which interferes with the activation of the host hemolymph prophenoloxidase. The coding region for Vn

The baculovirus expression vector system is an established platform for large-scale production of (glyco)proteins, subunit vaccines, virus-like particles, and recombinant adeno-associated virus (rAAV) vectors. We engineered a novel bacmid vector (BAC6) to improve genetic stability by deletion of the non-homologous repeat (hr) origin of DNA replication (ori) and to preserve product integrity and recovery through deletion of chitinase and cathepsin. Tn7-based transposition in E. coli and homologous recombination in insect cells were combined in BAC6 to drive expression from the odv-e56 and polyh

INTRODUCTION: The German cockroach (Blattella germanica) is a notorious urban pest with significant health impacts, owing in part to its highly sophisticated gustatory system that enables precise food discrimination and toxin avoidance. Odorant-binding proteins (OBPs) are known to play critical roles in insect chemoreception, yet their specific contributions to sweet and bitter compound recognition remain poorly understood.
METHODS: We investigated the function of three maxillary palp-enriched OBPs (BgerOBP10, BgerOBP44, and BgerOBP91) using fluorescence competitive binding assays, RNA interf

Entomopathogenic fungi of the genus Beauveria are widely used biological control agents that infect diverse insect hosts and can also associate with plants as rhizosphere colonizers and endophytes. Within this genus, Beauveria bassiana is a cosmopolitan generalist, whereas Beauveria brongniartii exhibits a narrower host range, primarily targeting soil-dwelling coleopteran larvae with limited evidence of plant colonization. To explore genomic differentiation associated with this ecological divergence, the commercially exploited B. brongniartii strain BIPESCO2 and B. bassiana ATHUM 4946 were seq

Influenza causes approximately one billion infections and hundreds of thousands of deaths each year. Occasionally, highly pathogenic new virus strains emerge. Research on emerging influenza viruses or multiple viral strains is often limited by the need for high biosafety level (BSL) laboratories. To overcome this limitation, we attempt to establish a BSL-1-compatible influenza research platform using baculovirus surface display technology. We constructed influenza virus pseudotypes HA-Bac and NA-Bac by displaying hemagglutinin (HA) and neuraminidase (NA) proteins from influenza viruses on the

Traditionally, vaccine development involves tradeoffs between immunogenicity and safety. Live-attenuated vaccines typically offer rapid and durable immunity but have reduced safety when compared to inactivated vaccines. In contrast, the inability of inactivated vaccines to replicate enhances safety at the expense of immunogenicity, often necessitating multiple doses and boosters. To overcome these tradeoffs, we developed the insect-specific alphavirus, Eilat virus (EILV), as a vaccine platform. To address the chikungunya fever (CHIKF) pandemic, we used an EILV cDNA clone to design a chimeric v

The degradation of cat immunoglobulin G (IgG) in blood-fed adult C. felis midguts was examined. SDS-PAGE analysis of dissected midgut extracts obtained from C. felis that had been blood fed for various times between 0 to 44 h revealed that by 24 h most of the high molecular weight proteins, including the heavy chain of IgG, were digested. A 31-kDa serine protease with IgG degrading activity was purified from fed C. felis midguts by benzamidine affinity chromatography, hydrophobic interaction chromatography, and cation exchange chromatography. Three primary cleavage products between 30- and 40-

UNLABELLED: In Latin America, Lutzomyia longipalpis is the main vector of the protozoan parasite Leishmania infantum, which is the causal agent of American Visceral Leishmaniasis. This insect uses male-produced pheromones for mate recognition. Elucidation of pheromone biogenesis or its regulation may enable molecular strategies for mating disruption and, consequently, the vector's population management. Motivated by our recent results of the transcriptomic characterization of the L. longipalpis pheromone gland, we performed a proteomic analysis of this tissue combining SDS-PAGE, and mass spect

This study was undertaken to assess the binding properties of BmK AS on both mammal and insect excitable cell membranes. It was found that BmK AS bound specifically to a single class of non-interacting binding sites on both rat brain and cockroach nerve cord synaptosomes with high affinity (K(d) = 1.49 +/- 0.14 and 0.79 +/- 0.29 nM) and low capacity (B(max) = 1.39 +/- 0.09 and 6.60 +/- 1.25 pmol/mg protein), respectively. Binding kinetics showed that BmK AS could bind and reach equilibrium quickly, and dissociate partially from its binding sites on both kinds of synaptosomes. The binding of Bm

A limitation to effective field use of naturally occurring nuclear polyhedrosis viruses (NPVs) is the slow rate at which they kill their host. In making NPVs a more attractive pest management tool, this problem has been addressed by modifying NPVs genetically to express insecticidal proteins resulting in substantial increases in their speed of action. One concern associated with these recombinant NPVs, however, is their effects on nontarget insects associated with pests targeted for control by applications of NPVs. Our studies evaluated the direct effects of wild-type Autographa californica NP
