BACKGROUND: Anticholinesterase insecticides (antiChE), including organophosphates and carbamates, are widely used to control crop pests in agriculture, household pests, and animal parasites, making them a potential source of poisoning in both humans and animals. This retrospective study aimed to document the frequency of antiChE positive samples in animal specimens analysed at the Department of Veterinary Pharmacology, Faculty of Veterinary Science, Chulalongkorn University from January 1, 2019, to December 31, 2024 using a cholinesterase kit based on Ellman’s method, to raise awareness and mi

Codling moths, Cydia pomonella L. (Lepidoptera: Tortricidae), can cause severe damage in apples (Malus domestica Borkh) (Rosales: Rosaceae) and are targets of the majority of insecticide applications. A novel insecticide, isocycloseram, presents a new mode of action with the potential to be incorporated into integrated pest management programs for codling moths. The objective of this study was to evaluate the efficacy of isocycloseram on codling moths in apples using both laboratory and field trials. In the laboratory trials, a direct toxicity test was conducted to compare the effectiveness of

Insecticide resistance threatens the durability of mosquito control. Extending the effective life span of available insecticides is therefore a key challenge. Resistance management strategies (RMS) have been proposed and incorporated into policy guidance, yet their relative effectiveness remains poorly resolved. This review synthesizes the theoretical and empirical evidence for major RMS: mixtures, rotations, mosaics, high-dose/refuge, dose moderation, and release of susceptible males. Among these RMS, mixtures currently have the strongest support, with models consistently predicting delayed r

Loxoscelism, caused by bites from Loxosceles spiders, is a global health concern. Control of these spiders with insecticides remains poorly understood and requires assessment at both biochemical and behavioural levels. We evaluated the ability of Loxosceles laeta to detect cypermethrin-treated surfaces and examined the effects of exposure on enzymatic activity, oxidative stress, biomolecule oxidation and locomotor behaviour. Spiders preferred untreated surfaces and increased movement speed on treated ones, indicating a repellent effect. Acetylcholinesterase activity remained unchanged, whereas

The large-scale use of insecticides remains a cornerstone of malaria vector control, but its long-term effectiveness is undermined by the evolution of quantitative insecticide resistance (qIR) in mosquito populations. We develop and analyze a mathematical model to identify optimal deployment strategies for two insecticides that differ only in their relative efficacy against target mosquito populations. Resistance is represented as a continuous phenotypic trait influencing mosquito fecundity and mortality, and the model accounts for successive deployment periods. Our results show that when muta

Insecticide use remains one of the most common strategies worldwide for controlling household pests and disease vectors, despite growing concerns over misuse, health risks, and the emergence of insecticide resistance. To document household insecticide use and flea infestation across a gradient of plague endemicity in Madagascar, we used a multidisciplinary approach. A descriptive household survey and an observational checklist were implemented with 422 respondents and combined with flea infestation assessment based on flea sampling from small mammals and candle traps. Household insecticide use

This commentary discusses an influential study from 1993 that demonstrated, among West African children, an overall mortality benefit of insecticide-impregnated bednets, and the reduction of malaria prevalence by chemoprophylaxis. Led by Brian Greenwood and colleagues in The Gambia, the trial also showed these tools to be affordable and practicable. In the years since, >2 billion bednets have been provided to high-risk populations and have contributed greatly to reductions in malaria-attributable mortality. Seasonal malaria chemoprevention now protects 50 million African children annually. Few

BACKGROUND: Information on the status of insecticide resistance in malaria vectors is critical for implementing effective malaria vector control. The Sierra Leone National Malaria Control Programme, in collaboration with the PMI VectorLink project, assessed the resistance status to insecticides commonly used in public health, and associated resistance mechanisms in Anopheles gambiae, the main vector of malaria in Sierra Leone.
METHODS: The susceptibility of An. gambiae against pyrethroids with and without piperonyl butoxide (PBO), chlorfenapyr, clothianidin, bendiocarb and pirimiphos-methyl w

Insecticide misuse has caused pest resistance, stressing the need for novel pesticides. The isoxazoline structure offers broad-spectrum effectiveness, mammalian safety, and no cross-resistance. Developing efficient insecticides with this scaffold remains challenging. Based on the lead compound L17, 31 novel isoxazoline compounds with benzamide were made and evaluated for insecticidal efficacy and mechanisms, all showing efficacy against Plutella xylostella. Especially, M31, designed based on the three-dimensional quantitative structure-activity relationship (3D-QSAR), displayed activity with L

We explore borrelidin and semisynthetic derivatives as promising pesticide candidates targeting threonyl-tRNA synthetase (ThrRS). Enzymatic inhibition assays and pull-down assays supported ThrRS as the target of borrelidins for insecticidal activity. Interestingly, derivative 6 showed weak in vitro inhibition but comparable in vivo insecticidal activity to borrelidin. Comparative efficacy tests via intravenous (i.v.) and intraperitoneal (i.p.) administration in silkworm larvae indicated improved pharmacokinetics for compound 6. These findings highlight ThrRS as a valuable insecticidal target a

Insecticide resistance in anopheline malaria vectors poses a major threat to malaria control and elimination efforts. We conducted spatial analyses using publicly available and published data to document and model the geospatial distribution of Anopheles spp. insecticide resistance in Tanzania at the national, regional, district, and subdistrict levels for the 2012-2021 period. An insecticide resistance map modeled for 2022 predicted that anopheline insecticide susceptibility remained above the 90% susceptibility threshold for all major insecticide classes, except for pyrethroids. We calculate

Insecticide resistance is under strong selective pressure in Anopheles mosquitoes due to widespread usage of insecticides in vector control strategies. Resistance mechanisms likely cause changes that profoundly affect mosquito physiology, yet it remains poorly understood how selective pressures imposed by insecticides may alter the ability of the mosquito to host and transmit a Plasmodium infection. From pyrethroid-resistant field-derived Anopheles gambiae s.l. mosquitoes, we established resistant (RES) and susceptible (SUS) colonies by either selection for, or loss of insecticide resistance.

Buprofezin (BUP) is an insecticide which belongs to the thiadiazine structural family and known to damage DNA in mice. Though its toxic effect on human is not known clearly, understanding the mechanism of interaction of BUP with DNA can prove useful when required. Multi-spectroscopic experiments such as UV-Vis, fluorescence, circular dichroism (CD) and 1H NMR coupled with viscosity measurements, urea effect and voltametric studies were performed to ascertain the mode of binding of BUP with calf thymus DNA (CT-DNA). Analysis of UV-Vis and fluorescence spectra indicated the formation of a comple

BACKGROUND: Myanmar is one of the six countries in the Greater Mekong Subregion (GMS) of Southeast Asia. Malaria vectors comprise many Anopheles species, which vary in abundance and importance in malaria transmission among different geographical locations in the GMS. Information about the species composition, abundance, and insecticide resistance status of vectorial systems in Myanmar is scarce, hindering our efforts to effectively control malaria vectors in this region.
METHODS: During October and November 2019, larvae and adult females of Anopheles mosquitoes were collected in three sentine

INTRODUCTION: Patients who require mechanical ventilation after self-poisoning with ingested organophosphorus (OP) insecticides often die. Aspiration of stomach contents may contribute to lung injury and lethality. This study was designed to assess the severity of direct and indirect pulmonary injury created by pulmonary instillation of mixtures of OP insecticide, solvent (Solv) and porcine gastric juice (GJ) compared to controls.
METHODS: Terminally anaesthetised minipigs (groups n = 5) were exposed to sham bronchoscopy or given mixtures (0.5 mL/kg) of: saline, GJ, OP insecticide and GJ (OP

BACKGROUND: Phlebotomine sand flies are prominent vectors of Leishmania parasites that cause leishmaniasis, which comes second to malaria in terms of parasitic causative fatalities globally. In the absence of human vaccines, sand fly chemical-based vector control is a key component of leishmaniasis control efforts.
METHODS AND FINDINGS: We performed a literature review on the current interventions, primarily, insecticide-based used for sand fly control, as well as the global insecticide resistance (IR) status of the main sand fly vector species. Indoor insecticidal interventions, such as resi
