Regulation of the developmental programs in Toxoplasma by a novel SNF2L-containing chromatin remodeling complex.

Toxoplasma gondii is an extremely successful parasite infecting one third of the human population and numerous animals. It has a complex lif … binding sites, thus ensuring that the gene expression and reproduction patterns are matched to the life cycle stages. Conditional depletion of SNF2L offers an opportunity to study the unique biology of the parasite during pre-sexual and sexual developments in vitro.

Conserved RNA Binding Activity of Phosphatidyl Inositol 5-Phosphate 4-Kinase (PIP4K2A).

Plasmodium falciparum is a causative agent for malaria and has a complex life cycle in human and mosquito hosts. During its life cycle, the malarial parasite Plasmodium goes through different asexual and sexual stages, in humans and mosquitoes. Expression of stage-specific proteins is important for successful completion of its life cycle and requires tight gene regulation. In the case of Plasmodium, due to relative paucity of the transcription factors, it is postulated that posttranscriptional regulation plays an important role in stage-specific gene expression. Translation repression of speci

EBV Early Lytic Antigens, EBNA2 and PDL-1, in Progressive Multiple Sclerosis Brain: A Coordinated Contribution to Viral Immune Evasion.

Epstein-Barr virus (EBV) infection shows the strongest causative association with multiple sclerosis (MS), but its contribution to disease progression and the mechanisms allowing for viral persistence in the MS brain are still elusive. Studies in post-mortem MS brain tissue indicate an ongoing yet ineffective antiviral immune reaction in advanced stages of the disease. EBV has evolved strategies to evade immune recognition and clearance by the host immune system during both the latency and lytic phase of its life cycle. Recent evidence demonstrates that cells expressing EBV latent membrane pro

PfFBXO1 is essential for inner membrane complex formation in Plasmodium falciparum during both asexual and transmission stages.

Plasmodium species replicate via schizogony, which involves asynchronous nuclear divisions followed by semi-synchronous segmentation and cytokinesis. Successful segmentation requires a double-membranous structure known as the inner membrane complex (IMC). Here we demonstrate that PfFBXO1 (PF3D7_0619700) is critical for both asexual segmentation and gametocyte maturation. In Toxoplasma gondii, the FBXO1 homolog, TgFBXO1, is essential for the development of the daughter cell scaffold and a component of the daughter cell IMC. We demonstrate PfFBXO1 forming a similar IMC initiation scaffold near t

New insights into the life cycle, host cell tropism, and infection amplification of Leishmania spp.

The genus Leishmania comprises several species of digenetic protozoan parasites that cause a spectrum of diseases known as leishmaniases, which are transmitted to humans and other mammals through the bite of hematophagous female sand flies. Leishmania spp. and their invertebrate vectors are widely distributed across the globe, putting more than a billion people at risk. Once inside mammalian hosts, these intracellular parasites reside within parasitophorous vacuoles of host cells. Although macrophages are the primary infected cells in lesions, Leishmania can also infect other cell types, whose

Environmental factors determining the survival of winter ticks (Dermacentor albipictus) at different life stages in Québec, Canada.

Ticks are responsible for large economic losses because of their impact on livestock and wildlife. In recent years, winter ticks (Dermacentor albipictus) have caused mass mortalities in moose (alces) populations of North America, leading to host mortality due to severe blood loss. During their off-host stages (i.e. egg and larvae), winter ticks are particularly vulnerable to mortality because of potential exposure to stressful abiotic conditions that could dehydrate eggs or freeze larvae. We conducted a field experiment in Québec, Canada, to better understand larval questing behaviour and asse

Organoid-based in vitro system and reporter for the study of Cryptosporidium parvum sexual reproduction.

Many advances have been made recently in our understanding of Cryptosporidium's asexual cycle and sexual differentiation. However, the process of fertilization, which is required for transmission of infectious oocysts, is not well understood. Typical cancer cell-based culture only allows robust exploration of asexual cycle and sexual differentiation of Cryptosporidium. To facilitate exploration of sexual reproduction in Cryptosporidium parvum, we developed an organoid-based culture system that supports Cryptosporidium's full life cycle and a novel fertilization reporter. Organoid-derived monol

The artemisinin-induced dormant stages of Plasmodium falciparum exhibit hallmarks of cellular quiescence/senescence and drug resilience.

Recrudescent infections with the human malaria parasite, Plasmodium falciparum, presented traditionally the major setback of artemisinin-based monotherapies. Although the introduction of artemisinin combination therapies (ACT) largely solved the problem, the ability of artemisinin to induce dormant parasites still poses an obstacle for current as well as future malaria chemotherapeutics. Here, we use a laboratory model for induction of dormant P. falciparum parasites and characterize their transcriptome, drug sensitivity profile, and cellular ultrastructure. We show that P. falciparum dormancy

LIFE CYCLE VARIATION AND HOST-PARASITE INTERACTIONS OF FOUR CONGENERIC SPECIES OF TURTLE ACANTHOCEPHALANS.

Turtle acanthocephalans in the genus Neoechinorhynchus have complex life cycles and are thought to involve an ostracod intermediate host and a turtle definitive host. For Neoechinorhynchus emydis, an additional snail paratenic host is part of its life cycle. However, how snails become infected and whether other species of Neoechinorhynchus can be found in snails remains unclear. This study was conducted to understand the survival, development, and host use of 4 species of turtle acanthocephalans (Neoechinorhynchus chrysemydis, N. emydis, Neoechinorhynchus emyditoides, and Neoechinorhynchus pse

Identification of three members of the multidomain adhesion CCp family in Babesia gibsoni.

Babesia gibsoni is an intraerythrocytic apicomplexan parasite transmitted by Haemaphysalis longicornis and causes canine babesiosis. Within the tick, the Babesia parasite undergoes sexual conjugation and the sporogony process of its life cycle. To control B. gibsoni infection, prompt and effective treatment of acute infections and curing chronic carriers are urgently needed. Gene disruption of Plasmodium CCps resulted in blocking the transition of sporozoites from the mosquito midgut to the salivary glands, showing that these proteins are potential targets for the development of a transmission

Healthcare waste in Bangladesh: Current status, the impact of Covid-19 and sustainable management with life cycle and circular economy framework.

COVID-19 has accelerated the generation of healthcare (medical) waste throughout the world. Developing countries are the most affected by this hazardous and toxic medical waste due to poor management systems. In recent years, Bangladesh has experienced increasing medical waste generation with estimated growth of 3 % per year. The existing healthcare waste management in Bangladesh is far behind the sustainable waste management concept. To achieve an effective waste management structure, Bangladesh has to implement life cycle assessment (LCA) and circular economy (CE) concepts in this area. Howe

Driving sustainable circular economy in electronics: A comprehensive review on environmental life cycle assessment of e-waste recycling.

E-waste, encompassing discarded materials from outdated electronic equipment, often ends up intermixed with municipal solid waste, leading to improper disposal through burial and incineration. This improper handling releases hazardous substances into water, soil, and air, posing significant risks to ecosystems and human health, ultimately entering the food chain and water supply. Formal e-waste recycling, guided by circular economy models and zero-discharge principles, offers potential solutions to this critical challenge. However, implementing a circular economy for e-waste management due to

Identification and molecular characterization of digenean trematode parasites of Aylacostoma chloroticum (Gastropoda: Thiaridae) from a Neotropical Basin.

Thiarids are pantropical gastropods distributed from South America to Australia and found in lentic and lotic freshwater environments. These gastropods act as intermediate hosts for several species of digeneans. There are difficulties in the correct identification of these digeneans in certain stages of the life cycle as the larvae have a reduced size and few morphological characteristics for diagnosis. Because of this, techniques based on molecular markers have become an extremely useful tool. Our objective was to molecularly characterize the digeneans found in gastropods from the Ivaí River,

A comparative life cycle assessment of anaerobic mono- and co-digestion of livestock manure in Bangladesh.

Proper management of biogenic residues, particularly livestock manure and food waste, is a major challenge for Bangladesh. While mono-digestion has traditionally been used on farms for treating manure, inadequate energetic output limits its applicability. Food waste, however, is typically landfilled in current practice. Co-digestion of biowaste emerged as an alternative due to synergistic yield and capacity to handle multiple waste streams. However, its environmental performance is underreported, particularly in developing countries. This study aimed to compare the environmental implications o

Performance of a green wall (Total Value Wall™) at high greywater loading rates and Life Cycle Impact Assessment.

Nature-based greywater (GW) treatment and reuse in urban areas has become an up-and-coming option. A 14.4 m2 green wall system called Total Value Wall (TVW) was installed at a terraced house in Gent (Belgium) for treating GW and reusing the effluent for toilet flushing. In a previous study, the TVW was loaded at 7 L.m-2.d-1 and efficiently removed TSS (67%), COD (43%), BOD5 (83%) and total coliforms (log 2), but a number of issues were reported related to nutrient leaching from the substrate, and the excessive retention time in the storage tanks. In this study results are reported from a follo

Adsorptive removal of some Cl-VOC's as dangerous environmental pollutants using feather-like γ-Al2O3 derived from aluminium waste with life cycle analysis.

Herein, we designed a cost-effective preparation method of nanocomposite γ-Al2O3 derived from Al-waste. The produced material has a feather-like morphology, and its adsorption of some chlorinated volatile organic compounds (Cl-VOC's) such as benzyl chloride, chloroform and carbon tetrachloride (C7H7Cl, CHCl3 and CCl4) was investigated due to their potential carcinogenic effect on humans. It showed a characteristic efficiency towards the adsorptive removal of these compounds over a long period, i.e., eight continuous weeks, at ambient temperature and atmospheric pressure. After 8-weeks, the ads

Annotation survey and life cycle transcriptomics of transcription factors in rust fungi (Pucciniales) identify a possible role for cold shock proteins in dormancy exit.

Fungi of the order Pucciniales are obligate plant biotrophs causing rust diseases. They exhibit a complex life cycle with the production of up to five spore types, infection of two unrelated hosts and an overwintering stage. Transcription factors (TFs) are key regulators of gene expression in eukaryote cells. In order to better understand genetic programs expressed during major transitions of the rust life cycle, we surveyed the complement of TFs in fungal genomes with an emphasis on Pucciniales. We found that despite their large gene numbers, rust genomes have a reduced repertoire of TFs comp

Differential expression profiles of miRNAs and their putative targets in Schistosoma mansoni during its life cycle.

BACKGROUND: Schistosomiasis is a disease caused by Schistosoma. Due to its complex life cycle, evolutionary position and sexual dimorphism, schistosomes have several mechanisms of gene regulation. MicroRNAs (miRNAs) are short endogenous RNAs that regulate gene expression at the post-transcriptional level by targeting mRNA transcripts. OBJECTIVES: Here, we tested 12 miRNAs and identified their putative targets using a computational approach. METHODS: We performed the expression profiles of a set of miRNAs and their putative targets during the parasite's life cycle by quantitative reverse tran

Life cycle process dependencies of positive-sense RNA viruses suggest strategies for inhibiting productive cellular infection.

Life cycle processes of positive-strand (+)RNA viruses are broadly conserved across families, yet they employ different strategies to grow in the cell. Using a generalized dynamical model for intracellular (+)RNA virus growth, we decipher these life cycle determinants and their dependencies for several viruses and parse the effects of viral mutations, drugs and host cell permissivity. We show that poliovirus employs rapid replication and virus assembly, whereas the Japanese encephalitis virus leverages its higher rate of translation and efficient cellular reorganization compared to the hepatit

Transcriptional ups and downs: patterns of gene expression in the life cycle of Toxoplasma gondii.

Toxoplasma gondii reproduces sexually in felines and asexually in virtually all warm-blooded animals, including humans. This obligate intracellular parasite alternates between biologically distinct developmental stages throughout its complex life cycle. Stage conversion is crucial for T. gondii transmission, persistence, and the maintenance of genetic diversity within the species. Genome-wide comparative transcriptomic studies have contributed invaluable insights into the regulatory gene networks underlying T. gondii development.
