Antimicrobial and antioxidant activities of the various extracts of Verbascum pinetorum Boiss. O. Kuntze (Scrophulariaceae).

BACKGROUND AND OBJECTIVES: The present study was performed to evaluate the in vitro antimicrobial and antioxidant properties of various extracts of Verbascum (V.) pinetorum, a member of Scrophulariaceae family. While the antimicrobial activity of various extracts of V. pinetorum was determined with agar-well diffusion method, the antioxidant activity was examined with two complementary test systems, namely 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging and beta-carotene/linoleic acid test systems. RESULTS: The hexane extract exhibits antimicrobial activity against few microorgan

Prostaglandin-H-synthase (PGHS)-1 and -2 microtiter assays for the testing of herbal drugs and in vitro inhibition of PGHS-isoenzyms by polyunsaturated fatty acids from Platycodi radix.

In order to test inhibition of prostaglandin-H-synthase-1 and -2 (PGHS-1 and -2) by plant extracts, we have established two enzyme based in vitro assays with enzyme immunoassay (EIA) evaluation. The assays have been evaluated with known synthetic inhibitors and with plant extracts. In a screening of traditionally used Chinese herbs for anti-inflammatory activity, a series of n-hexane and dichloromethane extracts showed significant inhibitory effect in comparison with the known specific PGHS-2 inhibitors NS-398 (IC(50) = 2.6 microM) and nimesulide (IC(50) = 36 microM). The lipophilic extracts o

Hypermethylation of Fads2 and altered hepatic fatty acid and phospholipid metabolism in mice with hyperhomocysteinemia.

Alterations in lipid metabolism may play a role in the vascular pathology associated with hyperhomocysteinemia (HHcy). Homocysteine is linked to lipid metabolism through the methionine cycle and the synthesis of phosphatidylcholine (PC) by phosphatidylethanolamine (PE) methyltransferase, which is responsible for the synthesis of 20-40% of liver PC. The goal of the present study was to determine if the reduced methylation capacity in HHcy is associated with alterations in liver phospholipid and fatty acid metabolism. Mice heterozygous for disruption of cystathionine beta-synthase (Cbs+/-) fed a

Selective reduction of bis(monoacylglycero)phosphate ameliorates the storage burden in a THP-1 macrophage model of Gaucher disease.

Bis(monoacylglycero)phosphate (BMP) assists lysosomal function by facilitating interaction of hydrolases and activator proteins with sphingolipid substrates. Impaired lysosomal degradation of the sphingolipid glucosylceramide (GC) occurs in Gaucher disease due to an inherited deficiency of acid β-glucosidase, with secondary BMP alterations. We investigated the nature of BMP accumulation and whether its correction reduced the storage burden in a THP-1 macrophage model of Gaucher disease. Using sucrose gradients and detergent solubility, 98% of BMP resided in the detergent-soluble membranes (DSM

The fatty acid composition of plasma and erythrocytes in Schistosoma mansoni-infected mice.

The fatty acid status of mice six weeks after infection with Schistosoma mansoni was investigated and compared with the status of uninfected mice. Fatty acids of several plasma and erythrocyte lipid fractions were determined by capillary gas-chromatography. Major fatty acids including linoleic acid, arachidonic acid and dihomo-gamma-linolenic acid, were significantly reduced in the plasma phospholipids of infected mice. Arachidonic acid was also reduced in plasma cholesterolesters. The degree of the plasma arachidonic acid reduction in infected animals correlated neither with the worm load nor

Plasma and red blood cell fatty acid composition in children with protein-calorie malnutrition.

Plasma and red cell fatty acids were measured in 44 children with third degree malnutrition: 22 children with marasmus, 11 children with kwashiorkor, and 11 with marasmic kwashiorkor (MK). The presence of edema, dermatosis, hair changes, and hypoalbuminemia were positively correlated with the red cell percentage linoleic acid and negatively correlated with red cell percentage arachidonic acid. Red cell percentage linoleic acid in kwashiorkor was decreased from the control value of 15.3% to 10.9%. In marasmus and MK, there were significantly greater decreases, to 7.7% and 8.0%, respectively. Re

Hydrogen abstraction from lipids by triplet states of derivatized benzophenone photosensitizers.

Laser photolysis techniques have been used to characterize the reactivity of triplet state lipoidal benzophenone derivatives toward fatty acids and glycerides in benzene solution. The reactivities of benzophenone-4-heptyl-4'-pentanoic acid (BHPA) toward fatty acid compounds having different configurations of olefinic bonds have been determined. The rates of hydrogen abstraction are found to be lower when compared with similar measurements using benzophenone alone. However, the contribution of physical quenching of the triplet derivative by double bonds also appears to be slightly lower than th

Undetected Association Between Fatty Acids and Dementia with Lewy Bodies: A Bidirectional Two-Sample Mendelian Randomization Study.

BACKGROUND: Although observational studies indicated connections between fatty acids (FAs) and Alzheimer's disease and dementia, uncertainty persists regarding how these relationships extend to dementia with Lewy bodies (DLB). OBJECTIVE: To explore the potential causal relationships between FAs and the development of DLB, thus clarifying these associations using genetic instruments to infer causality. METHODS: We applied a two-sample Mendelian randomization (MR) and multivariable Mendelian randomization (MVMR) approach. Genetic data were obtained from a DLB cohort, comprising 2,591 cases and

Evidence for Coupled Autoxidation of Coexisting Lipids in Aerosols Driven by Criegee Intermediates and OH Radicals.

Lipid oxidation driven by intermolecular interactions plays a pivotal role in human aging and indoor environmental quality, yet the underlying interaction mechanisms remain largely elusive. Here, we uncover a coupled autoxidation network in the heterogeneous co-ozonolysis of model skin lipids (cholesterol and fatty acids), using online atmospheric pressure photoionization coupled with an ultrahigh-resolution mass spectrometer. Individual cholesterol exhibited extremely low heterogeneous reactivity (determined as effective uptake coefficient, γeff, Chol = 2.94 × 10-6). However, upon co-ozonolys

Nitro-fatty acid signaling and intracellular biodistribution in plants.

Nitrated fatty acids (NO₂-FAs) are biologically active molecules formed by the reaction between unsaturated fatty acids and reactive species derived from nitric oxide (NO). They are characterised by their ability to release NO, their incorporation into complex lipids and their reactivity with nucleophiles such as cysteine and histidine, facilitating post-translational modifications by nitroalkylation. The main NO₂-FAs identified in plants are nitro-oleic acid (NO₂-OA), nitro-linoleic acid (NO₂-LA), nitro-conjugated linoleic acid (NO2-cLA) and nitro-linolenic acid (NO₂-Ln), whose concentration

Oxidized Lipids in Persistent Pain States.

Chemotherapy, nerve injuries, or diseases like multiple sclerosis can cause pathophysiological processes of persistent and neuropathic pain. Thereby, the activation threshold of ion channels is reduced in peripheral sensory neurons to normally noxious stimuli like heat, cold, acid, or mechanical due to sensitization processes. This leads to enhanced neuronal activity, which can result in mechanical allodynia, cold allodynia, thermal hyperalgesia, spontaneous pain, and may initiate persistent and neuropathic pain. The treatment options for persistent and neuropathic pain patients are limited; f

Control of brain fatty acids.

Saturated and monounsaturated fatty acids are mainly synthetized in the brain, but some of them could originate from the diet; in contrast polyunsaturated fatty acids are derived from dietary linoleic and linolenic acid. Saturated fatty acid biosynthesis occurs via three main pathways in mammalian cells. One is de novo synthesis of fatty acids from acetyl-CoA via malonyl-CoA; this system has been isolated in soluble form (the soluble system) from various animal tissues including brain. The second and third pathways involve elongation: in the mitochondrial system, acetyl CoA is the principal su

Biomarkers of dietary fat composition in young adults with a parental history of premature coronary heart disease compared with controls. The EARS Study.

The European Atherosclerosis Research Study (EARS) is a study of the biological expression of a paternal history of premature coronary heart disease. The study was carried out in 14 centres in 11 European countries, where the offspring of fathers who suffered a documented myocardial infarction before the age of 55 years (cases) were compared with age- and sex-matched controls. In this paper we describe the measurement of plasma cholesteryl esters as biomarkers for the fatty acid composition of the diet in the cases and controls. No significant differences were found.
