Biocatalytic transformation of Ganoderma lucidum by porcine pancreatic lipase in hydrophobic deep eutectic solvents.

Enzyme applications in industry are often limited by low stability, poor reusability and challenging recovery. This study presents a sustainable strategy for enhancing enzyme performance by immobilizing porcine pancreatic lipase (PPL) onto Ganoderma lucidum using a hydrophobic deep eutectic solvent (HDES) as a reaction medium to facilitate enzyme immobilization. Among several HDESs tested, menthol:decanoic acid (HDES 1) showed the highest lipase activity (198.38%) and was selected for immobilization. Under optimized conditions (3 h, 40 °C, pH 7.0), the immobilized PPL achieved 212.41% catalyti…

Tannic acid-crosslinked zein-lipase complex stabilized multichamber Pickering emulsion for efficient diacylglycerol production.

This study presents a food-grade, multi-chamber W/O/W high internal phase Pickering emulsion (HIPPE) system for interfacial catalysis, which stabilized by a zein nanoparticle (ZNP)-lipase complex crosslinked with tannic acid (TA). Experiments and molecular dynamics simulations confirmed that the cross-linking between TA, ZNPs, and lipase was predominantly mediated by hydrogen bonding, hydrophobic interactions, and π-π stacking. TA effectively modulated the zein-lipase interaction to form a stable, dispersed complex and substantially enhanced the thermal stability of the lipase. The multi-chamb…

Activity Enhancement of a Truncated Lipase-Phospholipase from the Marine Thraustochytrid Aurantiochytrium Sp. MG91.

Lipase/phospholipase (Lip/Plip) from thraustochytrids is a bifunctional enzyme capable of hydrolyzing both triglycerides and phospholipids and is characterized by an unusual 150-residue N-terminal domain whose function remains unclear. To elucidate the role of this region, we engineered an N-terminally truncated enzyme (S-270) and compared it with the full-length enzyme (L-420) from Aurantiochytrium sp. MG91 heterologously expressed in Escherichia coli. The present research is limited and directed toward comparing enzyme activities (specific activity, substrate selectivity, and growth-related …

Bioinformatics Analysis Identifies Sequence Determinants of Enzymatic Activity for the PHARC-Associated Lipase ABHD12.

In humans, PHARC (polyneuropathy, hearing loss, ataxia, retinitis pigmentosa, and cataract) is an early onset autosomal recessive neurological disorder caused by deleterious mutations to ABHD12 (α/β-hydrolase domain protein # 12). Biochemically, ABHD12 functions as a lipase and catalyzes the hydrolysis of lysophosphatidylserine (lyso-PS) (lyso-PS lipase). By doing so, it controls the concentrations and signaling pathways regulated by this potent signaling lysophospholipid in the mammalian brain. While genetic mapping efforts have identified over 30 mutations in ABHD12 from human PHARC subjects…

Bacterial lipase-responsive polydopamine nanoparticles for detection and synergistic therapy of wound biofilms infection.

Wound biofilms represent an elusive conundrum in contemporary treatment and diagnostic options, accredited to their escalating antibiotic resistance and interference in chronic wound healing processes. Here, we developed mesoporous polydopamine (mPDA) nanoparticles, and grafted with rhodamine B (Rb) as biofilm lipase responsive detection probe, followed by π - π stacking mediated ciprofloxacin (CIP) loading to create mP-Rb@CIP nanoparticles. mPDA NPs with a melanin structure could quench fluorescence emissions of Rb. Once encountering biofilm in vivo, the ester bond in Rb and mPDA is hydrolyze…

Unmasking crucial residues in adipose triglyceride lipase for coactivation with comparative gene identification-58.

Lipolysis is an essential metabolic process that releases unesterified fatty acids from neutral lipid stores to maintain energy homeostasis in living organisms. Adipose triglyceride lipase (ATGL) plays a key role in intracellular lipolysis and can be coactivated upon interaction with the protein comparative gene identification-58 (CGI-58). The underlying molecular mechanism of ATGL stimulation by CGI-58 is incompletely understood. Based on analysis of evolutionary conservation, we used site directed mutagenesis to study a C-terminally truncated variant and full-length mouse ATGL providing insi…

Monoacylglycerol Lipase Inhibition Using ABX-1431 Attenuates Cerebral Ischaemia Early After Traumatic Brain Injury.

An early event in the pathology of traumatic brain injury (TBI) is a reduction in cerebral blood flow (CBF), which exacerbates secondary injury development and inhibits brain recovery. The endogenous cannabinoid system signalling (eCBs) might be critical in TBI recovery due to modulating synaptic activity and exerting neuroprotective and anti-inflammatory effects. In the brain, eCBs predominantly occur at cannabinoid receptor type 1 via the eCB 2-arachidonoylglycerol (2-AG). The aim of this work was to test the efficacy of potentiating 2-AG signalling by monoacylglycerol lipase (MAGL) inhibiti…

Resolution of N-acetyl-DL-methionine methyl ester by the lipase from Brucella thiophenivorans.

In this study, lipase-catalyzed resolution of N-acetyl-DL-methionine methyl ester (N-Ac-DL-MetOMe) was evaluated. A lipase from Brucella thiophenivorans was prone to exhibit high activity and excellent enantioselectivity toward N-Ac-DL-MetOMe to produce the key chiral intermediate N-acetyl-L-methionine methyl ester (N-Ac-L-MetOMe). The results showed that the enzymatic reaction was carried out in 100 g/L racemic substrate for 2 h, the conversion reached 51.3%, the enantiomeric excess value N-Ac-L-MetOMe exceeded 99%, and the enantiomeric ratio value >200. Therefore, the lipase from B. thiophen…

Enhancing the Hydrolytic Activity of a Lipase towards Larger Triglycerides through Lid Domain Engineering.

Lipases have valuable potential for industrial use, particularly those mostly active against water-insoluble substrates, such as triglycerides composed of long-carbon chain fatty acids. However, in most cases, engineered variants often need to be constructed to achieve optimal performance for such substrates. Protein engineering techniques have been reported as strategies for improving lipase characteristics by introducing specific mutations in the cap domain of esterases or in the lid domain of lipases or through lid domain swapping. Here, we improved the lipase activity of a lipase (WP_07574…
