Dual role of HCN produced by the endophyte Pseudomonas urmiensis A32 in biocontrol and the defence response of pepper.

Plant-associated Pseudomonas species are widely recognised for their ecological versatility and potential as sustainable biocontrol and plant growth-promoting agents. Among their diverse metabolites, hydrogen cyanide (HCN) is traditionally considered a potent antimicrobial compound, yet its broader role in plant-microbe interactions remains underexplored. This study investigated Pseudomonas urmiensis A32, an endophytic bacterium previously shown to enhance drought tolerance and disease resistance in pepper (Capsicum annuum). Whole-genome analysis revealed genes involved in nutrient acquisition

Stanniocalcin-1 Overexpression Protects Porcine Intestinal Epithelial Cells Against TBHP-Induced Oxidative Stress by Preserving Mitochondrial Homeostasis.

Stanniocalcin-1 (STC-1) is a multifunctional glycoprotein; however, its role in protecting intestinal epithelial cells against oxidative injury has not been completely elucidated. This study investigated the cytoprotective effects and underlying molecular mechanisms of STC-1 overexpression in porcine intestinal epithelial (IPEC-J2) cells subjected to tert-butyl hydroperoxide (TBHP)-induced oxidative stress. IPEC-J2 cells were transfected with pcDNA3.1/STC-1 prior to TBHP challenge. STC-1 overexpression markedly rescued cells from TBHP-induced cytotoxicity and cell death, and was associated wit

Biomimetic dynamic nanomedicine SrHCF: Lattice self-reconstruction based on Fe2+ response to achieve synergistic "ferrous chelation-antioxidation-calcium antagonism-inflammation inhibition" quadruple treatment of CIRI.

Revascularization has revolutionized the treatment of ischemic stroke (IS), but its efficacy is limited by cerebral ischemia-reperfusion injury (CIRI). CIRI involves the dynamic interaction of multidimensional pathological mechanisms such as ferroptosis, calcium overload, oxidative stress, and subsequent inflammatory cytokine storm, which make many emerging single-target therapeutic strategies limited in alleviating CIRI. Strategies that simultaneously and synergistically target multiple key pathological factors in CIRI remain highly anticipated but extremely challenging. This study constructe

Genome-wide screening and expression of glutathione S-transferase genes reveal that GSTe4 contributes to sensitivity against β-cypermethrin in Zeugodacus cucurbitae.

Glutathione S-transferases (GSTs) are an essential multifunctional protein family with common detoxifying enzymes. In this study, 34 GST genes were identified from the melon fly, Zeugodacus cucurbitae, one of the most destructive pests worldwide. These GSTs include 32 cytosolic genes and two microsomal genes. Furthermore, these cytosolic GSTs were classified into six classes: 11 delta, 13 epsilon, three theta, one sigma, two zeta, and two omega. Most of these showed dynamic expression during the developmental stage, some of which showed stage-specific expression. The expression in various adul

Secreted neutral metalloproteases of Bacillus anthracis as candidate pathogenic factors.

To evaluate the pathogenic potential of Bacillus anthracis-secreted proteases distinct from lethal toxin, two neutral zinc metalloproteases were purified to apparent homogeneity from the culture supernatant of a non-virulent delta Ames strain (pXO1-, pXO2-). The first (designated Npr599) is a thermolysin-like enzyme highly homologous to bacillolysins from other Bacillus species. The second (designated InhA) is a homolog of the Bacillus thuringiensis immune inhibitor A. These proteases belong to the M4 and M6 families, respectively. Both enzymes digested various substrates, including extracellu

Molecular and biochemical characterization of 3-hydroxybenzoate 6-hydroxylase from Polaromonas naphthalenivorans CJ2.

Prior research revealed that Polaromonas naphthalenivorans CJ2 carries and expresses genes encoding the gentisate metabolic pathway for naphthalene. These metabolic genes are split into two clusters, comprising nagRAaGHAbAcAdBFCQEDJI'-orf1-tnpA and nagR2-orf2I''KL (C. O. Jeon, M. Park, H. Ro, W. Park, and E. L. Madsen, Appl. Environ. Microbiol. 72:1086-1095, 2006). BLAST homology searches of sequences in GenBank indicated that the orf2 gene from the small cluster likely encoded a salicylate 5-hydroxylase, presumed to catalyze the conversion of salicylate into gentisate. Here, we report physiol

Mutation of senataxin alters disease-specific transcriptional networks in patients with ataxia with oculomotor apraxia type 2.

Senataxin, encoded by the SETX gene, contributes to multiple aspects of gene expression, including transcription and RNA processing. Mutations in SETX cause the recessive disorder ataxia with oculomotor apraxia type 2 (AOA2) and a dominant juvenile form of amyotrophic lateral sclerosis (ALS4). To assess the functional role of senataxin in disease, we examined differential gene expression in AOA2 patient fibroblasts, identifying a core set of genes showing altered expression by microarray and RNA-sequencing. To determine whether AOA2 and ALS4 mutations differentially affect gene expression, we

XPF-ERCC1 acts in Unhooking DNA interstrand crosslinks in cooperation with FANCD2 and FANCP/SLX4.

DNA interstrand crosslinks (ICLs), highly toxic lesions that covalently link the Watson and Crick strands of the double helix, are repaired by a complex, replication-coupled pathway in higher eukaryotes. The earliest DNA processing event in ICL repair is the incision of parental DNA on either side of the ICL ("unhooking"), which allows lesion bypass. Incisions depend critically on the Fanconi anemia pathway, whose activation involves ubiquitylation of the FANCD2 protein. Using Xenopus egg extracts, which support replication-coupled ICL repair, we show that the 3' flap endonuclease XPF-ERCC1 co

The molecular basis of glutamate formiminotransferase deficiency.

Glutamate formiminotransferase deficiency, an autosomal recessive disorder and the second most common inborn error of folate metabolism, is presumed to be due to defects in the bifunctional enzyme glutamate formiminotransferase-cyclodeaminase (FTCD). Features of a severe phenotype, first identified in patients of Japanese descent, include elevated levels of formiminoglutamate (FIGLU) in the urine in response to histidine administration, megaloblastic anemia, and mental retardation. Features of a mild phenotype include high urinary excretion of FIGLU in the absence of histidine administration,

A DNA-Scaffolded Natural Killer Cell with Adaptable Multimodules for Enhanced Immunotherapy and Early Efficiency Reporting.

Adoptive cell therapy is promising with many clinical trials, but its application efficiency is significantly influenced by individual immune microenvironments and the varied responses of different patients. Equipping adoptive cells with multifunctional modules for function enhancement and real-time efficacy readouts is a key solution but remains challenging due to the dynamic nature of the cell membrane and the limited accessibility of available binding sites on the membrane. Herein, we report a multimodule intelligent NK (MI-NK) cell that encapsulates NK cells within a durable DNA scaffold a

Antibacterial gold nanoclusters: Protamine-coated, enzyme-inhibiting nanoparticles combat drug-resistant bacteria and accelerate wound healing.

Antimicrobial resistance (AMR) persists primarily due to the barrier effect of biofilms and the transient bactericidal activity of conventional antimicrobials, creating a demand for antimicrobial strategies independent of AMR mechanisms. To address this critical challenge, this study develops an innovative nanoplatform (Au@PS) by integrating protamine sulfate (PS) with gold nanoclusters. The synthesized Au@PS demonstrates broad-spectrum antimicrobial activity with significantly enhanced efficacy, leveraging the synergistic antimicrobial advantages of gold nanoclusters and PS. Specifically, PS

Exploration of 2D and 2.5D Conformational Designs Applied on Epoxide/Collagen-Based Integrative Biointerfaces with Device/Tissue Heterogeneous Affinity.

Collagen and multifunctional epoxides, which are respectively the common constituents of natural and polymer interfaces, were combined to fabricate integrative biointerfaces with device/tissue heterogeneous affinity. Further, the traditional 2D and advanced 2.5D conformational designs were achieved on collagen-based biointerfaces. The 2D conformational biointerfaces were formed by the self-entanglement of collagen molecules based on extensive hydrogen bonds among molecules, and the lamellar structures of 2D conformational biointerfaces could act as barriers to protect both biointerfaces and su
