S-nitrosothiols (SNOs) such as S-nitroso-L-cysteine (L-cysNO) are endogenous compounds with potent vasodilatory activity. During circulation in the blood, the NO moiety can be exchanged among various thiol-containing compounds by S-transnitrosylation, resulting in SNOs with differing capacities to enter the cell (membrane permeability). To determine whether the vasodilating potency of SNOs is dependent upon membrane permeability, membrane-permeable L-cysNO and impermeable S-nitroso-D-cysteine (D-cysNO) and S-nitroso-glutathione (GSNO) were infused into one femoral artery of anesthetized adult

AIMS: Arterial hypertension is a major risk factor for cardiovascular diseases. The kidney and its natriuretic function are in the centre of the prevailing models to explain the pathogenesis of hypertension; however, the mechanisms underlying blood pressure elevation remain unclear in most patients. Development of hypertension is strongly correlated with age, and this blood pressure increase typically accelerates in the fourth decade of life. The cause of age-dependent blood pressure elevation is poorly understood. This study aims to understand the role of procontractile G-protein-mediated sig

Cyclic AMP affects microvascular smooth muscle contraction and growth. Therefore, it is important to elucidate mechanisms regulating cyclic AMP production in microvascular smooth muscle. In this study, we determined whether several signal transduction pathways regulate receptor-induced cyclic AMP in isolated preglomerular microvessels and microvascular smooth muscle cells. Preglomerular microvessels were incubated with isoproterenol (beta-adrenoceptor agonist) and with and without U73122 (phospholipase C inhibitor), GF109203X (protein kinase C inhibitor), 1-butanol (phospholipase D inhibitor),

OBJECTIVES: The main function of myenteric neurons is the control of gut motility. As we recently showed that nitroxyl (HNO) induces intestinal smooth muscle relaxation, it was of interest to evaluate the effects of this signalling molecule on myenteric neurons in order to distinguish its properties in regard to myocytes.
METHODS: Myenteric neurons isolated from the ileum of 4-10 days old rats were used. HNO-induced changes in intracellular concentration of Ca2+ or membrane potential and ion currents were measured using the Ca2+-sensitive fluorescent dye fura-2 AM or by electrophysiological w

Endothelium-dependent vasorelaxation is partly mediated by small-conductance (SK3) and intermediate-conductance Ca2+ -activated K+ channels (SK4) in the endothelium that results in endothelium-dependent hyperpolarization (EDH). Apart from the electrical propagation through myoendothelial gap junctions, the K+ released from the endothelium facilitates EDH by increasing inward rectifier K+ channel (Kir) conductance in smooth muscle cells. The EDH-dependent relaxation of coronary artery (CA) and Kir current in smooth muscle cells (CASMCs) of hypertensive animals are poorly understood despite the

Calixarenes, owing to the ability to form supramolecular complexes with biologically important molecules and ions, can influence a course of biochemical processes and, accordingly, be considered as perspective molecular platforms for creation of physiologically active compounds. The work purpose is to study calixarene C-91 influence on systems of active Ca ions transport which are localized in subcellular membrane structures (mitochondria, sarcoplasmic reticulum, plasma membrane) of myometrial cells. It has been shown, that calixarene C-91 addition to incubation medium led to an increase in Ca

Asthma is a chronic respiratory condition characterized by airway inflammation, remodeling, and hyperresponsiveness to triggers that lead to airway constriction and impaired airflow. Bronchial smooth muscle (BSM) plays a central role in these processes by constricting the airways and producing proinflammatory cytokines in response to environmental triggers, allergens, and cytokines. Although current therapies, including bronchodilators, corticosteroids, and biologics, effectively treat many patients, additional strategies are needed for difficult-to-treat asthma. Emerging evidence suggests tha

AIMS: Myogenic urinary retention is a common urological disorder characterized by diminished detrusor contractility, reduced muscle mass, and disorganized muscle bundles, yet its molecular mechanisms are poorly defined. The adaptor protein PINCH2 is abundant in smooth muscle. This study aimed to determine whether the loss of PINCH2 induces myogenic urinary retention.
MATERIALS AND METHODS: C57BL/6 wild-type (WT) and PINCH2 global knockout (KO) mice were used for urodynamic assessments, post-void residual (PVR) measurement, and detrusor smooth muscle (DSM) strip electrical field stimulation (E

Smooth muscle cells (SMCs) play an important role in physiology and production in farm animals such as pigs. Here, we report the generation of a pig SMC line. Our original objective was to establish an enteroendocrine cell line from the pig ileum epithelium through lentiviral transduction of the Simian Virus (SV) 40 large T antigen. However, an initial expression analysis of marker genes in nine cell clones revealed that none of them were enteroendocrine cells or absorptive enterocytes, goblet cells, or Paneth cells, some of the major cell types existing in the ileum epithelium. A more detaile

Moderate hyperoxia (30%-60% O2) in premature infants promotes bronchial airway hyperresponsiveness (AHR) via airway smooth muscle (ASM), a key regulator of bronchoconstriction, bronchodilation, and remodeling. Understanding how O2 exposure drives long-term bronchial changes in prematurity is critical for developing therapies for airway disease across the lifespan. Premature lungs have immature antioxidant defenses, potentially due to disrupted mitochondrial dynamics, increasing susceptibility to O2-induced oxidative stress. Thus, mitochondrial homeostasis is highly relevant to ASM dysfunction

BACKGROUND: Diabetes constitutes a risk factor for atherosclerotic calcification, which is highly associated with phenotypic switching in vascular smooth muscle cells (VSMCs). Protein cysteine S-nitrosylation plays a crucial role in multiple cardiovascular diseases. The objective of this study is to examine whether diabetic atherosclerotic calcification is regulated by S-nitrosylation of AMP-activated protein kinase (AMPK), a regulator of VSMC phenotype switching.
METHODS: The atherosclerotic plaque was induced by feeding Apoe-/- mice a high-fat diet. A low dose of streptozotocin (STZ) was us

Aortic dissection (AD) is a devastating vascular disorder pathologically characterized by pathological phenotypic switching of medial vascular smooth muscle cells (VSMCs) and progressive breakdown of extracellular matrix (ECM) architecture. Conventional paradigms have primarily emphasized unidirectional injury originating within the vessel wall. Nevertheless, accumulating evidence indicates that perivascular adipose tissue (PVAT), which directly envelops the adventitia and is increasingly regarded as the "fourth layer" of the vascular wall, exerts a critical outside-in influence on vascular ho

Salvia officinalis L. (S. officinalis) or common sage has long been valued in traditional medicine for its broad therapeutic properties. Our study explored the antioxidant potential, the cytotoxic activity and the effects on smooth muscle (SM) contractility of a traditionally prepared sage aqueous infusion. A chromatographic analysis revealed the major phenolic constituents of the infusion, rosmarinic acid being the most abundant compound (38.16 mg/mL), followed by rutin (3.58 mg/g), caffeic acid (3.00 mg/g) and hesperidin (1.84 mg/g). Three spectrophotometric methods were used to determine th

Phenotypic switching of vascular smooth muscle cells (VSMCs) from the contractile to the synthetic is a key factor driving the formation of intracranial atherosclerotic stenosis (ICAS); however, the upstream mitochondrial mechanisms underlying this process and its therapeutic implications remain poorly defined. This study aims to investigate the role and underlying molecular mechanisms of the mitochondria-targeted hydrogen sulfide (H2S) donor AP39 in regulating mitochondrial function during VSMCs phenotypic switching and neointimal hyperplasia. The best way of treating vasospastic angina pharmacologically is to induce vasorelaxation under hypoxia only. We previously established a new method to quantify the contraction and relaxation of vascular smooth muscle cells (VSMCs) in real time. In the present study, we used this method to screen the effects of chemicals on VSMC dilation under normoxia, and we chose the chemicals with negative results as candidates. Next, we tested the effects of the candidates on VSMC dilation under hypoxia, and we found that cariporide and empagliflozin induced VSMC relaxation under hypoxia but not normoxia.

BACKGROUND: The development of thoracic aortic dissection (TAD) is closely associated with the loss of vascular smooth muscle cells (VSMCs). Androgen receptor (AR) signaling has increasingly been recognized as an important regulator of cell death in prostate cancer. However, the role of AR signaling in the development of TAD in men remains unknown.
METHODS: The expression of AR was analyzed in clinical specimens obtained from TAD patients undergoing surgical aortic replacement and control subjects receiving heart transplantation. Using β-aminopropionitrile (BAPN)-induced aortic dissection mou

Cerebral vascular deposition of the amyloid-β (Aβ) peptide, a condition known as cerebral amyloid angiopathy (CAA), is associated with intracerebral hemorrhaging and contributes to disease progression in Alzheimer's disease (AD) and vascular cognitive impairment and dementia (VCID). Familial mutations at positions 22 and 23 within the Aβ peptide lead to early onset and severe CAA pathology. Here, we evaluate the effects of fibrillar Aβ peptides on the viability of primary-cultured human cerebral smooth muscle (HCSM) cells, which are the major site of amyloid deposition in cerebral blood vessel

BACKGROUND: The impairment of vascular smooth muscle cells (VSMCs) causes many vascular diseases, such as atherosclerosis, hypertension, and heart conditions. Research has shown that N6-methyladenosine (m6A) modification regulates VSMCs' function in a vascular environment.
SUMMARY: By regulating RNA metabolism, m6A modification affects key biological processes in VSMCs, such as proliferation, migration, phenotypic transformation, and apoptosis. We aim to examine the role of m6A modification and its associated enzymes in vascular diseases caused by VSMC dysfunction and explore its potential as

Vascular smooth muscle cells (SMCs) can transition between a quiescent contractile or "differentiated" phenotype and a "proliferative-dedifferentiated" phenotype in response to environmental cues, similar to what in occurs in the wound healing process observed in fibroblasts. When dysregulated, these processes contribute to the development of various lung and cardiovascular diseases such as Chronic Obstructive Pulmonary Disease (COPD). Long non-coding RNAs (lncRNAs) have emerged as key modulators of SMC differentiation and phenotypic changes. In this study, we examined the expression of lncRNA

Neoatherosclerosis (NA) within stents has become an important clinical problem after coronary artery stent implantation. In-stent restenosis and in-stent thrombosis are the two major complications following coronary stent placement and seriously affect patient prognosis. As the common pathological basis of these two complications, NA plaques, unlike native atherosclerotic plaques, often grow around residual oxidized lipids and stent struts. The main components are foam cells formed by vascular smooth muscle cells (VSMCs) engulfing oxidized lipids at lipid residue sites. Current research mainly
