Central neural control of esophageal motility: a review.

We review recent studies on the central neural control of esophageal motility, emphasizing the anatomy and chemical coding of esophageal pathways in the spinal cord and medulla. Sympathetic innervation of the proximal esophagus is derived primarily from cervical and upper thoracic paravertebral ganglia, whereas that of the lower esophageal sphincter and proximal stomach is derived from the celiac ganglion. In addition to noradrenaline, many sympathetic fibers in the esophagus contain neuropeptide Y (NPY), and both noradrenaline and NPY appear to decrease blood flow and motility. Preganglionic

The olivocerebellar projection in the cat as studied with the method of retrograde axonal transport of horseradish peroxidase. II. The projection to the uvula.

Following injections of small quantities of horseradish peroxidase (HRP) suspension in the uvula of the cat, the distribution of labeled cells in the inferior olive has been mapped. The findings confirm the conclusion made on the basis of studies of retrograde cell loss in the olive following ablations of the uvula (Brodal, '40b) that two small olivary subdivisions, the nucleus beta and the dorsomedial cell column project heavily to the uvula. In addition the HRP-study shows that the uvula receives a smaller number of fibers from two circumscribed areas of the contralateral medial accessory ol

The periaqueductal gray and its role in the neural control of lower urinary tract function.

The periaqueductal gray (PAG) in the midbrain is recognized as a critical relay in lower urinary tract control, integrating ascending bladder signals with descending cortical and subcortical inputs, to coordinate micturition regulation. Animal studies reveal that coordinated pathways within the columnar organization of the PAG converge on the pontine micturition center (PMC), initiating or suppressing micturition. The ventrolateral column, in particular, exhibits robust glutamatergic projections to the PMC that facilitate bladder contractions, whereas local gamma-aminobutyric acid (GABA) inter

[Structure and Function of Thalamocortical Circuits Underlying Alloparental Behaviors].

The thalamus is widely recognized as a major source of input to the cerebral cortex and extensive knowledge of its anatomical connectivity has been accumulated through classical tracer studies. However, the quantitative characteristics of the thalamocortical connections and their physiological functions remain poorly understood. This issue is particularly important for higher-order cortical areas, such as the prefrontal cortex, which lack the prominent layer-4 bottom-up input characteristics of sensory cortices, raising the unresolved question of what types of inputs underlie higher cognitive

Brainstem Circuits for Rhythm Generation and Coordination of Orofacial Movements.

The head serves as a multifunctional nexus through which animals engage with the external world-gathering sensory input, breathing, feeding, and producing communication signals. These diverse functions must be executed with constrained anatomical resources, often requiring the same oropharyngeal structures to participate in multiple behaviors. Such overlap demands precise coordination to prevent catastrophic errors such as choking or aspiration, while ensuring that breathing and eating proceed without conflict. Operating largely outside conscious awareness, brainstem circuits choreograph these

Terra Incognita - Contributions of the Olivo-Cerebellar System to Autism Spectrum Disorder.

The inferior olivary nuclei (ION), a key component of the olivo-cerebellar system, remain understudied in autism spectrum disorder (ASD) research despite evident cerebellar involvement. This perspective piece aims to elucidate the critical role of the ION in cerebellar microcircuitry and its potential implications in ASD pathophysiology. We review the olivo- cerebellar system and the structural and functional alterations of the ION in autism, highlighting findings from neuroanatomical, neuroimaging, and behavioral studies. ION disruptions, although underexplored, may have a major role in the s

Local and long-range input balance: A framework for investigating frontal cognitive circuit maturation in health and disease.

Frontal cortical circuits undergo prolonged maturation across childhood and adolescence; however, it remains unknown what specific changes are occurring at the circuit level to establish adult cognitive function. With the recent advent of circuit dissection techniques, it is now feasible to examine circuit-specific changes in connectivity, activity, and function in animal models. Here, we propose that the balance of local and long-range inputs onto frontal cognitive circuits is an understudied metric of circuit maturation. This review highlights research on a frontal-sensory attention circuit

Multi-timescale neural dynamics for multisensory integration.

Carrying out any everyday task, be it driving in traffic, conversing with friends or playing basketball, requires rapid selection, integration and segregation of stimuli from different sensory modalities. At present, even the most advanced artificial intelligence-based systems are unable to replicate the multisensory processes that the human brain routinely performs, but how neural circuits in the brain carry out these processes is still not well understood. In this Perspective, we discuss recent findings that shed fresh light on the oscillatory neural mechanisms that mediate multisensory inte

Differential Brain Structural and Functional Patterns in Crohn's Disease Patients are Associated with Different Disease Stages.

BACKGROUND: Crohn's disease (CD) is an inflammatory, chronic disorder that alternates between a quiescent phase and inflammatory flare-ups. Research has begun to elucidate the impact of CD in modulating brain structure and function. The previous neuroimaging studies mainly involved CD patients in remission (CD-R); therefore, little is known about how inflammation influences brain-related features in different stages of the disease. We carried out a magnetic resonance imaging (MRI) study to explore whether the different levels of disease activity may differentially affect brain structure and fu

The non-specific matrix thalamus facilitates the cortical information processing modes relevant for conscious awareness.

The neurobiological mechanisms of arousal and anesthesia remain poorly understood. Recent evidence highlights the key role of interactions between the cerebral cortex and the diffusely projecting matrix thalamic nuclei. Here, we interrogate these processes in a whole-brain corticothalamic neural mass model endowed with targeted and diffusely projecting thalamocortical nuclei inferred from empirical data. This model captures key features seen in propofol anesthesia, including diminished network integration, lowered state diversity, impaired susceptibility to perturbation, and decreased corticoc

Cerebellar Contributions to the Basal Ganglia Influence Motor Coordination, Reward Processing, and Movement Vigor.

Both the cerebellum and the basal ganglia are known for their roles in motor control and motivated behavior. These two systems have been classically considered as independent structures that coordinate their contributions to behavior via separate cortico-thalamic loops. However, recent evidence demonstrates the presence of a rich set of direct connections between these two regions. Although there is strong evidence for connections in both directions, for brevity we limit our discussion to the better-characterized connections from the cerebellum to the basal ganglia. We review two sets of such

Frequency dependent emotion differentiation and directional coupling in amygdala, orbitofrontal and medial prefrontal cortex network with intracranial recordings.

The amygdala, orbitofrontal cortex (OFC) and medial prefrontal cortex (mPFC) form a crucial part of the emotion circuit, yet their emotion induced responses and interactions have been poorly investigated with direct intracranial recordings. Such high-fidelity signals can uncover precise spectral dynamics and frequency differences in valence processing allowing novel insights on neuromodulation. Here, leveraging the unique spatio-temporal advantages of intracranial electroencephalography (iEEG) from a cohort of 35 patients with intractable epilepsy (with 71 contacts in amygdala, 31 in OFC and 4

Multiscale neural gradients reflect transdiagnostic effects of major psychiatric conditions on cortical morphology.

It is increasingly recognized that multiple psychiatric conditions are underpinned by shared neural pathways, affecting similar brain systems. Here, we carried out a multiscale neural contextualization of shared alterations of cortical morphology across six major psychiatric conditions (autism spectrum disorder, attention deficit/hyperactivity disorder, major depression disorder, obsessive-compulsive disorder, bipolar disorder, and schizophrenia). Our framework cross-referenced shared morphological anomalies with respect to cortical myeloarchitecture and cytoarchitecture, as well as connectome…

Joint hub identification for brain networks by multivariate graph inference.

Recent developments in neuroimaging allow us to investigate the structural and functional connectivity between brain regions in vivo. Mounting evidence suggests that hub nodes play a central role in brain communication and neural integration. Such high centrality, however, makes hub nodes particularly susceptible to pathological network alterations and the identification of hub nodes from brain networks has attracted much attention in neuroimaging. Current popular hub identification methods often work in a univariate manner, i.e., selecting the hub nodes one after another based on either heuri

What's your name again? A review of the superior longitudinal and arcuate fasciculus evolving nomenclature.

Studies of the superior longitudinal fasciculus (SLF) have multiplied in recent decades owing to methodological advances, but the absence of a convention for nomenclature remains a source of confusion. Here, we have reviewed existing nomenclatures in the context of the research studies that generated them and we have identified their agreements and disagreements. A literature search was conducted using PubMed/MEDLINE, Web-of-Science, Embase, and a review of seminal publications, without restrictions regarding publication date. Our search revealed that diffusion imaging, autoradiography, and fi

Prefrontal cortex circuits in depression and anxiety: contribution of discrete neuronal populations and target regions.

Our understanding of depression and its treatment has advanced with the advent of ketamine as a rapid-acting antidepressant and the development and refinement of tools capable of selectively altering the activity of populations of neuronal subtypes. This work has resulted in a paradigm shift away from dysregulation of single neurotransmitter systems in depression towards circuit level abnormalities impacting function across multiple brain regions and neurotransmitter systems. Studies on the features of circuit level abnormalities demonstrate structural changes within the prefrontal cortex (PFC

Mapping the community structure of the rat cerebral cortex with weighted stochastic block modeling.

The anatomical architecture of the mammalian brain can be modeled as the connectivity between functionally distinct areas of cortex and sub-cortex, which we refer to as the connectome. The community structure of the connectome describes how the network can be parsed into meaningful groups of nodes. This process, called community detection, is commonly carried out to find internally densely connected communities-a modular topology. However, other community structure patterns are possible. Here we employ the weighted stochastic block model (WSBM), which can identify a wide range of topologies, t

Disrupted striatal neuron inputs and outputs in Huntington's disease.

Huntington's disease (HD) is a hereditary progressive neurodegenerative disorder caused by a CAG repeat expansion in the gene coding for the protein huntingtin, resulting in a pathogenic expansion of the polyglutamine tract in the N-terminus of this protein. The HD pathology resulting from the mutation is most prominent in the striatal part of the basal ganglia, and progressive differential dysfunction and loss of striatal projection neurons and interneurons account for the progression of motor deficits seen in this disease. The present review summarizes current understanding regarding the pro

A natural history of skills.

The dorsal pallium (a.k.a. cortex in mammals) makes a loop circuit with the basal ganglia and the thalamus known to control and adapt behavior but the who's who of the functional roles of these structures is still debated. Influenced by the Triune brain theory that was proposed in the early sixties, many current theories propose a hierarchical organization on the top of which stands the cortex to which the subcortical structures are subordinated. In particular, habits formation has been proposed to reflect a switch from conscious on-line control of behavior by the cortex, to a fully automated

Consensus Paper: Towards a Systems-Level View of Cerebellar Function: the Interplay Between Cerebellum, Basal Ganglia, and Cortex.

Despite increasing evidence suggesting the cerebellum works in concert with the cortex and basal ganglia, the nature of the reciprocal interactions between these three brain regions remains unclear. This consensus paper gathers diverse recent views on a variety of important roles played by the cerebellum within the cerebello-basal ganglia-thalamo-cortical system across a range of motor and cognitive functions. The paper includes theoretical and empirical contributions, which cover the following topics: recent evidence supporting the dynamical interplay between cerebellum, basal ganglia, and co
