SSUH2 Promotes Odontogenic Differentiation of SCAPs via the FOXM1/PDK1-Mediated Regulation of Mitochondrial Function.

AIM: Regenerative endodontics focuses on the restoration of the pulp-dentine complex by promoting odontogenic differentiation of stem cells of the apical papilla (SCAPs). Although SSUH2 has been implicated in developmental processes and dentine dysplasia type I (DD-I), the specific role of SSUH2 in regulating SCAPs differentiation remains unclear. In this study, we identified SSUH2 as a novel biomarker for SCAPs odontogenic differentiation and revealed its critical regulatory mechanism. METHODOLOGY: Immunohistochemical staining of mouse mandibular first molar sections and immunofluorescence s

Association between third molar agenesis and dental maturity in Brazilian children.

The aim of the current study is to investigate if third molar agenesis is associated with dental maturity variations in Brazilian children. Radiographs of children from 10 to 15 years old were examined and patients with 32 teeth were included in the control group and patients with at least one third molar agenesis were included in the agenesis group. Demirjian's method was used to calculate dental age and evaluate dental maturity. Dental age was calculated using the Dental Age mobile app. The dental maturity was determined by calculating the difference between dental age and chronological age

Delayed Dental Development in Children With Non-Syndromic Hypodontia: A Cross-Sectional Study Using a Machine Learning Approach to Dental Age Estimation.

OBJECTIVES: To investigate the influence of non-syndromic hypodontia on radiographic dental development and to estimate dental age in children with bilateral mandibular agenesis using a machine learning approach. MATERIALS AND METHODS: This retrospective cross-sectional study included 626 children aged 6-15 years (311 with dental agenesis, 315 matched controls). Dental age (DA) was assessed using the original Demirjian method. In cases with bilateral congenitally missing teeth in the mandible, DA was estimated using supervised machine learning models based on specific random forests, followin

Differences in osteogenic and odontogenic differentiation potential of DPSCs and SHED.

OBJECTIVE: Dental pulp stem cells (DPSCs) and stem cells from human exfoliated deciduous teeth (SHED) are types of human dental tissue-derived mesenchymal stem cells (MSCs) that have emerged as an interesting and promising source of stem cells in the field of tissue engineering. The aim of this work is to isolate stem cells from DPSCs and SHED, cultivate them in vitro and compare their odontogenic differentiation potential. METHODS: DPSCs and SHED were extracted from molars, premolars and canines of six healthy subjects aged 5-29 years. The cells were characterized, using flow cytometry, for

Effect of lactoferrin on odontogenic differentiation of stem cells derived from human 3rd molar tooth germ.

Stem cell technology has been a great hope for the treatment of many common tissue regeneration-related diseases. Therefore, the main challenge in hard tissue engineering is to make a successful combination of stem cells and efficient inductors such as biomaterials or growth factors, in the concept of stem cell conversion into odontogenic cell. Even though lactoferrin has been reported to promote bone growth in vivo, the molecular mechanism of teeth formation has not been elucidated yet. Different concentrations of lactoferrin were prepared for the analysis of cell toxicity and differentiation

Mechanism and control of tooth eruption: overview and clinical implications.

OBJECTIVES: To review pre- and post-emergent eruption, with particular emphasis on distinguishing isolated molar ankylosis from primary failure of eruption (PFE) and genetic considerations in eruption problems. MATERIAL AND METHODS: Radiographic review of eruption failure patients; animal and human experiments; high precision observations of movements of erupting teeth. RESULTS: In pre-emergent tooth eruption, the controlling element is the rate of resorption of overlying structures. A path is cleared, and then the erupting tooth moves along it. This has clinical importance in recognizing th

Sequential development of multiple supplemental premolars. Four-year follow-up report.

Multiple supernumerary teeth are usually associated with various syndromes, but they are relatively uncommon as an isolated dental anomaly. The presence of multiple supplemental premolars in all four quadrants and especially in the maxilla is rare. This report presents a unique case of multiple supplemental premolars in all four quadrants that were distributed unevenly (two erupted; five unerupted). A series of radiographs taken over four years demonstrated further sequential developing bilateral supplemental premolars in the maxilla, where they are considered the rarest. Appropriate managemen

Exposure to oral uranyl nitrate delays tooth eruption and development.

The risk of oral exposure to uranium potentially involves the population at large. Tooth eruption and development are ongoing processes that begin during fetal development and continue until the age of 18 y. Since one of the mechanisms involved in tooth eruption is bone formation and it is well documented that uranium inhibits bone formation, the aim of the present work was to study the effect of oral administration of uranyl nitrate (UN) on tooth eruption and development. Wistar rats aged 1 and 7 d were orally administered a single dose of 90 mg kg(-1) body weight of uranyl nitrate. Two age m

The heritability of malocclusion: Part 1--Genetics, principles and terminology.

The relative contribution of genes and the environment to the aetiology of malocclusion has been a matter of controversy throughout the twentieth century. Genetic mechanisms are clearly predominant during embryonic craniofacial morphogenesis, but environment is also thought to influence dentofacial morphology postnatally, particularly during facial growth. Orthodontic and orthopaedic techniques are used in the treatment of malocclusion and other dentofacial deformities, but with limited effectiveness. The key to the determination of the aetiology of malocclusion, and its treatability lies in t

Infantile osteopetrosis: a case report on dental findings.

In this paper we have described the case of a 7-yr-old Moroccan osteopetrotic boy, who had received a bone marrow transplant (BMT). He was transplanted from his older brother and, despite immunosuppressive therapy, developed chronic graft-versus-host disease and was placed on corticotherapy. Seven months after the bone marrow transplant, graft versus host disease (GVHD) was stabilized, but corticotherapy had inhibited growth. There was evidence of normalizing bone, his hearing was better but he had not recovered vision. Dental findings before the bone marrow transplant revealed some missing te

Presence of alpha-naphthyl acetate esterase enzymes in dental and oral epithelium of guinea pig embryos.

This study examines the distribution of alpha-naphthyl acetate esterase enzymes in reduced enamel epithelium, i.e., post secretory ameloblasts (PSA) and external cells of reduced enamel epithelium (ERE) of continuously growing incisor and molar tooth germs and in the covering oral epithelium (OE). Jaws of guinea pig embryos, 25-50 days of gestation age, were pretreated, frozen, serially cut, and incubated with alpha-naphthyl acetate as substrate and hexazotized pararosaniline as capture agent for demonstration of enzyme activity. In addition, sections were preincubated with various inhibitors

Associations of some variables to tooth formation in children with isolated cleft palate.

The timing of tooth formation in 251 children, affected with isolated cleft palate without syndromes or other concomitant visceral anomalies (CP), was evaluated from orthopantomograms. The age range was 6-12 yr. The formation of the permanent teeth was delayed approximately 0.7 yr in CP children compared with the data on the non-cleft reference group. There were no differences in sex distribution between the CP group and the non-cleft reference group and between the CP subgroups. No significant differences in tooth formation could be detected between the subgroups; with and without positive fa
