Lantadene A toxicity in sheep. A model for cholestasis.

Liver injury occurred after sheep were injected intravenously with the triperpene acid lantadene A, the toxic principle of the tropical plant Lantana camara. A single dose of 1--3 mg/kg of the compound caused mild hepatocellular injury characterized by transient rises in serum enzymes, with or without hyperbilirubinaemia. Higher doses resulted in hepatic necrosis. When low doses of the triterpene were given repeatedly over several days a cholestatic syndrome developed which appeared identical with that caused by consumption of the plant. This is a disease model which may be of value in experim

Extracts from Pygeum africanum and other ethnobotanical species with antiandrogenic activity.

Extracts from Pygeum africanum, Serenoa repens and Cucurbita pepo are used in the treatment of benign prostatic hyperplasia (BPH) and prostate cancer (PCa). The activity of the androgen receptor (AR) is known to control growth of the prostate. Here, we examined extracts of these plants for their antiandrogenic activity using an AR responsive reporter gene assay for drug discovery. A selective dichloromethane extract from the stem barks of Pygeum africanum revealed the highest antiandrogenic effect. Bioactivity-directed fractionation of this extract led to the isolation of N-butylbenzenesulfona

[Synthesis and antitumor activity of cyclopropane derivatives of betulinic and betulonic acids].

New cyclopropane derivatives of betulin were synthesized by attachment of dichlorocarbenes or dibromocarbenes to the double bond of betulin diacetate, followed by the deprotection of hydroxyl groups. The betulin cyclopropane derivative was obtained from 20,29-dihydro-20,29-dichloromethylenebetulin by treatment with lithium in tert-butanol. These compounds were converted into the corresponding derivatives of betulonic acid by oxidation with chromium trioxide. The reduction of oxo group with sodium borohydride led to the corresponding derivatives of betulinic acid. 20,29-Dihydro-20,29-dichlorome

Targeting glutathione with the triterpenoid CDDO-Im protects against benzo-a-pyrene-1,6-quinone-induced cytotoxicity in endothelial cells.

Epidemiological studies have exhibited a strong correlation between exposure to air pollution and deaths due to vascular diseases such as atherosclerosis. Benzo-a-pyrene-1,6-quinone (BP-1,6-Q) is one of the components of air pollution. This study was to examine the role of GSH in BP-1,6-Q mediated cytotoxicity in human EA.hy96 endothelial cells and demonstrated that induction of cellular glutathione by a potent triterpenoid, CDDO-Im (1-[2-cyano-3-,12-dioxooleana-1,9(11)-dien-28-oyl]imidazole), protects cells against BP-1,6-Q induced protein and lipid damage. Incubation of EA.hy926 endothelial

Extreme drought drives contrasting fates of labile and recalcitrant mangrove soil organic matter.

Mangroves are globally important blue carbon ecosystems, yet increasingly threatened by extreme climate events. We investigated sedimentary lipid biomarkers (n-alkanes, n-alcohols, sterols, triterpenoids, triterpenones) across tidal flat, Rhizophora mangle, and Avicennia schaueriana zones in the Piraquê-Açu and Piraquê-Mirim estuarine system (SE Brazil) during the 2015-2016 M-drought. High molecular weight n-alkanes (C29 dominance) and alcohols (C28-C30) indicated persistent vascular plant inputs, supported by elevated Carbon Preference Index values (CPILC = 2.0-4.6; CPIOH = 12.8-16.9).
