A century of andrology in Cell & Tissue Research: looking back while moving forward.

This article commemorates the 100th anniversary of the first issue of Cell & Tissue Research (CTR), the longest-running active journal dedicated to cell biology. Reflecting the significant contributions of spermatology and embryology to the early days of cell biology, the majority of articles in CTR's inaugural issue centered on plant and animal sperm cells. A brief synopsis of these articles provides a launching point for revisiting 100 years of research on the male germ cells and fertility in humans and animals and offers a perspective on the current state and future directions of the androl

Untargeted metabolomics reveals gender- and age- independent metabolic changes of type 1 diabetes in Chinese children.

INTRODUCTION: Type 1 diabetes (T1D) is a chronic condition associated with multiple complications that substantially affect both the quality of life and the life-span of children. Untargeted Metabolomics has provided new insights into disease pathogenesis and risk assessment. METHODS: In this study, we characterized the serum metabolic profiles of 76 children with T1D and 65 gender- and age- matched healthy controls using gas chromatography coupled with timeof-flight mass spectrometry. In parallel, we comprehensively evaluated the clinical phenome of T1D patients, including routine blood and

Metabolic coessentiality mapping identifies C12orf49 as a regulator of SREBP processing and cholesterol metabolism.

Coessentiality mapping has been useful to systematically cluster genes into biological pathways and identify gene functions1-3. Here, using the debiased sparse partial correlation (DSPC) method3, we construct a functional coessentiality map for cellular metabolic processes across human cancer cell lines. This analysis reveals 35 modules associated with known metabolic pathways and further assigns metabolic functions to unknown genes. In particular, we identify C12orf49 as an essential regulator of cholesterol and fatty acid metabolism in mammalian cells. Mechanistically, C12orf49 localizes to

Dietary niche and the evolution of cranial morphology in birds.

Cranial morphology in birds is thought to be shaped by adaptive evolution for foraging performance. This understanding of ecomorphological evolution is supported by observations of avian island radiations, such as Darwin's finches, which display rapid evolution of skull shape in response to food resource availability and a strong fit between cranial phenotype and trophic ecology. However, a recent analysis of larger clades has suggested that diet is not necessarily a primary driver of cranial shape and that phylogeny and allometry are more significant factors in skull evolution. We use phenome

Slowing ribosome velocity restores folding and function of mutant CFTR.

Cystic fibrosis (CF) is caused by mutations in the CF transmembrane conductance regulator (CFTR), with approximately 90% of patients harboring at least one copy of the disease-associated variant F508del. We utilized a yeast phenomic system to identify genetic modifiers of F508del-CFTR biogenesis, from which ribosomal protein L12 (RPL12/uL11) emerged as a molecular target. In the present study, we investigated mechanism(s) by which suppression of RPL12 rescues F508del protein synthesis and activity. Using ribosome profiling, we found that rates of translation initiation and elongation were mark

Identifying genetically driven clinical phenotypes using linear mixed models.

We hypothesized that generalized linear mixed models (GLMMs), which estimate the additive genetic variance underlying phenotype variability, would facilitate rapid characterization of clinical phenotypes from an electronic health record. We evaluated 1,288 phenotypes in 29,349 subjects of European ancestry with single-nucleotide polymorphism (SNP) genotyping on the Illumina Exome Beadchip. We show that genetic liability estimates are primarily driven by SNPs identified by prior genome-wide association studies and SNPs within the human leukocyte antigen (HLA) region. We identify 44 (false disco

Single-cell eQTL-based Mendelian randomization identifies immune cell subtype-specific regulators of epigenetic aging and prioritizes candidate therapeutic targets.

Epigenetic aging clocks offer precise measures of biological age, yet the causal contributions of immune gene expression within specific cell subtypes to epigenetic aging remain poorly understood. By integrating single-cell eQTL data from the OneK1K cohort with GWAS summary statistics for four epigenetic clocks (HannumAge Acceleration, IEAA, PhenoAge Acceleration, and GrimAge Acceleration), we performed two-sample Mendelian randomization across diverse immune cell subtypes, followed by colocalization analysis and gene-level phenome-wide association studies. We identified 11 eGene-cell type pai

Endothelial GPR68 Is Essential for Arteriogenesis and Represents a Therapeutic Target in a Model of Peripheral Artery Disease.

Peripheral artery disease (PAD) is characterized by arterial narrowing that reduces limb perfusion, causing significant morbidity. The body can compensate via shear stress-driven collateral artery growth (arteriogenesis). However, the primary receptors of mechanical forces on the endothelial cells in PAD remain poorly defined. Human phenome‑wide association study (PheWAS) shows that variants near GPR68, a gene encoding shear-stress sensitive G protein-coupled receptor (GPCR), are associated with increased PAD risk. To investigate the role of GPR68 in arteriogenesis in PAD, hindlimb ischemia (H

Opportunities and challenges in leveraging multiomics and biobanks for vision science.

PURPOSE OF REVIEW: Emerging biobank resources allow large-scale integration of eye-specific phenotypes with clinical, genomic, and multiomic data. This convergence enables unprecedented opportunities to systematically dissect the genetic architecture, epidemiology, and mechanistic pathways of both rare monogenic and common polygenic diseases. The review aims to critically examine how contemporary data extraction, multiomics, and analytic methodologies are reshaping disease classification, genetic discovery, and translational research in ophthalmology, while highlighting the associated challeng

Genetic epidemiological pipeline identifies candidate markers of clozapine-induced metabolic dysfunction: implications for precision prescribing.

OBJECTIVES: Clozapine, the gold-standard antipsychotic for treatment-resistant schizophrenia causes severe metabolic complications, including metabolic syndrome and increased type 2 diabetes (T2D) risk. A better understanding of the genetic factors influencing clozapine pharmacokinetics and the associated metabolic risk could inform precision medicine approaches to clozapine prescribing. METHODS: Using a series of genetic-epidemiological approaches, we aimed to identify candidate biomarkers associated with clozapine-induced metabolic dysfunction. Mendelian randomisation (MR) was employed to i

Common genetic mechanisms between obesity and COVID-19 severity: unravelling pleiotropic loci and biological pathways.

COVID-19 and obesity are complex conditions marked by immune and metabolic dysfunction, with the former still ranking among the leading causes of death from infectious diseases worldwide and the latter reaching pandemic proportions. Clinical evidence consistently shows that obesity increases the risk of severe COVID-19, yet the biological mechanisms underlying this association remain unclear. Given their physiological and clinical overlap, they may share genetic pathways. We investigated genetic variants jointly associated with body mass index (BMI) and COVID-19 using publicly available genome

Causal effects of cholelithiasis on hepatopancreatobiliary diseases: a multi-cohort Mendelian randomization study.

BACKGROUND: Cholelithiasis is commonly associated with multiple hepatopancreatobiliary diseases, yet whether these relationships reflect causal mechanisms or shared risk factors remains unclear. METHODS: We performed a phenome-oriented two-sample Mendelian randomization (MR) analysis to evaluate the causal impact of genetic liability to cholelithiasis across hepatopancreatobiliary outcomes. Independent genome-wide significant variants were selected as instrumental variables. Primary analyses used inverse variance weighting, complemented by sensitivity analyses, reverse MR, and multivariable M

Cryptococcal Infection Across Disease Stages: Host-Pathogen Interactions, Tissue Niches, and Translational Priorities.

Cryptococcosis often begins in the lungs and may later reach the central nervous system, where it can lead to severe or fatal disease. The course of infection is shaped by changing interactions between host cells, fungal adaptive states, and local tissue environments. Many virulence factors and immune responses have been studied in detail, but the evidence remains divided across organs, cell types, and experimental systems. As a result, pulmonary infection is better understood than later stages, including dissemination, crossing of the blood-brain barrier, CNS involvement, and long-term persis

Genetic Exploration of Targeting the Transient Receptor Potential Cation Channel Subfamily Member 6.

The transient receptor potential cation channel subfamily member 6 (TRPC6) represents an emerging druggable target with a broad therapeutic spectrum. TRPC6 Inhibitors are currently investigated for focal segmental glomerulosclerosis, acute respiratory distress syndrome due to COVID-19, and pulmonary hypertension. In the cardiovascular system, there is evidence that TRPC6 is critically involved in the development of cardiac hypertrophy, arrhythmia susceptibility and risk of restenosis after coronary stent implantation. However, data on systemic effects of TRPC6 modulation remain scarce. To asse

Precision Target Discovery for Migraine: An Integrated GWAS-eQTL-PheWAS Pipeline.

Migraine is a complex neurological disorder that severely compromises quality of life. Current therapies remain inadequate, creating an urgent need for precision medicine approaches. To bridge this gap, we integrated genome-wide association studies (GWASs) and multi-tissue expression quantitative trait loci (eQTL) data. Using Mendelian randomization (SMR/HEIDI) to identify putatively causal genes, followed by colocalization analysis, protein-protein interaction networks, and gene enrichment, we prioritized druggable targets. Phenome-wide association studies (PheWASs) further assessed their pot
