Gestational Protein Restriction Promotes Osteoporotic Phenotype During Aging.

Osteoporosis results from impaired bone remodeling and deterioration of bone microarchitecture, ultimately increasing fracture risk. Although early-life nutritional stress has been implicated in long-term health outcomes, its impact on skeletal remodeling mechanisms remains unclear. We investigated whether gestational protein restriction induces persistent alterations in the skeletal immune microenvironment that contribute to age-related bone fragility. Pregnant C57BL/6J mice were fed either a normal-protein or low-protein (LP) diet during gestation. Male offspring were evaluated at 12 and 18

Inflammation in Cardiomyopathies: Cellular Mechanisms Across Cardiac Phenotype.

Cardiomyopathies are traditionally classified by structural and genetic phenotypes, but emerging evidence highlights chronic myocardial inflammation as a pivotal driver of disease progression across different etiologies. This review synthesizes the current literature on the cellular and molecular inflammatory mechanisms underlying hypertrophic cardiomyopathy, Anderson-Fabry disease, cardiac amyloidosis, arrhythmogenic cardiomyopathy, and dilated cardiomyopathy. Across these distinct conditions, endogenous triggers such as metabolic substrates, misfolded amyloid fibrils, mechanical stress, or v…

Genomic insights into stroke recovery: cross-phenotype associations.

Stroke is a major cause of long-term disability with variable recovery. While clinical factors such as initial severity play a role, genetic … ity (modified Rankin Scale 0-2 versus 3-6), some of which were tested longitudinally, yielding nine phenotypes. Models were adjusted for age, sex, initial severity (NIH Stroke Scale score), and ancestry. Cross-phenotype associations were identified by evaluating single nucleotide polymorphisms (SNPs) associated (P < 5e-5) with multiple phenotypes. To determ …

The Audiologic and Otolaryngologic Phenotype in WHIM Syndrome.

OBJECTIVE: WHIM syndrome (warts, hypogammaglobulinemia, infections, and myelokathexis syndrome) is an ultra-rare primary immunodeficiency disease caused by autosomal dominant hyperfunctional mutations in the chemokine receptor CXCR4. Otolaryngologists are frequently consulted to evaluate WHIM patients because of recurrent acute ear and sinus infections, particularly in early childhood; however, a systematic assessment of otolaryngologic and audiometric phenotypes is lacking. This study comprehensively describes the auditory and otolaryngology phenotype of WHIM syndrome. STUDY DESIGN: Prospect…

Going Virtual as a Memory-Phenotype T Lymphocyte.

Conventional or 'true' memory CD8+ T cells (TTM) arise from immunologically naive T cells that circulate in the periphery after selection in the thymus. During infection or immunization by a foreign antigen, naive T cells can receive antigen-specific activation signals after recognition of MHC-antigenic peptide complexes. But the CD8+ T-cell population in immunologically naive hosts is not restricted to circulating naive T cells expecting cognate antigen encounter. Indeed, memory-phenotype T cells (TMP) develop in the absence of foreign antigen encounter and therefore exist in naive, pathogen-…

MIPD: Molecules, Imagings, and Clinical Phenotype Integrated Database.

Due to tumor heterogeneity, a subset of patients fails to benefit from current treatment strategies. However, an integrated analysis of imaging features, genetic molecules, and clinical phenotypes can characterize tumor heterogeneity, enabling the development of more personalized treatment approaches. Despite its potential, cross-modal databases remain underexplored. To address this gap, we established a comprehensive database encompassing 9965 genes, 5449 proteins, 1121 metabolites, 283 pathways, 854 imaging features, and 73 clinical factors from colorectal cancer patients. This database iden…

LRG1 Alters Pericyte Phenotype and Compromises Vascular Maturation.

Upregulation of leucine-rich alpha-2-glycoprotein-1 (LRG1) contributes to aberrant neovascularization in many different diseases. In contras … h the maturation of developing retinal vessels and drive vessel development towards a dysfunctional phenotype. These observations deliver further evidence that LRG1 is an angiopathic factor and highlight the therapeutic potential of blocking LRG1 in diseases characterized by pathogenic angiogenesis or vascular remodelling.

Cortical inexcitability in ALS: correlating a clinical phenotype.

BACKGROUND: Cortical inexcitability, a less studied feature of upper motor neuron (UMN) dysfunction in amyotrophic lateral sclerosis (ALS), was identified in a large cross-sectional cohort of ALS patients and their demographic and clinical characteristics were contrasted with normal or hyperexcitable ALS cohorts to assess the impact of cortical inexcitability on ALS phenotype and survival. METHODS: Threshold-tracking transcranial magnetic stimulation (TMS) technique with measurement of mean short interval intracortical inhibition (SICI) differentiated ALS patients into three groups (1) inexci…

Management of patients with myocarditis and arrhythmogenic phenotype.

Acute myocarditis (AM) is an inflammatory condition of the myocardium that may lead to severe complications, including acute heart failure and life-threatening ventricular arrhythmias (VAs). In-hospital VAs are estimated to affect 2.5% of adult patients with AM. Recent insights suggest a genetic predisposition to develop VA in a subset of patients with AM. This review will focus on arrhythmogenic manifestations of AM, highlighting risk stratification for VA after an acute episode and the contribution of genetic factors, emphasizing the need to integrate clinical, imaging, and genetic findings.

Congenital and infantile nephrotic syndrome: genotype-phenotype associations.

BACKGROUND: Congenital nephrotic syndrome (CNS) and infantile nephrotic syndrome (INS) are disorders of podocytes in the slit diaphragm. CNS manifests during the first three months of life, and INS between 3-12 months, with severe proteinuria due to mutations in the NPHS1 and NPHS2 genes. This study aimed to establish specific genotype-phenotype characteristics of CNS and INS in the North American population. METHODS: Eleven Pediatric Nephrology Research Consortium (PNRC) sites retrospectively reviewed charts of 36 patients born between 1998-2019 who had CNS or INS and underwent genetic testi…
