Polystyrene Nanoparticles Disrupt Oxidative Phosphorylation and Impair Placental Development in Mice.

BACKGROUND: Microplastics and nanoplastics, as pervasive and persistent environmental pollutants, are raising growing concerns regarding the … e placenta. Differentially expressed genes were enriched primarily in pathways related to oxidative phosphorylation, the tricarboxylic acid (TCA) cycle, and ErbB signalling. CONCLUSIONS: These changes collectively led to decreased mitochondrial ATP production, increased oxidative stress in the placenta, and potentially altered placental barrier function and trop …

KIF2A downregulation links amyloid-β to Tau phosphorylation in Alzheimer's disease.

Microtubules are essential components of the cytoskeleton. Dysfunctions of microtubules and microtubule-associated proteins are prominent features of neurodegenerative disorders. In Alzheimer's disease, changes in microtubule composition and hyperphosphorylation of Tau are more closely related to neurodegeneration than amyloid plaque formation. However, the accumulation of amyloid beta (Aβ) species is the earliest event in Alzheimer's disease pathology and induces Tau toxicity. KIF2A is a microtubule depolarizing kinesin with important roles during cortical development. KIF2A expression is mai…

Tau phosphorylation homeostasis: Mechanisms, targets, and therapeutic implications in Alzheimer's disease.

Neurofibrillary tangles, composed of excessively phosphorylated tau, are a core neuropathological hallmark of Alzheimer's disease (AD). However, therapeutic strategies aimed at directly clearing neurofibrillary tangles have demonstrated limited clinical efficacy, shifting the research focus towards the fundamental underlying mechanism- the dysregulation of tau phosphorylation. Evidence indicates that tau physiological phosphorylation is indispensable for microtubule stability and normal neuronal function, while its aberrant hyperphosphorylation drives neurodegeneration. Consequently, restoring…

Therapeutic potential of liposome@azoramide in cancer by suppressing oxidative phosphorylation.

OBJECTIVE: Targeting cancer cell metabolism represents a promising therapeutic strategy; however, metabolic reprogramming in tumor cells com … isms. RESULTS: We showed that azoramide effectively inhibits tumor growth by suppressing oxidative phosphorylation (OXPHOS) both in vitro and in vivo. Azoramide downregulated OXPHOS proteins, reduced mitochondrial respiration, and induced reactive oxygen species (ROS), leading to decreased mitochondrial ATP generation. Additionally, azoramide downregulated PGC-1 …

Piezo1 accelerates osteoarthritis progression via promotion of HBB-dependent oxidative phosphorylation.

The mechanosensitive ion channel Piezo1 serves as a key regulator of osteoarthritis (OA) progression, but the underlying mechanism remains l … unit beta (HBB) in chondrocytes. The upregulation of HBB initially enhanced mitochondrial oxidative phosphorylation (OXPHOS), but subsequently impaired mitochondrial function owing to excessive reactive oxygen species (ROS) production, thereby driving chondrocytes an energy metabolism shift from OXPHOS to glycolysis to meet the energy requirements. Notably, intra …

Expanding the Phenotype of TUFM -Related Combined Oxidative Phosphorylation Deficiency 4.

Combined oxidative phosphorylation deficiency 4 (COXPD4) is a rare mitochondrial condition caused by biallelic deleterious variants in the nuclear-encoded gene TUFM. To date, most individuals with COXPD4 have presented with encephalopathy, hypotonia, and abnormal brain imaging. Many of the reported individuals died in infancy. We aim to expand the clinical and biochemical phenotype of COXPD4 by reporting on an adult with this condition. Our proband has a homozygous TUFM c.1025T>G, p.(Val342Gly) variant. He has sensorineural hearing loss, hyperlactatemia with mild illness, and reduced activity …

Metixene hydrochloride hydrate mitigates kidney tubulointerstitial fibrosis by inhibiting Smad3 phosphorylation.

Chronic Kidney disease (CKD), in which renal fibrosis is the defining pathological feature, poses significant global health and economic cha … ters and expression of renal fibrotic markers. Mechanistically, MHH suppressed TGF-β1-induced Smad3 phosphorylation both in vitro and in vivo. Our findings indicate that MHH exerts potent antifibrotic effects by downregulating the TGF-β1/Smad3 signaling pathway and suppressing the expression of fibrotic factors in renal cells and obstructed kidneys. Therefore, MH …

Roles of the phosphorylation sites of distinct α1A-adrenergic receptor domains.

Phosphorylation participates in the signaling, localization, and trafficking of G protein-coupled receptors. In this study, the amino acids known to be phosphorylated in α1A-adrenergic receptors were substituted with nonphosphorylatable residues in intracellular loop 3 (IL3) mutant, the carboxyl terminus (CTerm) mutant, and both structural regions (IL3-CTerm mutant). Their functions were compared with those of the wild-type (WT) receptor. Agonist- and protein kinase C-induced receptor phosphorylation was markedly decreased in the IL3-CTerm mutant, confirming the major roles of both domains in …

Brassinosteroids promote sugar synthesis by inhibiting BIN2 phosphorylation of phosphoenolpyruvate carboxykinase.

Sugar is both an essential energy source and the major substrate for cell wall biosynthesis during plant growth, yet how growth-promoting hormones regulate sugar synthesis remains unclear. Here we show that the brassinosteroids (BRs) promote gluconeogenic and photosynthetic sugar synthesis by activating phosphoenolpyruvate carboxykinase (PCK), which catalyses the conversion of oxaloacetate to phosphoenolpyruvate, a central step in primary metabolism. Arabidopsis BR-deficient mutants display reduced PCK1 activity and elevated phosphorylation at conserved Ser-62 and Thr-66 residues. BR treatment…

RASD2 Drives Renal Clear Cell Carcinoma Progression via RAF1 (Ser338) Phosphorylation.

Clear cell renal cell carcinoma (ccRCC) is a highly aggressive malignancy with limited treatment options. Although RASD2, which encodes a Ra … -immunoprecipitation and LC-MS/MS, we identified RAF1 as a key binding partner. RASD2 enhanced RAF1 phosphorylation at Ser338, thereby activating the P38/ERK-MAPK pathway. RAF1 phosphorylation at Ser338 was critical for ccRCC cell growth, migration, and invasion. The RAF1 inhibitor BAY43-9006 reduced p-RAF1 (Ser338)/RAF1 levels, suppressed ccRCC proliferation, mi …

Phosphorylation and DNA damage resolution coordinate SOX2-mediated reprogramming in vivo.

The stem cell factor SOX2 can reprogram resident glial cells into neurons in the adult mammalian central nervous system, but the molecular mechanisms underlying this process remain poorly understood. Here, we show that both SOX2 phosphorylation and the PRKDC-dependent nonhomologous end joining (NHEJ) pathway are essential for SOX2-mediated in vivo glia-to-neuron reprogramming. A phospho-mimetic SOX2 mutant significantly enhances reprogramming output without altering neuronal fate. Conversely, loss of PRKDC or knockdown of core NHEJ components KU80 and LIG4 abolishes reprogramming. Notably, p53…

Vitamin B12 Supports Skeletal Muscle Oxidative Phosphorylation Capacity in Male Mice.

BACKGROUND: Vitamin B-12 is a cofactor in folate-mediated 1-carbon metabolism, which generates nucleotides {thymidylate [deoxythymidine monophosphate (dTMP)] and purines} and methionine. Depressed de novo thymidylate (dTMP) synthesis leads to uracil accumulation in DNA. OBJECTIVES: This study aimed to determine how B-12 availability affects mitochondrial DNA (mtDNA) integrity and mitochondrial function in skeletal muscle. B-12 deficiency was modeled in young-adult mice. Intramuscular B-12 injection in aged mice assessed the role of B-12 supplementation in age-related changes in skeletal muscl

Mycobacterium tuberculosis modulates phosphorylation of host ATP6V1E1 to promote intracellular survival.

Intracellular pathogens such as Mycobacterium tuberculosis (Mtb) can promote their survival within infected cells by preventing lysosomal acidification. Here, we report that Mtb secretes a protein (Rv1184, or acyltransferase Chp2) that inhibits lysosomal acidification by targeting the host vacuolar ATPase (V-ATPase). We show that phosphorylation of the V-ATPase E1 subunit (ATP6V1E1) at Tyr56/57 suppresses lysosomal acidification through inhibition of V-ATPase assembly. Further investigation reveals that tyrosine kinase BMX promotes phosphorylation of ATP6V1E1. Strikingly, Chp2 increases BMX-de…

Salmonella effector kinase SteC is activated by phosphorylation at Serine 379.

The pathogen Salmonella, which causes significant human morbidity and mortality, encodes an effector kinase, SteC, which mediates actin polymerisation and cell migration. Given the minimal nature of its kinase domain, it remains unclear how SteC is catalytically active and how this activity is regulated. Here, we show that SteC is activated following the phosphorylation of the highly conserved S379 residue, which can be carried out by a host kinase. Phosphorylation of S379 dramatically increases nucleotide binding affinity of SteC, enabling substrate phosphorylation and promoting actin polymer…

Idebenone Protects Photoreceptors Impaired by Oxidative Phosphorylation Disorder in Retinal Detachment.

PURPOSE: Oxidative phosphorylation (OXPHOS) is an aerobic metabolic mechanism, and its dysfunction plays an important role in the pathological changes of ischemic diseases. However, systematic studies on the occurrence of retinal detachment (RD) are lacking. METHODS: Single-cell RNA sequencing (scRNA-seq) of the human retina was performed to detect the metabolic changes of various retinal cells after RD. In this study, animal experiments were conducted to explore the OXPHOS activity after RD. In addition, idebenone, a coenzyme Q10 (CoQ10) analog currently used to treat Leber hereditary optic …
