The pars tuberalis: The site of the circannual clock in mammals?

Accurate timing and physiological adaptation to anticipate seasonal changes are an essential requirement for an organism's survival. In contrast to all other environmental cues, photoperiod offers a highly predictive signal that can be reliably used to activate a seasonal adaptive programme at the correct time of year. Coupled to photoperiod sensing, it is apparent that many organisms have evolved innate long-term timekeeping systems, allowing reliable anticipation of forthcoming environmental changes. The fundamental biological processes giving rise to innate long-term timing, with which the …

Androgenic activity in 15-day-old male rats: role of the maternal pineal gland.

Female Sprague-Dawley rats, exposed to a long (18L:6D) or a short (6L:18D) photoperiod from 21 days of age, were mated when they reached 55 days of age. On Day 2 of gestation, dams were pinealectomized or sham-operated. Pre- and postnatal photoperiods were identical, and offspring were killed at 15 days of age. Maternal pinealectomy had no effect when rats were kept on 18L:6D. Rats born to sham-operated mothers and kept on 6L:18D had higher testicular testosterone and androstenedione content than offspring raised on the long photoperiod. This stimulatory effect of the short photoperiod was blo…

Some reflections on the phylogeny and function of the pineal.

The pineal gland is a universal feature of vertebrate organization and has been implicated in the control of rhythmic adaptations to daily and seasonal cycles. This paper considers three aspects of pineal function; the generation of a rhythmical endocrine signal (the nocturnal synthesis of melatonin) and the use of the signal in the regulation of circadian and photoperiodic functions. The shape of the nocturnal signal is determined by an interaction of afferent neural control and biochemical processes intrinsic to the pinealocyte. The nature of the effect of the signal upon circadian systems i…

Photic and pineal modulation of food anticipatory circadian activity rhythms in rodents.

Restricted daily feeding schedules entrain circadian oscillators that generate food anticipatory activity (FAA) rhythms in nocturnal rodents … ies of FAA mechanisms, we quantified FAA in rats and mice alternately maintained in a standard full photoperiod (12h of light/day) and in a skeleton photoperiod (two 60 min light pulses simulating dawn and dusk). In both species, FAA was significantly and reversibly enhanced in the skeleton photoperiod compared to the full photoperiod. In a third experiment, …

The saccus vasculosus of fish is a sensor of seasonal changes in day length.

The pars tuberalis of the pituitary gland is the regulatory hub for seasonal reproduction in birds and mammals. Although fish also exhibit r … se genes are expressed in the coronet cells of the saccus vasculosus, suggesting the existence of a photoperiodic signalling pathway from light input to neuroendocrine output. In addition, isolated saccus vasculosus has the capacity to respond to photoperiodic signals, and its removal abolishes photoperiodic response of the gonad. Although the physiological r …

Effects of lighting patterns in pubertal development and metabolism of female wistar rats.

Modern lifestyle is characterized by constant exposure to artificial light, which is associated with alterations in biological rhythms, abnormalities to reproductive cycles and metabolic changes. In this study, we aimed to evaluate the effects of four different lighting patterns on puberty timing and on possible metabolic changes in female Wistar rats. Additionally, we developed a machine learning algorithm to automatically classify the stages of the estrous cycle. Adult Wistar rats mated during a week at a photoperiod station where they were exposed to combined red-green-blue lights (RGB) dur…

Photoresponsive Fischer 344 Rats are reproductively inhibited by melatonin and differ in 2-[125I] lodomelatonin binding from nonphotoresponsive Sprague-Dawley rats.

Many temperate-zone species use photoperiod as an environmental cue to regulate reproductive timing. Strains of laboratory rats differ in their responsiveness to photoperiod, with the Fischer 344 (F344) strain being the most responsive known. F344 rats and closely related strains that differ in photoresponsiveness may be useful models to study the mechanisms and genetic basis for photoresponsiveness. We tested two hypotheses: (i) that melatonin mediates photoresponsiveness in F344 rats, as is the case in all other mammals tested, and (ii) that the location, abundance, or affinity of melatonin …

A test of the coincidence and duration models of melatonin action in Siberian hamsters: the effects of 1-hr melatonin infusions on testicular development in intact and pinealectomized prepubertal Phodopus sungorus.

The pineal hormone melatonin is known to play an important role in mediating photoperiodic messages to the reproductive system in seasonal breeding animals. Our goal was to test, in a single experimental paradigm, two hypotheses that have been forwarded to describe how the circadian rhythm of pineal melatonin transmits photoperiodic information to the reproductive system: 1) induction, i.e., a short-day effect, occurs when secreted melatonin and a circadian rhythm of sensitivity to melatonin coincide in time; 2) induction occurs following exposure to elevated circulating melatonin levels for a…

Molecular basis of photoinduced seasonal energy rheostasis in Japanese quail (Coturnix japonica).

Seasonal rhythms in photoperiod are a predictive cue used by many temperate-zone animals to time cycles of lipid accumulation. The neuroendocrine regulation of seasonal energy homeostasis and rheostasis are widely studied. However, the molecular pathways underlying tissue-specific adaptations remain poorly described. We conducted two experiments to examine long-term rheostatic changes in energy stability using the well-characterized photoperiodic response of the Japanese quail. In experiment 1, we exposed quails to photoperiodic transitions simulating the annual photic cycle and examined the m…

Retrospective Study of 23 Patients with Traumatic Posterolateral Tibial Plateau Fracture Treated in a Single Center Between 2017 and 2019 with Lateral Arthrotomy, Reduction, and Plate Fixation Using the Frosch Approach.

The Institute of Physiology of the Czech Academy of Sciences (CAS) has been involved in the field of chronobiology, i.e., in research on temporal regulation of physiological processes, since 1970. The review describes the first 35 years of the research mostly on the effect of light and daylength, i.e., photoperiod, on entrainment or resetting of the pineal rhythm in melatonin production and of intrinsic rhythms in the central biological clock. This clock controls pineal and other circadian rhythms and is located in the suprachiasmatic nuclei (SCN) of the hypothalamus. During the early chronobi…

The circadian system of ruin lizards: a seasonally changing neuroendocrine loop?

Previous studies have shown that the amplitude of daily melatonin production in cultured ruin lizard pineal organs explanted in the summer is significantly higher than that from organs explanted in the winter. To test whether seasonal photoperiodic changes are decoded autonomously by the pineal gland, pineals explanted in summer were cultured in vitro and exposed to changes between winter and summer photoperiods. The changes in photoperiod duration did not affect the daily profiles of in vitro melatonin production. The discrepancy between the present in vitro results and those from lizards exp…

Why does the effectiveness of light trapping for tea leafhoppers decline sharply in late autumn?

Light trapping is a common monitoring and control method for Empoasca onukii Matsuda (Hemiptera: Cicadellidae), a major pest in tea gardens. However, in late autumn (late October), despite large populations of leafhoppers in tea gardens around 30°N latitude, the effectiveness of light trapping declines sharply. The factors causing this decline are unclear. In this study, field experiments revealed significant differences in ambient temperature and photoperiod corresponding to variation in light trapping effectiveness across different leafhopper peaks. Through a series of bioassays, we found th…

Effects of exogenous leptin on seasonal reproductive responses to interacting environmental cues in female Siberian hamsters.

Animals living in temperate climates respond to environmental cues that signal current and future resource availability to ensure that energy resources are available to support reproduction. Siberian hamsters (Phodopus sungorus) undergo robust gonadal regression in short, winter-like photoperiods as well as in response to mild food restriction in intermediate photoperiods. The goal of the present study was to investigate whether leptin is a relevant metabolic signal in regulating gonadal regression in response to diminishing food availability. Adult female hamsters housed in short-day (winter-…

Discrete thalamic lesions attenuate winter adaptations and increase body weight.

The midline thalamus (e.g., the paraventricular thalamic nuclei and the reuniens nucleus) of Siberian hamsters and other mammals has been reported to contain specific binding sites for melatonin, a hormone that is essential for photoperiodically induced winter adaptations such as reproductive quiescence, loss of body weight, daily torpor, and the winter molt. The first experiment investigated whether the midline thalamus is necessary for these winter adaptations. Adult Siberian hamsters received discrete neurotoxic lesions of the paraventricular thalamic nuclei or the reuniens nucleus while un…

We found that a long-day photoperiod (16 h light/8 h dark cycle; 100 lx) significantly improved the survival of R6/2 female mice by 2.4 weeks, compared to mice kept under standard conditions (12 h light/12 h dark cycle). Furthermore, a long-day photoperiod improved the nocturnality of …

Melatonin, serotonin, 5-hydroxyindole-3-acetic acid and N-acetyltransferase in the pineal of the European hamster (Cricetus cricetus) kept under natural environmental conditions: lack of a day/night rhythm in melatonin formation in spring and early summer.

In female European hamsters killed in spring and early summer, pineal melatonin content exhibited no day/night rhythm. Absolute levels measu … ean that in the European hamster it is not melatonin but another substance that is of importance in photoperiodism. An absence of melatonin rhythm, however, could also be simply a peculiar pattern of melatonin production observed at a given period of the year. In this case, melatonin would be able to transduce photoperiodic information in the European hamster …

Effects of pinealectomy and melatonin or 5-methoxytryptamine on testicular LH and PRL receptors in Syrian hamsters (Mesocricetus auratus).

The pineal has been previously shown to be an important factor in the regulation of testicular function in photoperiodic mammals. The effects of lack or increase in pineal hormones on testicular hormonal receptors has, therefore, been examined. Pinealectomy decreased the concentration of testicular LH receptors in hamsters exposed to either a long or short photoperiod but had no effect on the concentration of testicular PRL receptors. In animals exposed to a short photoperiod, pinealectomy prevented testicular regression and the concomitant decreases in total LH and PRL receptor contents. Trea…

Time of day of birth and absence of endocrine and uterine contractile activity rhythms in sheep.

To determine whether 24-h rhythms characterize hormone secretion and uterine activity in the pregnant sheep, blood samples were drawn every 1-4 h for 48 h from ewes and fetuses from day 120 of gestation to term. Repetitive 24-h rhythms were absent for cortisol, progesterone, and prolactin in maternal and fetal circulation and for hourly mean uterine contraction rate and amplitude. To test whether photoperiod or pineal melatonin contributes to the absence of rhythms, pineal-intact and pinealectomized ewes and their fetuses were studied in reverse photoperiod. Again, there was little evidence to…

The underlying mechanisms of vertebrate seasonal reproduction.

Animals make use of changes in photoperiod to adapt their physiology to the forthcoming breeding season. Comparative studies have contributed to our understanding of the mechanisms of seasonal reproduction in vertebrates. Birds are excellent models for studying these phenomena because of their rapid and dramatic responses to changes in photoperiod. Deep brain photoreceptors in birds perceive and transmit light information to the pars tuberalis (PT) in the pituitary gland, where the thyroid-stimulating hormone (TSH) is produced. This PT-TSH locally increases the level of the bioactive thyroid h…
