Plant regeneration from cultured immature embryos of Sorghum bicolor (L.) Moench.

Immature embryos of 20 sorghum genotypes were cultured on MS 5 medium containing MS mineral salts supplemented with 2,4-D, zeatin, glycine, niacinamide, Ca-pantothenate, L-asparagine, and vitamins. For regeneration, calli were transferred onto the same medium with the exception that IAA was substituted for 2,4-D. In general, immature embryos obtained 9-12 days after pollination resulted in the best redifferentiation. Ability of calli to regenerate varied among genotypes; cultivars C401-1 and C625 had the highest redifferentiation frequencies. Ability to redifferentiate was heritable and acted …

Heat stress in cereal crops: reproductive development and grain filling.

Increasingly frequent extreme heat events threaten cereal production and food security under a changing climate. The reproductive-to-grain f … evelopmental processes, including inflorescence development, gametophyte development, flowering and pollination, fertilization, and grain filling. We then integrate emerging evidence on cross-cutting mechanisms that connect stage-specific heat responses, focusing on hormonal and redox homeostasis, carbohydrate metabolism and source-sink coordination, proteost …

Polycomb protein ZmEMF1a restricts endosperm proliferation and directs differentiation via stage-specific H2Aub1 and H3K27me3 landscapes in maize.

Polycomb group (PcG) proteins serve as pivotal epigenetic repressors that govern the transcriptional programs underlying cell growth and dif … -specific epigenetic regulatory program: it predominantly mediates H3K27me3 deposition at 6 d after pollination (DAP), while coordinately regulating the deposition of both H3K27me3 and H2Aub1 at 10 DAP. Loss of ZmEMF1a leads to ectopic hyperproliferation of differentiated endosperm tissues, specifically the basal endosperm transfer layer (BETL) and aleurone ( …

Protecting honey bees through microbiome engineering.

Honey bees are indispensable insects. Their pollination services support modern agriculture and natural ecosystems. Managed honey bee colonies face increasing threats to their survival, ranging from environmental stressors that include agrochemicals to infestations of arthropod pests and infections with microbial pathogens. Like humans, honey bees have a native gut microbiome that supports their health. However, the bee gut microbiome has a simpler composition than the gut microbiome of mammals, and its main constituent bacterial species can be easily cultured outside of the host. This experim…

Bioinsecticide spinosad poses multiple harmful effects on foragers of Apis mellifera.

There are multifactorial causes for the recent decline in bee populations, which has resulted in compromised pollination and reduced biodiversity. Bees are considered one of the most important non-target insects affected by insecticides used in crop production. In the present study, we investigated the effects of acute oral exposure to spinosad on the survival, food consumption, flight behavior, respiration rate, activity of detoxification enzymes, total antioxidant capacity (TAC), brain morphology, and hemocyte count of Apis mellifera foragers. We tested six different concentrations of spinos…

Outdoor airborne allergens: Characterization, behavior and monitoring in Europe.

Aeroallergens or inhalant allergens, are proteins dispersed through the air and have the potential to induce allergic conditions such as rhinitis, conjunctivitis, and asthma. Outdoor aeroallergens are found predominantly in pollen grains and fungal spores, which are allergen carriers. Aeroallergens from pollen and fungi have seasonal emission patterns that correlate with plant pollination and fungal sporulation and are strongly associated with atmospheric weather conditions. They are released when allergen carriers come in contact with the respiratory system, e.g. the nasal mucosa. In addition…

OsCER1 regulates humidity-sensitive genic male sterility through very-long-chain (VLC) alkane metabolism of tryphine in rice.

Humidity-sensitive genic male sterility (HGMS) is a novel type of environment-sensitive male sterility (EGMS) which plants are male sterile … n revealed abnormal tryphine structure, potentially representing the basis of HGMS. Furthermore, co-pollination with mixed OsCER1Cas mutant and maize (Zea mays L.) pollen could rescue the fertility of the mutant, thereby establishing the key role of tryphine in germination on stigmas. OsCER1 knockout might affect VLC alkane metabolism and therefore alter the …

Climate change, environment and allergy.

Climate change with global warming is a physicometeorological fact that, among other aspects, will also affect human health. Apart from cardiovascular and infectious diseases, allergies seem to be at the forefront of the sequelae of climate change. By increasing temperature and concomitant increased CO(2) concentration, plant growth is affected in various ways leading to prolonged pollination periods in the northern hemisphere, as well as to the appearance of neophytes with allergenic properties, e.g. Ambrosia artemisiifolia (ragweed), in Central Europe. Because of the effects of environmental…

The Capsella rubella genome and the genomic consequences of rapid mating system evolution.

The shift from outcrossing to selfing is common in flowering plants, but the genomic consequences and the speed at which they emerge remain poorly understood. An excellent model for understanding the evolution of self fertilization is provided by Capsella rubella, which became self compatible <200,000 years ago. We report a C. rubella reference genome sequence and compare RNA expression and polymorphism patterns between C. rubella and its outcrossing progenitor Capsella grandiflora. We found a clear shift in the expression of genes associated with flowering phenotypes, similar to that seen in

Genetics, evolution, and adaptive significance of the selfing syndrome in the genus Capsella.

The change from outbreeding to selfing is one of the most frequent evolutionary transitions in flowering plants. It is often accompanied by … e that evolution of the selfing syndrome was at least partly driven by selection for efficient self-pollination. Thus, our studies pave the way for a molecular dissection of selfing-syndrome evolution.

Carbamate-Modified Pyrido[1,2-a]pyrimidinones: Design and Synthesis of Insecticides with High Efficacy and Low Bee Toxicity.

Megoura crassicauda, a sap-feeder on legumes, remains a persistent threat, yet current control depends largely on neonicotinoids that pose serious risks to beneficial pollinators. We herein describe a set of new pyrido[1,2-a]pyrimidinone derivatives bearing a carbamate moiety attached via a methylene spacer at the 2-position. Insecticidal bioassays identified H3 as the most potent analogue against M. crassicauda, comparable to the commercial standard TFM. Encouragingly, H3 exhibited remarkably low toxicity to honeybees, with acute oral and contact LD50 values of 24.0 and 35.1 μg a.i./bee, resp

Extraction and Quantification of Phenolic Compounds in Maize.

Plants accumulate hundreds of thousands of specialized metabolites that participate in their interactions with the environment. Among these compounds, phenolics represent a large class, and they play important physiological roles, such as providing a first barrier against pathogens, cues to pollinators, and radiation protection. Maize is one of the most important crops worldwide for food, animal feed, and biofuels, and it has the potential to accumulate different phenolics in vegetative tissues as well as in seeds. Recent studies have identified a large number of phenolic compounds-with a dive

Biological damage of monocrotaline on the brain and intestinal tissues of Apis mellifera.

Monocrotaline (MCT) is a toxic alkaloid present in plants, posing a threat to animals in terrestrial ecosystems. However, little is known about its potential impacts on pollinating insects. Here, we report the effects of of MCT on the brains and intestines of foraging honeybees (Apis mellifera). MCT exposure resulted in a reduction in head weight and swelling of the abdomen in honeybees. Additionally, MCT exposure caused morphological damage to the brain, characterized by decreased antioxidant capacity and increased apoptosis, along with intestinal tissue damage that was accompanied by increas

Identification of volatile organic compounds in purple and white soybean flowers by HS-SPME/GC-MS.

This study aimed to establish a method for the extraction, enrichment, and identification of volatile organic compounds (VOCs) released by the flowers of purple (BRS 399) and white (DONMARIO 6563) soybean varieties. We tested the Static Headspace (HS) and Solid Phase Microextraction (SPME) methods using various fibre types: PDMS (Polydimethylsiloxane), PDMS/DVB (Divinylbenzene), and PDMS/DVB/CAR (Carboxen). We employed gas chromatography-mass spectrometry (GC-MS) to identify the VOCs. The SPME method with PDMS/DVB and PDMS/DVB/CAR fibres yielded the highest number of extracted compounds for bo

Consensus recommendations for use of maintenance immunosuppression in solid organ transplantation: Endorsed by the American College of Clinical Pharmacy, American Society of Transplantation, and the International Society for Heart and Lung Transplantation.

Advances in maintenance immunosuppression over the past three decades have improved solid organ transplantation outcomes dramatically. Uninterrupted access to immunosuppression is paramount to minimize rejection and maintain allograft and patient survival. There is no standardized approach to maintenance immunosuppression management. Agents used vary based on transplanted organ, center-specific protocol, provider expertise, insurance formularies, ability to cover co-pays, recipient characteristics and tolerability. Published data reflects this heterogeneity. Despite this limitation, maintenanc

Changes in hypopharyngeal glands of nurse bees (Apis mellifera) induced by pollen-containing sublethal doses of the herbicide Roundup®.

Decreasing pollinator populations worldwide has generated great concern and stimulated countless studies to understand the origin of colony losses. One main cause is the indiscriminate use of different pesticides, producing subtle negative effects on bee physiology and behavior. Royal jelly synthesized in the hypopharyngeal glands is an essential protein for feeding all individuals of the hive, especially the queen. Therefore, the present study aimed to determine the effect of sublethal concentrations of Roundup® on the hypopharyngeal glands of nursing workers, including its interference with

Attractant for vinegar fly, Drosophila busckii, and cluster fly, Pollenia rudis (Diptera: Drosophilidae et Calliphoridae).

A field test carried out in an industrial greenhouse in Lithuania revealed the attractiveness of synthetic methyl salicylate (MeSa) to two dipteran species: the vinegar fly, Drosophila busckii (Drosophilidae), and the cluster fly, Pollenia rudis (Calliphoridae). The attractant for the former fly species was especially effective, as sticky traps containing 0.25 ml of MeSa captured (814 +/- 55) D. busckii flies/trap on average compared to (12 +/- 4) flies/trap in control traps. The mean capture of P. rudis [(42 +/- 4) flies/trap] was significantly higher in MeSa-baited traps compared to the cont
